In-Depth Characterization of Crown Gall Disease of Tobacco in Serbia

AB  - In August 2020, the unusual appearance of crown gall symptoms was observed on the
tobacco plants (hybrid PVH2310) grown in fields in the Golubinci (Srem district, Serbia) locality. The
causal agent isolated from galls located on tobacco roots formed circular, convex, and glistening
light blue colonies, and then dark to olive-green-colored bacterial colonies on a semi-selective D1
medium. Molecular analysis based on multiplex PCR and multi-locus sequence analysis (MLSA)
using concatenated sequences of the atpD, dnaK, glnA, and rpoB genes as well as 16S rRNA identified
Serbian tobacco isolates such as Agrobacterium tumefaciens (biovar 1). Two duplex PCR methods
confirmed the presence of the virD2 and virC genes in tobacco isolates. Pathogenicity tests performed
on carrot discs and squash fruits resulted in tumor/gall formation after 12 to 16 days post inoculation,
respectively. Pathogenicity was also confirmed on tobacco plants, where isolates caused tumor
development 21−25 days after inoculation. API 50 CH generated results regarding the biochemical
features of the Serbian tobacco isolates. As A. tumefaciens (biovar 1) as a cause of tobacco crown gall
has previously been documented solely in Japan, there is presently no data on its wider occurrence.
Therefore, this first detailed investigation of A. tumefaciens isolated from naturally infected tobacco in
Serbia will contribute to a better understanding of it at the global level.

Pseudomonas cerasi, the new wild cherry pathogen in Serbia and the potential use of recG helicase in bacterial identification

AB  - In May 2016, an unusual appearance of leaf spot (water-soaked, brown-purple, round to angular surrounded with yellow halos) was observed on the leaves of wild cherry specimens grown in Rimski Šančevi, Vojvodina (North Serbia). The causal pathogen was isolated from the wild cherry diseased leaves on Nutrient Agar supplemented with 5% sucrose and identified as Pseudomonas cerasi based on multilocus sequence analysis (MLSA). PCR amplification and sequencing of four housekeeping genes—gapA, gltA, rpoD and gyrB—showed 100% (gapA, gltA), 99.81% (rpoD) and 99.67% (gyrB) identity with P. cerasi type strain CFBP8305T (=58T = LMG28609T) and strain PL963 sequences from NCBI database. Pseudomonas cerasi isolates (coded as RE10-RE19) were LOPAT +− − − + (Pseudomonas Group Ia) and GATTa + − + −, produced fluorescent pigment, were able to utilise lactic and aspartic acid, but not tartaric acid. All isolates were pathogenic on wild cherry seedlings and leaves, immature sweet and sour cherry fruitlets, as well as on lilac leaves and green bean pods. This study also aimed to design new primers (recG-F/recG-R) for amplification of recG gene (encoding ATP-dependent DNA helicase RecG) known for its crucial role in DNA recombination and repair. Amplification of this gene enabled high identification ability of our wild cherry isolates as well as isolates belonging to the closely related stone fruit pathogenic Pseudomonas species. The obtained results highlight the potential of using recG gene for the specific detection and the identification of pathogenic Pseudomonas syringae complex. This research presents the first report of P. cerasi infecting wild cherry as well as first description of this bacterium in Serbia. Obtained results indicate the risk from further spread of this bacterium with the infected cherries rootstock, having a key role in its epidemiology.

Incidence of Grapevine Fanleaf Virus (GFLV) and Grapevine Leafroll-Associated Viruses (GLRaV 1− 3) in Vojvodina Province

AB  - During May and June of 2021, a total of 123 grapevine leaf samples were collected and analyzed for infection by grapevine fanleaf virus (GFLV) and three viruses from the grapevine leafroll-associated virus complex -1, -2 and -3(GRLaV-1, GRLaV-2 and GRLaV-3, respectively). The samples were collected from commercial vineyards, small backyard vineyards and grapevine nurseries located across the entire Vojvodina Province (Bačka, Banat and Srem).OEPP/EPPO sampling protocols were followed during the sampling of leaf tissues showing possible virus infection symptoms. Among the 123 samples, 47 were collected from Bačka region (Bačko Gradište − 11, Bečej − 10, Temerin− 15, Vrbas − 1, Hajdukovo − 10), 50 from Banat region (Srpska Crnja − 10, Vojvode Stepe − 10, Čoka − 10,Uljma −20) and 26 from Srem region (Šid − 6, Banoštor − 10, Sremski Karlovci − 10) and serological ELISA testswere performed for virus detection. GFLV was detected in five samples (4.06%), GLRaV-1 was detected in six samples (4.87%), while GLRaV-2 was not detected in any of the analyzed grapevine samples. GLRaV-3 was present in five samples (4.06%). When infection rates were examined in relation to cultivars, GFLV was detected in Cardinal, Zalagyöngye, Black Muscat, Italian Riesling and Dornfelder. GRLaV-1 was detected in cultivars Cardinal,Black Muscat, Italian Riesling and Merlot, and GRLaV-3 in cultivars Othello and Italian Riesling. Based on these results, it can be concluded that GFLV and GRLaV-1 and GRLaV-3 are present in vineyards across Vojvodina Province and affect different grapevine cultivars. To effectively control virus infections and their spreading, continuous monitoring of these viruses and their vectors is required along with the planting of healthy propagation material.

NEW DATA ON PRESENCE OF CANDIDATUS PHYTOPLASMA SOLANI ON POTATO IN VOJVODINA PROVINCE

AB  - BACKGROUND Bačka (Vojvodina, Serbia) is one of the main potato-growing areas in Serbia. The presence of the stolbur-associated symptoms in the form of reddening and leaf rolling, shortened internodes, and aerial tuber formation were frequently noticed on the potato plants. Therefore, the identification of the causal agent of the disease was initiated, considering that presence of phytoplasmas was not earlier reported in Maglić locality. OBJECTIVES This research aimed to identify the causal agent of the phytoplasmas’ disease in potato field (unknown cultivar) in locality Maglić (Bačka) in 2021. METHODS Samples for the identification within the survey site consisted of pooled multiple diseased plants (leaves and tubers). Genomic DNA from the ten pooled samples was extracted using the Cetyltrimethyl ammonium bromide (CTAB) method. Identification was performed with polymerase chain reaction (PCR) using universal phytoplasma 16S rRNA primer pair P1/P7. The obtained sequences were checked for identity with the deposited strains in the NCBI database using the BLASTn search tool. Further, Neighbour-joining (NJ) tree was made to reconstruct the phylogeny of potato strains in relation to deposited strains in the NCBI database. RESULTS PCR amplification performed with primer pair P1/P7 resulted in amplicon generation at 1800 bp for three (two from tubers and one from leaves) out of a total of ten tested samples. BLASTn analysis revealed 100% identity of potato strains with deposited strains of Candidatus Phytoplasma solani, the 16SrXII-A ribosom al subgroup. NJ phylogenetic analysis grouped potato strains in the same tree cluster with NCBI deposited strains of Ca. Phytoplasma solani.

AB  - The present study was conducted on 106 leaf samples of Prunus species (44 plum, 47 myrobalan plum, 10 apricot, 3 peach, 1 blackthorn, and 1 sour cherry) collected from the Vojvodina province of Serbia and assessed for the presence of Plum pox virus (PPV) with the aim of establishing whether trees growing along roadsides and in backyards could be sources of PPV infection in commercial orchards. Analyses confirmed PPV infection in 68.2%, 38.6%, and 20% of plum, myrobalan plum and apricot samples, respectively. The infection level varied considerably across locations, with 80% noted in Uljma, followed by 69% in Sremska Kamenica, 60% in Bečej, 50% in Temerin, 50% in Senta, 40% in Petrovaradin, 31% in Novi Sad, 30% in Susek, and 20% in Subotica. As no difference in infection rates was noted between young and older plum trees, the infections seemed to have occurred in their early life stage. Trees growing along roadsides were infected to a higher extent (89.6%) than those in backyards (66.6%). In some cases, symptomless trees were also proven
to be PPV positive, indicating that they are a potentially important source of further virus infections. Due to the widespread presence of the virus, tolerant or hypersensitive cultivars are recommended for planting new orchards.
