primene: ciprodinil + fludioksonil (25μg/10ml vode), fluksapiroksad + difenokonazol
(100μl/10ml vode), azoksistrobin + difenokonazol (50μl/10ml vode), difenokonazol
(25μl/10ml vode) i azoksistrobin (50μl/10ml vode).
koncentraciju (MIC).
Najviši stepen tokstičnosti ispoljila je kombinacija azoksistrobina i difenokonazola, sa
prosečnom vrednosti minimalne koncentracije inhibicije od 0,003 μl/ml; zatim kombinacija
ciprodinila i fludioksonila - MIC 0,02 μg/ml; kombinacija fluksapiroksada i difenokonazola -
MIC 0,08 μl/ml, dok je najnižu toksičnost ispoljio difenokonazol - MIC 0,2 μl/ml.

AB - Identifying the precise pathogens responsible for specific plant diseases is imperative for implementing targeted and efficient interventions and mitigating their spread. Dieback and shoot blight significantly diminish the lifespan and productivity of blueberries, yet the causative agents remain largely unidentified. To determine the identity and prevalence of the causal agents of branch dieback and shoot blight, we conducted multi-year and multi-site sampling of diseased highbush blueberries (Vaccinium corymbosum cv. Duke) in Serbia. Sixty-nine monosporic isolates were collected and characterized based on morphological, physiological features and multi-locus phylogenetic analysis of internal transcribed spacer (ITS), β-tubulin (TUB2) and translation elongation factor 1-alpha (TEF1) sequence data. Five species were identified as causal agents: Diaporthe eres (36 isolates), D. foeniculina (3 isolates), Neopestalotiopsis vaccinii (9 isolates), N. rosae (6 isolates) and Neofusicoccum parvum (15 isolates). The results of the pathogenicity tests performed with the 23 representative isolates confirmed the role of these species as primary pathogens in causing dieback and shoot blight of blueberry, with N. parvum being the most aggressive and D. eres the least. Our study underscores the diversity of genera and species of ascomycetes capable of causing blueberry dieback and shoot blight. Furthermore, our findings indicate that the agents responsible for the disease in Serbia differ from those identified in other regions worldwide.

AB - Garlic (Allium sativum L.) is the second most cultivated Allium species worldwide. Many viral, fungal and bacterial diseases attack garlic plants and can be a limiting factor to garlic production. Fungal diseases control is mainly conducted by fungicides, which have detrimental impact on the environment. In this work, we explore the possibility of utilizing essential oils (EOs) as antifungal agents against a fungal isolate originated from symptomatic garlic cloves from Kraljevci locality, Srem District, Serbia, morphologically and molecularly identified as Fusarium verticillioides. EOs used in this study originated from several medicinal plants: Turkish pickling herb (Echinophora tenuifolia), oregano (Origanum vulgare), basil (Ocimum basilicum) and myrtle (Myrtus communis). Minimum inhibitory concentrations (MIC) were determined by microdilution method in 96 well microtiter plates. Microtiter plates were incubated for five days at 28°C. The experiment was repeated four times with trifloxystrobin as a positive control. The lowest concentrations without visible growth were defined as the minimal concentrations inhibiting fungal growth. Fungal spores were washed from the surface of potato dextrose agar (PDA) and spore suspension was adjusted to a concentration of approximately 5.0x104 in a final volume of 100μl per well. The values of minimal inhibitory concentration (MIC) were carried out by Duncan’s multiple range tests. An analysis of variance was performed on MIC data for four EOs applied on F. verticillioides. A significance was evaluated at p<0.05. STATISTICA v.7 (StatSoft, Inc.) was used for statistical analyses. The results of the antimicrobial activity of EOs using microdilution method showed a wide range of antifungal activity against F. verticillioides. The basil EO proved to be the most potent one (MIC-0,325±5,10μg/mL), followed by oregano (MIC-0,775±0,05μg/mL), myrtle (MIC-5,5±0,05μg/mL) and Turkish pickling herb (MIC-55±5,10μg/mL). The data obtained here suggest that the selected EOs can be applied as inhibitors to prevent growth of the phytopathogenic fungus F. verticillioides.

AB - Tomato (Solanum lycopersicum, L.) is one of the most widely cultivated crops with high content of vitamins and antioxidant lycopene, which are very important for human health. During the growing season and postharvest storage tomato is susceptible to various diseases caused by pathogenic fungi. In July 2019, tomato (cv. Balkan) with symptoms of blue mold decay were collected from market in Belgrade, Serbia. Macroscopic morphology of three obtained monosporic isolates were observed after growth on Czapek yeast autolysate agar (CYA), creatine sucrose agar (CREA), and malt extract agar (MEA) for seven days at 25ºC. Also, selected isolates were incubated at 5, 25, and 37°C for one week on CYA to monitor the effect of different temperature incubation conditions. Colony characteristics and micromorphology of the fungi agreed with the literature descriptions of Penicillium expansum. The conidiophores of isolates were hyaline, mainly terverticillate; stipes usually smooth-walled; metulae and phialides cylindrical; conidia ellipsoidal (3-3,86-4 × 3-3,13-4 μm), greenish, smooth-walled. Total DNA was extracted using DNeasy Plant Mini Kit (Qiagen, Hilden, Germany) and partial β-tubulin (BenA) sequence was amplified with primers Bt2a/Bt2b. BenA sequence of representative isolate ParP/1 was deposited in NCBI GenBank (Accession No. ON186699). Phylogenetic analysis clustered our isolate with other isolates of P. expansum. Pathogenicity test was conducted on symptomless, detached tomato fruits. All tested isolates caused typical blue mold symptoms on tomato fruits after seven days of incubation. To our knowledge, this is the first report of P. expansum causing postharvest fruit decay on tomato in Serbia.

To our knowledge, this is the first report of P. expansum causing postharvest fruit decay on tomato in Serbia.",

AB - The distribution and frequency of potato leaf roll virus in the four most important potato growing regions in Serbia were studied during the seven years (2012–18). One hundred randomly collected potato tubers were sampled from each seed lot. The young leaves that developed in three weeks were sampled and tested to record infection rate. The presence of potato leaf roll virus was detected by double-antibody sandwich enzyme-linked immunosorbent assay and disease incidence was calculated using standard formula. The obtained result showed that the highest prevalence of potato leaf roll virus was detected from seed potato samples originated from the Raški region during 2018 (20.7%), while in the Moravički region, only 2.3–11.1% of the potato leaf roll virus was detected every year. The average annual potato leaf roll virus infection was the highest in 2012 (8.4%) and 2018 (8.0%). For further confirmation of potato leaf roll virus infection, reverse transcription (RT)-PCR was performed using specific primers PLRVCPvEcoRI /PLRVCPcNcoI, designed to amplify a 650 bp fragment of the full-length coat protein gene. The PCR products derived from 26 isolates were directly sequenced using the same primer pair as in RT-PCR. The coat protein sequence analysis revealed that the Serbian potato leaf roll virus isolates showed very low nucleotide diversity (95.9–100%). They shared the highest nt identities of 98.08–99.36% with the sequences of potato leaf roll virus isolates deposited in the GenBank from other parts of the world. Phylogenetic analysis and the haplotype network of the coat protein gene sequences showed that the Serbian potato leaf roll virus isolates could be classified in two different groups indicating two possible introductions of the virus to Serbia. The results of this study confirmed the importance of potato leaf roll virus in seed potato production in Serbia. Additionally, this research highlights the need for a continuous monitoring of the potato seeds produced in Serbia as well as imported seeds for the presence of potato leaf roll virus.

AB - Leaf spot disease caused by Cercospora beticola Sacc. is the most destructive foliar disease of beet. Cercospora leaf spot is controlled primarily by fungicides because the non-chemical alternatives do not provide commercially viable control. One of the ways of reducing chemical application is the use of different essential oils (EOs) or antagonistic plant growth-promoting rhizobacteria (PGPB). This study evaluates several EOs and PGPB belonging to Pseudomonas chlororaphis as possible control agents of this pathogen. Antifungal properties were determined by in vitro microdilution method against five C. beticola monosporial isolates originated from the locality Brus, Serbia (53°53’ N, 21°04’E and 429 m above sea level) using EOs from medicinal plants: Turkish pickling herb (Echinophora tenuifolia), oregano (Origanum vulgare), basil (Ocimum basilicum), and myrtle (Myrtus communis) obtained by a hydro-distillation method. All tested oils displayed some antifungal activity against the fungal isolates. Origanum vulgare EO demonstrated the strongest antifungal activity (MIC - 0.0055±0.0051mg/mL), Ocimum basilicum slightly lower (MIC - 0.075±0.045mg/mL), followed by Myrtus communis (MIC - 0.775±0.045 mg/ mL) and Echinophora tenuifolia (MIC - 7.75±4.5 mg/mL). Five tested P. chlororaphis strains exhibited some antagonistic effect against C. beticola. Overnight culture (ONC) of P. chlororaphis strain E65 induced the highest percentage of inhibition (75.8%), followed by N3 (72.0%). A cell-free supernatant (CFS) and the CFS treated with EDTA (CFS-EDTA) of these strains showed similar inhibition of 60.2 and 56.0%, and both strains suppressed C. beticola growth. P. chlororaphis strains M1 and K113 also reduced the fungal growth by 67-70% using ONC and between 48-57% using different CFS fractions. The strains L1 and B25 caused inhibition of 60% using ONC and 50% by CFS. The lowest inhibition (~40%) by CFS-EDTA and heat-treated cell-free supernatant (HT-CFS) was recorded for B25, which was used as a reference strain. The tested isolates of C. beticola were susceptible to all selected essential oils and P. chlororaphis strains E25, N3, M1, and K113 in vitro, making them a promising non-chemical control agent. It is recommended that these findings should be tested in field conditions.

AB - Talaromyces minioluteus is one of the important species of genus Talaromyces, which has cosmopolitan distribution and is encountered on a wide range of different habitats. This species has not been considered as an important plant pathogen, even though it has been isolated from various plant hosts. Fruits and vegetables with Penicillium-like mold symptoms were collected from 2015 to 2017 from markets in Serbia. Isolates originating from quince, tomato, and orange fruits, onion bulbs, and potato tubers were identified and characterized on a morphological, physiological, and molecular level. Morphological and physiological examination included observing micromorphology, testing growth on six different media and at five different temperatures, and production of three enzymes. Molecular identification and characterization were performed using four molecular markers: internal transcribed spacer, β-tubulin, calmodulin, and DNA-dependent RNA polymerase II second largest subunit. The results of morphological and molecular analyses were in agreement, and they proved that the obtained isolates are T. minioluteus. In the pathogenicity assay, T. minioluteus was confirmed as a pathogen of all species tested with the exception of potato tubers. This is the first report of T. minioluteus as a postharvest plant pathogen on quince, tomato, and orange fruit and onion bulbs. Also, this is the first record of T. minioluteus in Serbia.

AB - Aims: A multidisciplinary approach was used to compare phenolic composition, radical scavenging and antimicrobial activity of propolis samples from different geographical localities, and plant resin against various microorganisms. Methods and Results: Using UHPLC-qqqMS quantitative analysis, 28 phenolic compounds were determined. Caffeic and p-coumaric acids were identified as main phenolic acids in poplar propolis samples, except samples from Russia (P6) and China (P7). Radical scavenging activity (applying DPPH spectrophotometric assay) showed the highest activity of Serbian (40·51%) and Chinese (53·21%) propolis samples. Broth microdilution method was used for the oral cavity, fungal phytopathogenic and human vaginal isolates which have been identified at a molecular level. The most sensitive bacterial isolates were Lactobacillus acidophilus (MIC of 0·03–0·13 mg ml−1) and the oral streptococci isolates (MIC values of 0·19–0·13 mg ml−1). The most sensitive fungal phytopathogenic isolate was Fusarium oxysporum (MIC 0·003 mg ml−1). All samples, except propolis from Serbia (P4) and Turkey (P5), showed a strong antifungal activity against Fusarium sporotrichioides, Fusarium subglutinans and Fusarium proliferatum. Conclusion: The results of various tests indicate good radical scavenging and antimicrobial activity against important human and plant pathogens. Significance and Impact of the Study: A detailed propolis analysis is important when proposing a preparation of new biological antimicrobial products which have a positive impact on human health and reduce antibacterial resistance.

AB - The liquid chromatography tandem mass spectrometry (LC-MS/MS) method for the citrinin (CIN) determination in red rice was developed. The mycotoxin was extracted from red rice using anetonitrile/water/acetic acid mixture followed by the clean-up step on Captiva EMR cartridges. The developed method was validated according to the Commission Regulation No. 401/2006/EC. The validation data were evaluated in terms of recoveries, reproducibility, limits of quantification (LOQ), limit of detection (LOD), specificity, linearity and matrix effects for CIN in matrix using 13C20-OTA as an internal standard. The obtained method performance parameters indicate that the method is suitable for the CIN routine analysis.

AB - This study assesses the potential of antibiotics-producing Pseudomonas chlororaphis strains to increase potato yield and to inhibit the mycelial growth of phytopathogenic fungi Fusarium oxysporum (Fo) isolated from potato. P. chlororaphis subsp. aurantiaca strain Q16 (PchlQ16) caused the highest growth inhibition (67.07%) of FoA2 isolate in vitro. In field trials the effect of PchlQ16 was measured as the number of stems, number and weight of tubers and a total potato yield of the Rudolph potato variety. Application of P. chlororaphis and the number of treatments exhibited a significant effect on the yield. Two treatments of PchlQ16 increased the total yield of tubers from 4.9% to 33.05%, while four treatments from 9.3% to 92.35%, compared to the control. Based on our field results we can recommend a frequent application of PchlQ16 (4 times) during potato growth season. The results of our in vitro experiment support these findings as the bacterial strain suppressed growth of F. oxysporum. In this investigation PchlQ16 was confirmed as an effective growth promoting agent in potato production and can be highly effective in prevention of F. oxysporum infection.

AB - The undemanding LC-MS/MS method was developed for the synchronized analysis of atropine and scopolamine in maize crop. The dSPE was carried out with 1% acetic acid in acetonitrile/water and a mixture of magnesium sulphate, sodium chloride and sodium citrate. The analytes were separated on a Zorbax XDB C18 column using methanol/water as the mobile phase in gradient mode. The detection was done using a tandem masss spectrometry (MS/MS) in the positive ESI. The tropane alkaloids exhibited excellent linearity in the range of 2-20 µg/kg with the LOQ of 5 µg/kg for maize. The extraction recoveries of atropine and scopolamine were 65.7 and 85.5% with the intraday RSDr 10.25 and 4.29%, respectively. The validated method was appled to real maze samples. One sample contained 18.8 µg/kg of atropine and 6.3 µg/kg of scopolamine.

abstract = "The undemanding LC-MS/MS method was developed for the synchronized analysis of atropine and scopolamine in maize crop. One sample contained 18.8 µg/kg of atropine and 6.3 µg/kg of scopolamine.",

AB - It is essential that certified potatoes are free from known viruses which can negatively affect quality and yield. However, very little is known about the distribution and frequency of Potato virus S (PVS) in Serbia. Until 2014, PVS was present sporadically in the domestic seed potato production. The incidence of PVS was studied by a molecular method over 3 years (2014-2016) in four important potato-growing regions (Moravidd. Zlatiborski, Raski and Macvanski) and in different cultivars. The results showed that the incidence of PVS increased steadily over 3 years from 1.52 to 8.84%. The Moravicki region had the highest incidence (13.06%) and Desiree was the most susceptible cultivar with a mean PVS incidence of 8.2% followed by Marabel and Riviera. The highest significant statistical difference was between the cultivars and in the interaction between cultivars and regions. A detailed phylogenic analysis of the tested isolates contained that Serbian PVS belongs to PVSO. Of the 18 Serbian PVS isolates included in this study, eight were grouped into the PVSO cluster and formed a subgroup (O-I) with isolates from the USA, Syria, Korea and Chile. Ten Serbian isolates of PVS together with the isolates from Iran were clustered in a branch of subgroup O-VII. This study constitutes the fast report of PVS isolates in Serbia which are capable of infecting Chenopodium quinoa and inducing the symptoms of local chlorotic lesions.

AB - The control of Plasmopara viticola and Botrytis cinerea, two of the most dangerous pathogens on grapevine, requires frequent treatments with chemicals based on weather conditions. Numerous applications of fungicides have resulted in developing fungicide resistance. Active ingredients based on copper have been used very successfully for a long time to protect grapevines against these pathogens. Application of a copper citrate formulation with high degree dissociation at a very low concentration (1%) was evaluated in field trials. The efficacy of two concentrations of copper citrate, 0.5 and 1.0%, were tested against P. viticola on grapevine in three locations, and against B. cinerea in two locations during 2018. Our results demonstrated that the concentration of 1.0% copper citrate was highly effective against P. viticola (87.4%) and B. cinerea (63.7%), compared to standard treatment (90.6 and 53.1%), under a high level of infection.

AB - The essential oil of Lavandula stoechas was examined by GC and GC-MS. Discs (5 mmi.d.) of the tested fungi (Alternaria alternata, Fusarium oxysporum and Botritys cinerea) were inoculated separately onto each assay plate and incubated at 25 degrees C for 7 days. The oil yield of dried parts (v/dw) obtained by hydro distillation was 2.9%. Thirty-two compounds representing 98.3% of the essential oil were determined. Linalool (49.9%), linalyl acetate (14.4%), lavandulyl acetate (5.7%), alpha-terpineol (5.6%), terpinene-4-ol (5.1%), lavandulol (3.7%), (E)-beta-ocimene (2.6%) and (Z)-beta-ocimene (2.4%) were identified as the main constituents of the oil. In addition, both doses of the lavender oil showed varying levels of inhibitory effects on the mycelial growth of tested fungi used in the experiment. The results demonstrated the strongest effect on B.cinerea, followed by A.alternata and F.oxysporum. The inhibitory effect is probably dependent on the concentration of essential oils.

AB - Fusarium species are widely distributed and responsible for several plant diseases in different medicinal plants. Fungi of this genera cause very important economic losses in Serbian plantation. Antibiotic production by plant-associated microorganisms represents an environmentally acceptable method of disease control, esspecialy in cultivation of medicinal and aromatic plants. Among the plant growth promoting bacteria (PGPB), Pseudomonas have been recognized as the most frequent antagonists of plant fungal pathogens and antibiotic producers. This is probably due to the widely distribution of this diverse group of bacteria in temperate soils and their often predomination among bacteria from plant rhizosphere. In this study, we examined the antifungal activity of eleven indigenous Pseudomonas isolates (PB4, PB5, K38, Q34, PBA12, PD5, C7, C8, Q16P, K29 and K35) against eight phytopathogenic fungi belonging to genus Fusarium (Fusarium tricinctum, F. sambucinum, F. equiseti, F. heterosporum, F. sporotrichioides, F. semitectum, F. verticillioides and F. oxysporum), which had infected anise (Pimpinella anisum L., fam. Apiaceae), using in vitro growth inhibition tests. The obtained results demonstrated that all Pseudomanas isolates showed more or less pronounced antifungal activity, whereby the most pronounced activity was observed for K29 and K35 strains. F. oxysporum and F. verticillioides showed the highest sensitivity to antibiotic-producing Pseudomanas isolates. In general, it has been concluded that studied Pseudomonas isolates have potential in controlling plant diseases caused by Fusarium spp., whereby the bacterial isolates with the highest inhibitory potential will be selected for further experiments.
