Characterization of Pseudomonas syringae Strains Associated with Shoot Blight of Raspberry and Blackberry in Serbia

AB  - During May 2016, severe blight symptoms were observed in several raspberry and blackberry fields in Serbia. In total, 22 strains were isolated: 16 from symptomatic raspberry shoots, 2 from asymptomatic raspberry leaves, and 4 from symptomatic blackberry shoots. Additionally, eight raspberry strains, isolated earlier from two similar outbreaks, were included in the study. Pathogenicity of the strains was confirmed on detached raspberry and blackberry shoots by reproducing the symptoms of natural infection. The strains were Gram-negative, fluorescent on King’s medium B, ice nucleation positive, and utilized glucose oxidatively. All strains were levan positive, oxidase negative, nonpectolytic, arginine dihydrolase negative, and induced hypersensitivity in tobacco leaves (LOPAT + −−− +, Pseudomonas group Ia). Furthermore, all strains liquefied gelatin and hydrolyzed aesculin but did not show tyrosinase activity or utilize tartrate (GATTa + + − −). Tentative identification using morphology, LOPAT, GATTa, and ice-nucleating ability tests suggested that isolated strains belong to Pseudomonas syringae. The syrB gene associated with syringomycin production was detected in all strains. DNA fingerprints with REP, ERIC, and BOX primers generated identical profiles for 29 strains, except for strain KBI 222, which showed a unique genomic fingerprint. In all, 9 of 10 selected strains exhibited identical sequences of four housekeeping genes: gyrB, rpoD, gapA, and gltA. Five nucleotide polymorphisms were found in strain KBI 222 at the rpoD gene locus only. In the phylogenetic tree based on a concatenated sequence of all four housekeeping genes, strains clustered within phylogroup 2 (i.e., genomospecies 1) of the P. syringae species complex, with pathotype strains of P. syringae pv. aceris and P. syringae pv. solidagae as their closest relatives. There was no correlation between genotype and geographic origin, particular outbreak, host, or cultivar.

PROUČAVANJE MIKROBIOTE U TUMORIMA POVEZANIM SA BAKTERIOZNIM RAKOM KUPINE

AB  - Bakteriozni rak predstavlja ekonomski veoma značajno i široko rasprostranjeno
oboljenje voćaka, vinove loze i pojedinih ukrasnih biljaka. Bolest prouzrokuju tumorogeni
pripadnici familije Rhizobiaceae, posebno rodova Agrobacterium, Allorhizobium i Rhizobium.
Tokom prethodnih proučavanja ove bolesti na biljkama bestrne kupine, kao prouzrokovač
bakterioznog raka identifikovana je nova vrsta iz roda Rhizobium, nazvana Rhizobium
tumorigenes. Međutim, poznato je da tumore ne naseljavaju samo patogene tumorogene
bakterije, već kompleksna zajednica mikroorganizama, sastavljena od različitih vrsta
bakterija. Cilj istraživanja bio je da se prouči raznovrsnost populacije bakterija koja naseljava
tumore na biljkama kupine.
Tokom 2021. godine, na području Lučana, prikupljeno je devet uzoraka kupine sa
simptomima bakterioznog raka. Identifikacija bakterija koje čine mikrobiom bakterioznog
tumora, vršena je primenom metode sekvenciranja nove generacije (NGS). Ukupna DNK
izolovana je iz tkiva tumora, nakon čega je urađeno umnožavanje a zatim sekvenciranje
produkta 16S rRNK gena. Dobijene varijante umnoženih sekvenci (ASV) taksonomski su
determinisane primenom „blastn“ programa.
Rezultati bioinformatičkih analiza pokazali su da je vrsta R. tumorigenes prisutna u
svim testiranim uzorcima sa relativnom zastupljenošću od 14 - 95%. Takođe, ova vrsta bila je
dominantna u pet testiranih uzoraka (42 - 95%). U preostala četiri uzorka, najveću
zastupljenost imale su vrste roda Rachnella (24 - 63%), dok je kod prethodnih pet uzoraka taj
udeo bio znatno niži (0,4 - 9%). U pojedinim uzorcima, detektovano je prisustvo Bacillus sp. (6
- 25%), Rhizobium spp. (8 - 20%), Pectobacteriaceae (~13%), Pantoea sp. (~11%),
Novosphingobium sp. (~7%) i/ili Burkholderiaceae (~5%). Preostale vrste bakterija koje su
dektovane u pojedinim uzorcima, imale su relativnu zastupljenost manju od 5%.
Pored vrste R. tumorigenes, koja je bila prisutna u svim uzorcima tumora kupine, a
koja je i u prethodnim istraživanjima identifikovana kao prouzrokovač bolesti, ostale
detektovane vrste takođe mogu ostvariti različite odnose sa biljkom domaćinom. Prema
literaturnim podacima, neke od njih mogu imati ulogu u biološkoj kontroli i/ili kao stimulatori
rasta biljaka.

Differentiation of Pseudomonas syringae pathovars originating from stone fruits

AB  - Due to an overlapping host range, similar symptomatology and many common characteristics, Pseudomonas syringae pathovars originating from stone fruits can easily be misidentified. In order to select tests for rapid and efficient differentiation of P. s. pvs. syringae, morsprunorum and persicae, we studied the suitability and differentiating potential of some standard bacteriological and molecular methods. Differentiation of the strains was performed using LOPAT, GATTa and ice nucleation tests, nutrient sucrose broth growth and utilization of various carbon sources. PCR method enabled the detection of toxin-producing genes: syrB and syrD in P. s. pv. syringae, and cfl gene in P. s. pv. morsprunorum race 1. Syringomycin production by pv. syringae was confirmed in bioassay using Geotrichum candidum, Saccharomyces cerevisiae and Rhodotorula pilimanae as indicator organisms. Pathogenicity test on lemon and immature nectarine fruits, as well as on string bean pods, showed different intensity of reaction of the inoculated material which could separate pv. syringae from the other two pathovars. PCR-based repetitive sequences, Rep-PCR with REP, ERIC and BOX primers revealed different genetic profiles within P. syringae pathovars.

AB  - Timley and reliable detection of the hazelnut pathogen Xanthomonas arboricola pv. corylina (Xac) is essential for the production of good quality disease-free planting material. In order to improve knowledge on diagnostic tools for this quarantine bacterium, the effectiveness of laboratory methods recomended by EPPO and additional modified procedures developed within COST873 was compared. Methods included the classical, biochemical, serological, pathogenicity and molecular approach. To confirm the presence of the pathogen in plant tissue, a combination of different techniques is necessary. For successful isolation of Xac cells from different hazelnut organs, general nutrient agar or semi-selective glucose amended media are recommended. Traditional biochemical techniques are still necessary for the identification of isolates at the species level. ELISA and IF are useful as screening assays for the early and rapid pathogen detection. In addition, PCR-based methods proved to be useful for the rapid detection of Xac cells, although highly pathovar-specific genetic tool is not available. Identification at the pathovar level requires pathogenicity tests on hazelnut plants that enable confirmation of strains' pathogenic nature and reproduction of symptoms. However, a choice of various pathogenicity testing procedures should be made based on testing material available and type and dynamics of symptom development.

Bakterioze i fitoplazmoze koštičavih voćaka

AB  - U voćarstvu naše zemlje koštičave voćke zauzimaju značajno mesto i njihovo gajenje predstavlja profitabilnu delatnost. Međutim, rentabilnost voćarske proizvodnje mogu ugroziti različite biljne bolesti. Po značaju, raširenosti, ali i nemogućnosti uspešne kontrole, izdvajaju se oboljenja bakteriozne i fitoplazmozne prirode. Bakteriozno izumiranje, koje prouzrokuje Pseudomo nas syringae sa svoja dva patogena varijeteta syringae i morsprunorum, zadaje ozbiljne probleme voćarima u našoj zemlji. Značajan prob lem, takođe, predstavlja i Agrobacterium tumefaciens, prouzrokovač bakterioznog raka korena, koji nanosi štete u rasadničarskoj proizvodnji. U radu su pred stavljena i oboljenja koja nisu prisutna u našoj zemlji, a u svetu su opisana kao izuzetno destruktivna. To su bakteriozno izumiranje breskve, koje prouzrokuje P. s. pv. persicae i bakteriozna pegavost lišća koštičavih voćaka, čiji je prouzrokovač Xanthomonas arboricola pv. pruni. U Srbiji je prisutno i fitoplazmozno oboljenje Evropsko žutilo koštičavih voćaka na kajsiji i šljivi, koje za sada ne predstavlja ozbiljan problem. Cilj rada je da doprinese poznavanju simptomatologije i epidemiologije bakterioznih i fitoplazmoznih oboljenja koštičavih voćaka, kako bi se donela pravilna i pravovremena dijagnoza od koje zavisi uspešnost mera zaštite.
