Revision of the European Lysiphlebus species (Hymenoptera: Braconidae: Aphidiinae) on the basis of COI and 28SD2 molecular markers and morphology

AB  - Members of the genus Lysiphlebus Foerster are medium-sized endoparasitoid aphidiine wasps with over 30 species distributed worldwide. They attack numerous aphid hosts, and some species are important biological control agents. All European Lysiphlebus species are revised based on sequence information from the mitochondrial COI barcoding gene and the nuclear 28SD2 gene, morphological traits, and on information about their host specificity. The taxonomic status of 13 European Lysiphlebus species is confirmed, while 11 are redescribed and illustrated. The following four Lysiphlebus species are synonymized: L. melandriicola Stary, 1961 [= L. fabarum (Marshall, 1896)], L. hirtus Stark, 1985 (= L. confusus Tremblay & Eady, 1978), L. hispanus Stary, 1973 (= L. desertorum Stary, 1965), and L. safavii Stary, 1985 (= L. hirticornis Mackauer, 1960). Two new Lysiphlebus species are described: L. volkli sp.n. and L. brachycaudi sp.n. Our results, based on both COI and 28SD2, confirm the existence of three monophyletic species groups within the genus: (1) the 'fabarum' group, which includes L. fabarum, L. cardui (Marshall, 1896), L. confusus, L. hirticornis, L. brachycaudi sp.n., and L. volkli sp.n.; (2) the 'testaceipes' group, which includes L. testaceipes (Cresson, 1880), L. orientalis Stary & Rakhshani, 2010, L. fritzmuelleri Mackauer, 1960, L. balcanicus Stary, 1998, L. desertorum, [and probably L. dissolutus (Nees, 1811)]; and (3) the 'alpinus' group, consisting solely of L. alpinus Stary, 1971. Geometric morphometrics of fore wing shape proved to be a powerful tool to delineate sibling species within the fabarum' group. We were able to confirm for the first time sexual lines of L. cardui and L. confusus. Furthermore, an additional potentially asexual Lysiphlebus taxon in Europe is revealed based on the discovery of an asexual line of L. volkli sp.n. in Iran. An illustrated key for identification of the European Lysiphlebus species is provided.

AB  - Background: Morphological divergence often increases with phylogenetic distance, thus making morphology taxonomically informative. However, transitions to asexual reproduction may complicate this relationship because asexual lineages capture and freeze parts of the phenotypic variation of the sexual populations from which they derive. Parasitoid wasps belonging to the genus Lysiphlebus Foerster (Hymenoptera: Braconidae: Aphidiinae) are composed of over 20 species that exploit over a hundred species of aphid hosts, including many important agricultural pests. Within Lysiphlebus, two genetically and morphologically well- defined species groups are recognised: the "fabarum" and the "testaceipes" groups. Yet within each group, sexual as well as asexual lineages occur, and in L. fabarum different morphs of unknown origin and status have been recognised. In this study, we selected a broad sample of specimens from the genus Lysiphlebus to explore the relationship between genetic divergence, reproductive mode and morphological variation in wing size and shape (quantified by geometric morphometrics). Results: The analyses of mitochondrial and nuclear gene sequences revealed a clear separation between the "testaceipes" and "fabarum" groups of Lysiphlebus, as well as three well-defined phylogenetic lineages within the "fabarum" species group and two lineages within the "testaceipes" group. Divergence in wing shape was concordant with the deep split between the "testaceipes" and "fabarum" species groups, but within groups no clear association between genetic divergence and wing shape variation was observed. On the other hand, we found significant and consistent differences in the shape of the wing between sexual and asexual lineages, even when they were closely related. Conclusions: Mapping wing shape data onto an independently derived molecular phylogeny of Lysiphlebus revealed an association between genetic and morphological divergence only for the deepest phylogenetic split. In more recently diverged taxa, much of the variation in wing shape was explained by differences between sexual and asexual lineages, suggesting a mechanistic link between wing shape and reproductive mode in these parasitoid wasps.

AB  - In this paper we apply an integrative approach combining morphometric and molecular analyses to explore parasitoids of the Praon dorsale-yomenae s.str. complex. These parasitoids occur in natural and agricultural ecosystems throughout the Palaearctic and parasitize aphid hosts belonging to the tribe Macrosiphini. The P. dorsale-yomenae species complex represents a morphologically cryptic group, consisting of several hidden taxa with specific host adaptations and distributions. For the morphometric analyses we used a large dataset comprising 230 female specimens that emerged from 30 different species of aphid hosts throughout the Palaearctic. The molecular analysis included a reduced dataset of 44 specimens that emerged from 19 aphid hosts. The mitochondrial cytochrome oxidase subunit I (COI) and the nuclear second expansion segment of the 285 rRNA gene (28SD2) were used to estimate a genetic diversity within this complex. Although all Praon species clustered closely together in the 28SD2 tree, confirming their close relatedness, the molecular characterization based on COI identified five clearly separate taxa with sequence divergences in the range of 4.7-8.9%. These taxa also exhibited significant differences in forewing shape as revealed by geometric morphometric analyses. Classical morphometric analyses revealed the length of m-cu vein as a new taxonomic character, but suggested that one commonly used trait, the color pattern of the Rs + M and m-cu veins, cannot be used for species distinction as it was highly variable in one of the taxa. Based on the combined morphometric and genetic data, we confirm the species status of Praon dorsale, P. yomenae, P. longicorne and P. volucre, and describe and illustrate a new parasitoid species of the "dorsale-yomenae" group, i.e. Praon staticobii n.sp. associated with Staticobium limonii on Limonium angustifolium. It has rapidly become widespread in coastal areas continuing gradually to expand inland. L. testaceipes exploited a large number of aphids in Europe, including new hosts and significantly changed the relative abundance of the native parasitoids. This behavior may reflect a broad oligophagy of the introduced parasitoid or it may require the evolution of host specialization that results in genetically differentiated subpopulations on different hosts. To address this issue we used the mitochondrial cytochrome oxidase subunit I and seven microsatellite loci to analyze the structure of genetic variation for L. testaceipes samples collected from 12 different aphid hosts across seven European countries, as well as some samples from Benin, Costa Rica, USA, Algeria and Libya for comparison. Only five COI haplotypes with moderate divergence were identified overall. There was no evidence for the association of haplotypes with different aphid hosts in the European samples, but there was geographic structuring in this variation. Haplotype diversity was highest in France, where L. testaceipes was introduced, but only a single haplotype was detected in areas of south-eastern Europe that were invaded subsequently. The analysis of microsatellite variation confirmed the lack of host-associated genetic structure, as well as differentiation between populations from south-western and southeastern Europe. The parasitoid L. testaceipes in Europe is thus an opportunistic oligophagous species with a population structure shaped by the processes of introduction and expansion rather than by host exploitation. Haplotype diversity was highest in France, where L. testaceipes was introduced, but only a single haplotype was detected in areas of south-eastern Europe that were invaded subsequently. The analysis of microsatellite variation confirmed the lack of host-associated genetic structure, as well as differentiation between populations from south-western and southeastern Europe. The parasitoid L. testaceipes in Europe is thus an opportunistic oligophagous species with a population structure shaped by the processes of introduction and expansion rather than by host exploitation. AB  - The genus Praon represents a large group of aphid endoparasitoids and is exemplary for the problems encountered in their taxonomy because of a great variability of morphological characters. To investigate the intraspecific variability and to ascertain cryptic speciation within the Praon abjectum Haliday group, biotypes in association with the aphid hosts Aphis sambuci L., Longicaudus trirhodus Walker, and Rhopalosiphum spp. were examined. We combined molecular and geometric morphometric analyses, that is, partial sequences of the mitochondrial cytochrome oxidase subunit I and nuclear 28SD2 genes and the shape of the forewing. Low variation of 28SD2 sequences confirmed the close relatedness of species from the genus Praon. Analysis of the cytochrome oxidase subunit I sequences however identified three separate taxa within the P. abjectum group with substantial genetic divergence. The biotype of P. abjectum associated with L. trirhodus differed from those associated with Rhopalosiphum sp. and A. sambuci by 5.4-6.5% and 7.7% sequence divergence, respectively, while the genetic distance between the latter two biotypes ranged from 9.5 to 10%. The main changes in the forewing shape that discriminate these three biotypes as revealed by geometric morphometrics are related to the stigma shape and the position of the radial nerve. Based on the differences determined in mitochondrial sequences and in the shape of the wing, we describe two new cryptic species within the P. abjectum group as follows: P. sambuci sp. n. in association with A. sambuci/S. nigra and P. longicaudus sp. n. in association with L. trirhodus/T. aquilegifolium.
