Genetic diversity of Pseudomonas syringae pv. syringae isolated from sweet cherry in southern and northern regions in Serbia

AB  - Bacterial canker and leaf spot caused by plant pathogenic bacterium Pseudomonas is among the most destructive cherry diseases worldwide. Nowadays in Serbia, sweet cherry production significantly increased and the new plantations, mainly grown from imported planting material are being raised every year. During spring, 2018 and 2019, occurrence of bacterial canker and leaf spot symptoms was observed on a newly planted sweet cherry plantations in two localities, Zitorada (Southern region) and Karavukovo (Northern region-Vojvodina). Typical P. syringae colonies were isolated on Nutrient Sucrose Agar supplemented with 5% sucrose (NSA). A total of fifteen isolates were selected and identified. Results of the LOPAT test (+---+) determined them to belong to fluorescent Pseudomonas Group Ia, while results of G(+)A(+)T(-)Ta(-) tests indicate presence of Pseudomonas syringae pv. syringae. Pathogenicity was confirmed on immature sweet and sour cherry fruitlets by forming of black, sunken lesions for all tested isolates. Genes syrB and syrD were successfully detected in all tested isolates. DNA sequencing using gapA, gltA, gyrB and rpoD housekeeping genes determined tested isolates to belong to P. s. pv. syringae using the National Center for Biotechnology Information (NCBI) nucleotide BLAST. The Serbian isolates shared 99.47% to 100% (Zitorada) and 99.38% to 100% (Karavukovo) identity with bacterium P. s. pv. syringae. Phylogenetic analysis grouped isolates from Zitorada in one tree cluster, separate from the Karavukovo isolates,indicating presence of two genetically diverse groups of causal pathogen P. s. pv. syringae, obtained from two geographically distinct localities in Serbia. Phylogeographic analysis grouped isolates from Zitorada in multilocus haplotype coded as REz and isolates originated from Karavukovo in multilocus haplotype coded as REk. Considering that during last few years P. syringae continuously occurs mainly in young sweet cherry plantations, where imported material is used for raising, health status check is recommended to be included as obligatory measure when nursery material is used from import.

Identification and phylogenetic analysis of Fusarium proliferatum isolated from elephant garlic Allium ampeloprasum L

AB  - Symptoms of cloves rot of A. ampeloprasum were noticed during 2018 in storage conditions. 16 isolates were obtained (AMP1-AMP16) and according to morphological and cultural characteristics they belong to F. proliferatum (sex. stage Gibberella intermedia), species of Gibberella fujikuroi complex. To confirm morphological identification, total genomic DNA was extracted from mycelium of the 16 isolates by amplification of TEF-1a gene, using polymerase chain reaction (PCR) that was performed with the primer pair EF1 and EF2. Results presented in this article clearly indicated that the new host of Fusarium proliferatum as the causal agent of cloves rot is "elephant garlic" Allium ampeloprasum. Pathogenicity test was confirmed on Allium ampeloprasum cloves. Pathogenicity assays revealed that all isolates caused symptoms on tested Allium spp., like naturally infected cloves.
AB  - Simptomi truleži uskladištenih čenova belog luka na vrsti poznatoj kao "elephant garlic" (A. ampeloprasum) zapaženi su tokom 2018. godine. Prikupljen je veliki broj zaraženih uzoraka i nakon izolacije odabrano je 16 izolata (AMP1-AMP16) za dalja istraživanja. Na osnovu morfoloških i odgajivačkih karakteristika je ustanovljeno da je prouzrokovač truleži čenova gljiva F. proliferatum (telemorf Gibberella intermedia), koja pripada kompleksu Gibberella fujikuroi. Proučavanjem patogenosti svi izolati su prouzrokovali simptome truleži na različitim vrstama roda Allium spp. koji su identični prirodnoj infekciji. U cilju potvrde morfoloških odlika izvršena je molekularna identifikacija metodom lančane reakcije polimeraze (PCR) korišćenjem para prajmera EF1 i EF2 koji amplifikuju TEF-1a gen, sekvencioniranje DNK i filogenetska analiza dobijenih sekvenci. Dobijeni rezultati potvrdili su da je Fusarium proliferatum prouzrokovač truleži na vrsti A. ampeloprasum kao novom domaćinu.

Foliar pathogens of sweet and sour cherry in Serbia

AB  - In recent years, sweet and sour cherry production in Serbia has increased. Under Serbian agroecological conditions, pathogens causing leaf diseases threaten the success of sweet and sour cherry production. In the period 2012-2019, the health status of cherries was monitored in more than 30 locations. Depending on the production system, the following leaf pathogens were identified: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina and Pseudomonas syringae pv. syringae and morsprunorum race 1. Leaf pathogens caused premature defoliation, which adversely affected bud formation for the next growing season and increased susceptibility to freezing. Therefore, attention should be focused on correct leaf pathogen identification, and proper selection, application and rotation of fungicides.
AB  - Poslednjih godina proizvodnja trešnje i višnje u Srbiji je povećana. U našim agroekološkim uslovima, patogeni lista ugrožavaju proizvodnju ovih perspektivnih voćnih vrsta. U periodu 2012-2019. godine praćeno je zdravstveno stanje trešnje i višnje, na više od 30 lokaliteta. avisno od sistema proizvodnje identifikovani su sledeći patogeni lista: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina, Pseudomonas siringae pv. syringae i morsprunorum rasa 1. Patogeni lista izazivaju prevremenu defolijaciju, što negativno utiče na formiranje pupoljaka za narednu vegetaciju i povećanu osetljivost na izmrzavanje. Stoga pažnju treba usmeriti na ispravnu identifikaciju patogena, pravilan izbor i rotaciju fungicida različitog mehanizma delovanja, kao i optimalno vreme za njihovu primenu.

AB  - During the past few years, symptoms of garlic clove rot have appeared in storage facilities at different localities in Serbia. Thirteen fungal isolates were isolated from samples of decayed garlic cloves (Allium sativum L.). Based on the colony and fungal morphology, isolated fungi were tentatively identified as F. proliferatum. Pathogenicity of 13 F. proliferatum strains were confirmed by inoculating garlic cloves of Allium sativum (spring cv. Labud, Sedef; winter cv. Bosut, Ranko) and Allium ampeloprasum (cv. Biser). Isolate JBL532 was classified as the most aggressive, while isolate B3 was the least pathogenic. To confirm morphological identification of 13 Fusarium isolates, amplification of TEF-1α gene was performed using polymerase chain reaction (PCR) with the primer pair: EF1 and EF2. All isolates were cryopreserved at -80°C and deposited in the NCBI GenBank.
AB  - Tokom proteklih godina u skladištima na različitim lokalitetima u Srbiji pojavili su se simptomi truleži i popadanja belog luka (Allium sativum L.). Izolacijom je dobijeno 13 izolata gljiva roda Fusarium koji su na osnovu morfoloških karakteristika identifikovani kao F. proliferatum. Patogenost izolata je potvrđena inokulacijom različitih genotipova belog luka Allium sativum (prolećne sorte Labud, Sedef, jesenje sorte Bosut, Ranko) i Allium ampeloprasum (sorta Biser). Na osnovu ocene patogenosti, izolat JBL532 se izdvojio kao najagresivniji, dok se izolat B3 pokazao kao najmanje patogen. Morfološka identifikacija potvrđena je amplifikacijom TEF-1α gena, korišćenjem prajmera EF1 i EF2. Svi izolati se čuvaju na -80°C i deponovani su u NCBI bazu gde su im dodeljeni jedinstveni pristupni brojevi.

Evaluation of cherry cultivar susceptibility to bacterial canker and leaf spot disease

AB  - In this study, the susceptibility of different sweet (19) and sour cherry (3) cultivars (cvs.) to the bacterial pathogens Pseudomonas syringae pv. syringae and P. s. pv. morsprunorum race 1 causing bacterial canker (sensu stricto) and leaf spot was evaluated. Susceptibility to bacterial canker was examined by artificial inoculations of 1-year-old rooted trees, which revealed susceptibility of all cvs. to both P. syringae pathovars. Susceptibility level differed among cvs.; sweet cherry cvs. Katalin, Linda, Summit, New Star and Bigarreau Burlat were highly susceptible, sour cherry cvs. erdi botermo and sweet cherry Drogan's Yellow, Carmen, Germersdorfer and Noir de Meched were susceptible. Sour cherry cvs. Spanska and ujfehertoi furtos, as well as sweet cherry cv. Rita were found to be less susceptible to bacterial canker. Susceptibility to bacterial leaf spot disease was evaluated on naturally infected (field conditions) and artificially inoculated leaves (laboratory conditions). Severe symptoms of bacterial spot with shot hole appearance were observed in cherry leaves at the beginning of May under field conditions. In all collected leaf samples, presence of P. s. pv. morsprunorum race 1 was determined. Sweet cherries Valery Chkalov, Summit, Bigarreau Burlat and New Star were highly susceptible to this bacterium. No symptoms of bacterial spot were found on sweet cherries Solomary gomboly, Linda, Germersdorfer, Hedelfinger, Katalin, Margit, Carmen, Aida, Sunburst, Alex, Peter, Rita and Kavics and sour cherry erdi botermo. In laboratory conditions, susceptibility of the tested cvs. varied and mostly depended on the inoculation method. Nonetheless, sweet cherry cvs. Carmen and Margit and sour cherry erdi botermo exhibited a certain level of resistance.
