LC-MS/MS Determination of Tropane Alkaloids in Maize Crop

AB  - The undemanding LC-MS/MS method was developed for the synchronized analysis of atropine and scopolamine in maize crop. The dSPE was carried out with 1% acetic acid in acetonitrile/water and a mixture of magnesium sulphate, sodium chloride and sodium citrate. The analytes were separated on a Zorbax XDB C18 column using methanol/water as the mobile phase in gradient mode. The detection was done using a tandem masss spectrometry (MS/MS) in the positive ESI. The tropane alkaloids exhibited excellent linearity in the range of 2-20 µg/kg with the LOQ of 5 µg/kg for maize. The extraction recoveries of atropine and scopolamine were 65.7 and 85.5% with the intraday RSDr 10.25 and 4.29%, respectively. The validated method was appled to real maze samples. One sample contained 18.8 µg/kg of atropine and 6.3 µg/kg of scopolamine.

The Incidence and Genetic Diversity of Potato virus S in Serbian Seed Potato Crops

AB  - It is essential that certified potatoes are free from known viruses which can negatively affect quality and yield. However, very little is known about the distribution and frequency of Potato virus S (PVS) in Serbia. Until 2014, PVS was present sporadically in the domestic seed potato production. The incidence of PVS was studied by a molecular method over 3 years (2014-2016) in four important potato-growing regions (Moravidd. Zlatiborski, Raski and Macvanski) and in different cultivars. The results showed that the incidence of PVS increased steadily over 3 years from 1.52 to 8.84%. The Moravicki region had the highest incidence (13.06%) and Desiree was the most susceptible cultivar with a mean PVS incidence of 8.2% followed by Marabel and Riviera. The highest significant statistical difference was between the cultivars and in the interaction between cultivars and regions. A detailed phylogenic analysis of the tested isolates contained that Serbian PVS belongs to PVSO. Of the 18 Serbian PVS isolates included in this study, eight were grouped into the PVSO cluster and formed a subgroup (O-I) with isolates from the USA, Syria, Korea and Chile. Ten Serbian isolates of PVS together with the isolates from Iran were clustered in a branch of subgroup O-VII. This study constitutes the fast report of PVS isolates in Serbia which are capable of infecting Chenopodium quinoa and inducing the symptoms of local chlorotic lesions.

Efficacy of copper citrate in grapevine disease control

AB  - The control of Plasmopara viticola and Botrytis cinerea, two of the most dangerous pathogens on grapevine, requires frequent treatments with chemicals based on weather conditions. Numerous applications of fungicides have resulted in developing fungicide resistance. Active ingredients based on copper have been used very successfully for a long time to protect grapevines against these pathogens. Application of a copper citrate formulation with high degree dissociation at a very low concentration (1%) was evaluated in field trials. The efficacy of two concentrations of copper citrate, 0.5 and 1.0%, were tested against P. viticola on grapevine in three locations, and against B. cinerea in two locations during 2018. Our results demonstrated that the concentration of 1.0% copper citrate was highly effective against P. viticola (87.4%) and B. cinerea (63.7%), compared to standard treatment (90.6 and 53.1%), under a high level of infection.
AB  - Zaštita vinove loze od prouzrokovača plamenjače vinove loze - Plasmopara viticola i sive truleži - Botrytis cinerea, je vrlo kompleksna i zahteva primenu većeg broja hemijskih tretmana, u skladu sa vremenskim uslovima. Učestala primena fungicida uslovljavala je pojavu rezistentnih izolata patogena na fungicide. Različite forme bakarnih jedinjenja primenjuju se u zaštiti vinove loze dugi niz godina prilično uspešno. Primena bakar-citrata - formulacije sa visokim stepenom disocijacije u niskoj koncentraciji (1,0%) ispitivana je u poljskim uslovima. Efikasnost dve koncentracije bakar-citrata - 0.5 i 1.0% je testirana u suzbijanju P. viticola i B. cinerea na vinovoj lozi na tri (dva) lokaliteta, tokom 2018 godine. Naši rezultati pokazuju da je ispitivana koncentracija od 1,0% bakar-citrata ispoljila zadovoljavajući efekat na P. viticola (87,4%) i B. cinerea (63,7%) u odnosu na primenjene standarde (90,6% i 53,1%) u uslovima visokih zaraza.

AB  - The effects of copper citrate on defoliation of one year old fruit stocks in a nursery are presented in this paper. The investigation aimed to evaluate the defoliation efficacy of copper citrate as a new formulation, compared with other compounds recommended for that purpose. Field trials were conducted in the region of Trstenik, a center of nursery production in Serbia in 2016 and 2017. Two years of investigation showed that copper citrate could be used as an efficient compound for defoliation in nursery production. A stronger defoliation effect was observed on plum stocks, compared to apple stocks. Better results were achieved with the highest concentration of copper citrate (2%), while the effectiveness decreased with lower concentrations (0.5%, 1.0% and 1.5%). Defoliation degree on nursery apple stocks after treatment with 2.0% concentration of copper citrate varied from 80.6% to 95.6%, while it reached 100% on plum stocks.
AB  - U radu su prikazani efekti defolijacije sadnica jabuke i šljive, primenom različitih koncentarcija bakar citrata, kao nove formulacije, i standardnih preparata koji se koriste u ove svrhe (bordovska čorba, urea). Ogledi su sprovedeni u regionu Trstenika (Ljubava i Milutovac), koji je centar rasadničke proizvodnje u Srbiji. Tretiranja su obavljena tokom 2016. i 2017. godine. Na osnovu dobijenih rezultata zaključeno je da bakar citrat pokazuje visok potencijal za upotrebu u svrhu defolijacije sadnica jabuke i šljive. Korišćene koncentracije ovog jedinjenja su pokazale veću efikasnost u primeni na sadnicama šljive nego na sadnicama jabuke. Najbolji rezultati u oba ogleda su zabeleženi sa maksimalno primenjenom koncentracijom (2%), dok efekat defolijacije na sadnicama jabuke opada sa smanjenjem koncentracije. Stepen defolijacije jabuke primenom bakar citrata u koncentraciji od 2% kretao se od 80,6% do 95,6%, a kod šljive čak 100%. Potvrđeno je i da uspešna defolijacija sadnica jabuke i šljive značajno zavisi od vremena aplikacije kao i meteoroloških uslova.

Molecular characterization of potato virus Y inducing potato tuber necrotic ringspot disease in Serbia

AB  - The Potato virus Y (PVY) is the most important limiting factor for potato seed production in Serbia. Currently, PVY is a major concern for the potato seed growers. Initially, serological (ELISA) tests were carried out on 100 potato seed tubers from each of the seven potato cv. during 2013. The infection rates with the PVYN was between 5 and 36%. A complete genome sequencing of the most common Serbian isolate of PVY (3D), followed by molecular characterization and phylogenetic analysis has been performed to show what group it belongs to. Our isolate's complete genome sequence (KJ946936) showed that the Serbian PVY isolate (3D) is 99.7% identical at nt level, with other tuber necrosis strain group (PVYNTN) from Europe. Phylogenetic analysis revealed three consistent lineages of isolates, showing that our isolate was clustered with the isolates from Europe and North America in the PVYN lineage which induces potato tuber necrotic ringspot disease (PTNRD). The Serbian isolate of PVYNTN together with the isolates from Europe was clustered in the branch of European sublineage, with a high bootstrap support and no genetic diversity. This is the first study in Serbia demonstrating phylogenetic distinction between our isolate and other isolates of PVY.

Initially, serological (ELISA) tests were carried out on 100 potato seed tubers from each of the seven potato cv. during 2013. The infection rates with the PVYN was between 5 and 36%. A complete genome sequencing of the most common Serbian isolate of PVY (3D), followed by molecular characterization and phylogenetic analysis has been performed to show what group it belongs to. Our isolate's complete genome sequence (KJ946936) showed that the Serbian PVY isolate (3D) is 99.7% identical at nt level, with other tuber necrosis strain group (PVYNTN) from Europe. Phylogenetic analysis revealed three consistent lineages of isolates, showing that our isolate was clustered with the isolates from Europe and North America in the PVYN lineage which induces potato tuber necrotic ringspot disease (PTNRD). AB  - Anise (Pimpinella anzsum L.) is an important medicinal spice plant that belongs to the family Apiaceae. Anise seeds are rich in essential oils and this is a reason why anise production in Serbia has increased over the last decade. During a routine health inspection on anise seeds collected from three localities in the province of Vojvodina (Mogorin, Veliki Radinci and Ostojkevo) during 2012 and 2013, it was found out that Fusarium spp. were a commonly observed fungi. The presence of Fusarium fungi on the seed samples ranged from 3.75-13.75%. The aim of this study was to isolate and identify the strains of Fusarium species present on anise seed samples as it is necessary that commercially used anise seeds are completely free of Fusarium. Based on morphological, microscopic characteristics and a molecular identification by sequencing of TEF gene, the presence of the following species was confirmed on the anise seeds: F. trzeinctum, F. proliferatum, F. equiseti, F. oxysporum, F. sperotrzchozcles, F. incarnatum and F. verttallzoteles. According to our knowledge and research, this is the first report of F. trianctum and F. sporotrichoi des as pathogens on anise seeds in the world. All seven isolates of Fusarium species are pathogenic to the anise seedlings, while the most virulent species were F. oxysporum, F. tricznetum and F. incarnation.

AB  - Primary and secondary infections with PVYNTN were investigated on forty plants of the potato cv. Romano inoculated in a greenhouse in Serbia in 2012 and 2013. PVY isolates were collected from the potato growing region of Čačak and identified by ELISA and RT-PCR methods. The sequence of the Serbian isolate 3D (Acc. No. KJ946936) showed 100% match with seven PVY isolates deposited in GenBank and described as NTN. A significant difference was detected between PVYNTN symptoms exibited on leaves of the cv. Romano under primary and secondary infections. The findings are significant because they are based on symptoms observed, so that it is clear that there are two distinct types of infection: primary and secondary. Symptoms of primary and secondary infection were the same on potato tubers and had the form of necrotic rings.
AB  - Upoređivani su simptomi primarnih i sekundarnih zaraza na četrdeset biljaka sorte Romano inokulisanih izolatom PVYNTN u uslovima staklare, tokom 2012 i 2013 godine. Izolat PVY je kolekcionisan u lokalitetu Čačak, identifikovan je ELISA testom i okarakterisan RT-PCR metodom kao PVYNTN soj. Sekvenca CP gena odabranog izolata 3D (Acc. No. KJ946936) ispoljila je 100% nukleotidnu identičnost sa sedam sekvenci PVY izolata deponovanih u GenBank opisanih kao NTN soj. Ispoljeni simptomi na biljnoj masi primarno i sekundarno zaraženih biljaka opisanim sojem su značajno različiti. Na osnovu ovih razlika može se zaključivati da li su se biljke zarazile u tekućoj godini ili su sađene već zaražene krtole. Prstenasta nekroza je zajednički simptom na krtolama primarno i sekundarno zaraženih biljaka.

This disease cause a several disease-causing agents, and the most important is Didymella applanata, causal agent of raspberry spur blight. With regard to the biology of the pathogen fungicide use postharvest is not efficient enough because pat­hogen can infect raspberry far earlier. Therefore, the aim of this study is testing the efficacy of newer organic fungicides applied before and after the harvest raspberries and mutual comparison thereof on biological efficacy. The results show that the tested fungicides ex­hibited very high efficacy against D. apllanata on the raspberry leaves and the canes (from 83.0 - 96.5%).
AB  - Prouzrokovači 'kompleksa sušenja' maline predstavljaju najveći problem u njenoj proizvodnji smanjujući prinose i životni vek zasada. Ovaj kompleks sačinjen je od više prouzrokovača bolesti, a najznačajniji u tom kompleksu je Didymella applanata, prouzrokovač kestenaste pegavosti maline. S obzirom na biologiju patogena tretiranje biljaka tek nakon berbe nije dovoljno efikasno jer patogen u povoljnim uslovima može da zarazi malinu daleko ranije. Cilj rada je ispitivanje efikasnosti novijih organskih fungicida primenjenih pre i posle berbe maline kao i međusobno poređenje njihove biološke efikasnosti. Dobijeni rezultati pokazuju da su ispitivani fungicidi ispoljili veoma visoku efikasnost u suzbijanju D. applanata kako na listu tako i na izdancima maline (od 83.0 - 96.5%).

The effectiveness of potassium phosphite and captan mixture in controlling Venturia inaequalis in apple orchards

AB  - Venturia inaequalis is a common apple disease in Serbia and its intensity depends on weather conditions. The most successful method of combating apple scab is to use a lower dose of an active ingredient and increase its effectiveness by adding substances that do not cause environmental damage. The effectiveness of a fungicide mixture of captan and potassium phosphite against V. inaequalis was investigated in apple orchards in 2008 and 2009. A relevant EPPO standard method was used in all trials. Captan 50 WP was used as a standard fungicide. In order to determine the exact conditions marking the onset of apple infection, key meteorological conditions were monitored in the orchards using an automated iMetos weather station and a Lufft electronic instrument. The results showed high efficacy of the tested fungicides which remained similar in both years of investigation. The intensity of leaf infection after treatment with the mixture of captan and potassium phosphite was 0.7-2.4%, while fruit infection was 0.5-3.0%. The efficacy of this mixture ranged from 96.3-97.9% on leaves and 87.4-98.7% on fruits. The standard fungicide Captan 50 WP showed an efficacy ranging from 95.3-96.7% on leaves and from 87.8-99.3% on fruits. No statistically significant differences were found between the fungicide tested and the standard fungicide.
AB  - Venturia inaequalis je široko rasprostranjena bolest jabuke u Srbiji, a njen intenzitet zavisi od vremenskih uslova. Najuspešniji metod borbe protiv čađave krastavosti jabuke je korišćenje manjih doza aktivnih materija uz istovremeno povećavanje efikasnosti dodavanjem supstanci koje ne prouzrokuju štetu u životnoj sredini. Tokom 2008 i 2009 godine, proučavana je efikasnost fungicidne mešavine kaptana i kalijum fosfita protiv V. inaequalis u jabučnjacima. U svim ogledima je korišćen standardni EPPO metod. Captan 50 WP je korišćen kao standardni fungicid. Kako bi se tačno odredili uslovi za početak zaraze na jabuci, praćeni su osnovni meteorološki parametri u voćnjacima pomoću iMetos stanice i elektronskog uređaja Lufft. Rezultati su pokazali visoku efikasnost testiranih fungicida koja je bila slična u obe godine istraživanja. Intenzitet zaraze lista nakon tretmana mešavinom kaptana i kalijum fosfita bio je 0.7-2.4%, dok je zaraženost plodova bila 0.5-3.0%. Efikasnost mešavine bila je 96.3-97.9% na listovima, a 87.4-98.7% na plodovima. Standardni fungicid Captan 50 WP pokazao je efikasnost u opsegu 95.3-96.7% na listovima i 87.8-99.3% na plodovima. Nisu konstatovane statistički značajne razlike između ispitivanog i standardnog fungicida.

The results showed high efficacy of the tested fungicides which remained similar in both years of investigation. The intensity of leaf infection after treatment with the mixture of captan and potassium phosphite was 0.7-2.4%, while fruit infection was 0.5-3.0%. The efficacy of this mixture ranged from 96.3-97.9% on leaves and 87.4-98.7% on fruits. The standard fungicide Captan 50 WP showed an efficacy ranging from 95.3-96.7% on leaves and from 87.8-99.3% on fruits. No statistically significant differences were found between the fungicide tested and the standard fungicide., Venturia inaequalis je široko rasprostranjena bolest jabuke u Srbiji, a njen intenzitet zavisi od vremenskih uslova. Najuspešniji metod borbe protiv čađave krastavosti jabuke je korišćenje manjih doza aktivnih materija uz istovremeno povećavanje efikasnosti dodavanjem supstanci koje ne prouzrokuju štetu u životnoj sredini. Tokom 2008 i 2009 godine, proučavana je efikasnost fungicidne mešavine kaptana i kalijum fosfita protiv V. inaequalis u jabučnjacima. U svim ogledima je korišćen standardni EPPO metod. Captan 50 WP je korišćen kao standardni fungicid. Kako bi se tačno odredili uslovi za početak zaraze na jabuci, praćeni su osnovni meteorološki parametri u voćnjacima pomoću iMetos stanice i elektronskog uređaja Lufft. Rezultati su pokazali visoku efikasnost testiranih fungicida koja je bila slična u obe godine istraživanja. Intenzitet zaraze lista nakon tretmana mešavinom kaptana i kalijum fosfita bio je 0.7-2.4%, dok je zaraženost plodova bila 0.5-3.0%. Efikasnost mešavine bila je 96.3-97.9% na listovima, a 87.4-98.7% na plodovima. Standardni fungicid Captan 50 WP pokazao je efikasnost u opsegu 95.3-96.7% na listovima i 87.8-99.3% na plodovima. Nisu konstatovane statistički značajne razlike između ispitivanog i standardnog fungicida.",

AB  - Copper citrate is a complex compound of copper, which is characterized by a higher degree of dissociation in relation to the other copper compounds that are presently in the use and can be used in lower concentrations for the control of plant pathogens. Apple scab (Venturia inaequalis) is on of the economically most important diseases of apples in all regions where this fruit species grown. A successful apple production is not possible without intensive chemical control. The use of copperbased products to protect apples, positioned at the start of the growing season. The earliest apple infections are the most destructive, and the effective protection of the apples in this period is very important. The aim of this study was to evaluate in vitro the influence of copper citrate at different concentrations applied to the growth of V. inaequalis colonies. The results show that copper citrate in significantly lower concentrations of application 0.2 and 0.1 % higher than the standard product, effectively inhibiting the growth of V. inaequali mycelium (colony), which is the result of significantly higher amounts of copper ions, that is released from compound.
AB  - Bakar-citrat je kompleksno jedinjenje bakra koje se odlikuje višim stepenom disocijacije u odnosu na druga bakarna jedinjenja koja su trenutno u primeni pa se može koristiti u nižim koncentracijama za suzbijanje biljnih patogena. Čađava pegavost lista i krastavost plodova (Venturia inaequalis) je jedna od ekonomski najštetnijih bolesti jabuke u svim regionima gde se ova voćna vrsta gaji. Uspešna proizvodnja jabuke nije moguća bez izvođenja intenzivnih hemijskih mera zaštite. Primena preparata na bazi bakra u zaštiti jabuke, pozicionirana je na sam početak vegetacije. Najranije zaraze jabuke su i najdestruktivnije, pa je efikasna zaštita jabuke u ovom periodu veoma važna. Cilj ovog rada je da se u in vitro uslovima ispita uticaj bakar-citrata u različitim koncentracijama primene na porast kolonija Venturia inaequalis. Dobijeni rezultati pokazuju da bakar citrat u znatno nižim koncentracijama primene 0,2 i 0,1% nego sandardni preparat, efikasno blokira porast micelije (kolonija) fitopatogene gljive V. inaequalis, što je rezultat znatno veće količine bakarnih jona koje ovo jedinjenje oslobađa.

AB  - Due to unfavorable terrain configuration on the great of deposols of the 'Kolubara' open cast mine, recultivation under forest cultures has been performed. Tests of basic agrochemical properties as well as quantitative abundance of some groups of microorganisms in deposols of different oldness and under different forest cultures have been done. The tested deposols have shown favorable pH, low content of humus and total nitrogen, readily available to phosphorus and potassium. The biogenity shown is satisfactory but the total microflora, fungi and Clostridium abundance is lower compared to the control soil while the other physiological groups of microorganisms are considerably more abundant.
AB  - Zbog nepovoljne konfiguracije terena, na većini trajno odloženih zemljišta (deposola) rudarskog basena 'Kolubara' rekultivacija je izvedena pošumljavanjem. Izvršeno je ispitivanje osnovnih agrohemijskih osobina, kao i kvantitativne zastupljenosti pojedinih grupa mikroorganizama u deposolima različite starosti i pod različitim šumskim kulturama. Ispitivani deposoli su pokazali povoljniji pH od kontrolnog zemljišta, niži sadržaj humusa, ukupnog N i lako pristupačnog K2O, a viši sadržaj lako pristupačnog P2O5. Opšta biogenost je bila zadovoljavajuća, s tim što je zastupljenost ukupne mikroflore, gljivica i bakterija iz roda Clostridium bila manja nego kod kontrolnog zemljišta, dok je zastupljenost ostalih fizioloških grupa mikroorganizama bila znatno veća.

AB  - Graft transmissibility of the plum sectorial resistance to Plum pox virus (PPV) was investigated. Sectorial resistance is the resistance which is manifested in a sector or portion of infected plant, where virus is not present. Sectorial resistance to PPV is exhibited in the crown or in primary shoots of the plum tree. Double budding method was applied. One bud was carrying sectorial resistance to PPV, while the other one was infected with the virus. Infection on shoots was evaluated by symptoms appearance and serological test for PPV presence.Among three plum cultivars the best results were obtained with cv. "Crvena ranka". Sectorial resistance to PPV has been successfully transmitted by buds of "Crvena ranka". Plum cv. "Č ačanska rodna" failed to transmit sectorial resistance. Plum cv. "Stanley" was between those two above-mentioned plum cultivars.

Etiology of diseases of some medicinal plants in plantation in Serbia

AB  - Marshmallow, coneflower, St. John's wort and sage, which grown in plantation, were the hosts of numerous fungi species that cause a variety of symptoms. A total of 34 species from 22 genera were determined. Some species pathogen on only one host (Puccinia malvacearum on marshmallow, Diaporthe eres and Seimatosporium hypericum on St. John's wort), while others were present at all examined hosts (species of the genus Fusarium are present in all examined hosts). The most abundant species in the seed was Alternaria alternate. The percentage of contaminated seeds ranged from 5% to 69%. Fusaium species were isolated from 2.5% to 29% of seeds, depending on the host and year. From the root of the studied medicinal plants seven species of Fusarium (F.graminearum, F. oxysporum, F.proliferatum, F.semitectum, F.solani, F.subglutinans and F.verticilioides) and one of the genus Sclerotinia (S.sclerotiorum) were isolated. Thirteen fungi species belonging to 10 genera were isolated from stem, leaves and flower of medicinal plants.
AB  - Plantažno gajeni beli slez, ehinacee, kantarion i žalfija su domaćini brojnih vrsta gljiva koje prouzrokuju različite simptome. Ukupno je determinisano 34 vrsta iz 22 roda. Neke vrste parazitiraju samo jednog domaćina (Puccinia malvacearum na belom slezu, Diaporthe eres i Seimatosporium hypericum na kantarionu), dok druge ispoljavaju izrazitu polifagnost (vrste iz roda Fusarium prisutne su na svim ispitivanim domaćinima). Najzastupljenija vrsta na semenu bila je Alternaria alternata, koja je uvek prisutna na semenu u visokom procentu, a pojednih godina i preko 60 %. Na korenu ispitivanih lekovitih biljaka izolovano je sedam vrsta gljiva iz roda Fusarium (F.graminearum, F.oxysporum, F.proliferatum, F.semitectum, F.solani, F.subglutinans i F verticilioides) i jedna iz roda Sclerotinia (S.sclerotiorum). abstract = "Marshmallow, coneflower, St. John's wort and sage, which grown in plantation, were the hosts of numerous fungi species that cause a variety of symptoms. A total of 34 species from 22 genera were determined. Some species pathogen on only one host (Puccinia malvacearum on marshmallow, Diaporthe eres and Seimatosporium hypericum on St. John's wort), while others were present at all examined hosts (species of the genus Fusarium are present in all examined hosts). The most abundant species in the seed was Alternaria alternate. The percentage of contaminated seeds ranged from 5% to 69%. Fusaium species were isolated from 2.5% to 29% of seeds, depending on the host and year. From the root of the studied medicinal plants seven species of Fusarium (F.graminearum, F. oxysporum, F.proliferatum, F.semitectum, F.solani, F.subglutinans and F.verticilioides) and one of the genus Sclerotinia (S.sclerotiorum) were isolated. Thirteen fungi species belonging to 10 genera were isolated from stem, leaves and flower of medicinal plants., Plantažno gajeni beli slez, ehinacee, kantarion i žalfija su domaćini brojnih vrsta gljiva koje prouzrokuju različite simptome. Ukupno je determinisano 34 vrsta iz 22 roda. Neke vrste parazitiraju samo jednog domaćina (Puccinia malvacearum na belom slezu, Diaporthe eres i Seimatosporium hypericum na kantarionu), dok druge ispoljavaju izrazitu polifagnost (vrste iz roda Fusarium prisutne su na svim ispitivanim domaćinima). Najzastupljenija vrsta na semenu bila je Alternaria alternata, koja je uvek prisutna na semenu u visokom procentu, a pojednih godina i preko 60 %. Godišnji indeks porasta zaraze kretao se od 2 do 6, a smanjenje prinosa zelene mase između 30-50%. Primenom RFLP i analize sekvenci 16S rDNK, identifikovano je prisustvo fitoplazmi u uzorcima lucerke koje pripadaju 16SrIII-B i 16SrXII-A grupama. Fitoplazma Stolbur tipa bila je dominantna i dokazana je u 80% uzoraka simptomatičnih biljaka.

Identification of Colletotrichum acutatum from nectarine fruit

AB  - Isolates of Colletotrichum spp. obtained from nectarine fruits with typical anthracnose symptoms in 2010 were identified using morphological and molecular methods. Pathogenicity test was conducted on symptomless, detached nectarine fruits. All tested isolates caused anthracnose lesions on nectarine fruits after 7 days of incubation. On PDA medium nectarine isolates forming dark green to dark gray mycelia. Growth rates of all isolates and reference strain of C.acutatum were lower at 25°C compared with reference strain of C.gloeosporioides. The conidia were hyaline, aseptate, and fusiform. Appressoria were dark brown, smooth, simple, and clavate to ovate. Using the primer set CaInt2/ITS4, the 490 bp DNA fragment was amplified from all nectarine isolates and reference strain of C. acutatum - CBS 294.67. Based on these results, the causal agent of anthracnose on nectarine fruits in Serbia was identified as C.acutatum.
AB  - Izolati Colletotrichum spp. dobijeni 2010. godine, sa antraknoznih plodova nektarine identifikovani su pomoću morfoloških i molekularnih metoda. Test patogenosti je obavljen sa reprezentativnim izolatima, na odabranim, zdravim plodovima. Svi ispitivani izolati prouzrokuju antraknozne lezije na plodu nektarine, 7 dana nakon inokulacije. Na PDA podlozi izolati formiraju tamno zelenu do tamno sivu miceliju. Stopa rasta izolata sa nektarine i referentnog soja C.acutatum je bila niža u odnosu na referentni soj C.gloeosporioides. Konidije su hialinske, neseptirane i fusiformne. Apresorije su tamno braon boje, glatke, jednostavne, okruglastog ili oblika izdužene palice. Korišćenjem para prajmera CaInt2/ITS4 iz genoma DNA izolata sa ploda nektarine i referentnog soja C. acutatum - CBS 294.67, amplifikovan je fragment veličine 490 bp. AB  - In concordance with phytosanitary regulations and the Program of continuous monitoring of quarantine and economically harmful nematodes that are intensively conducted every year, the presence of these organisms is determined in the territory of the Republic of Serbia and in the samples of plant material. Globodera pallida and G.rostochiensis are the most important parasites of potato in our country and their presence is monitored since 2005 and 2000 respectively. Morphology, i.e. examination of morphological characters of different populations of potato cyst nematodes determines their mutual similarities (differences) within species, between species and the similarity with the type populations. This paper presents the morphological descriptions of cysts and second stage larvae of two populations of G.rostochiensis found in localities Rujevac and Tabanovici in 2012.
AB  - U skladu sa Programom stalnog praćenja karantinskih i ekonomski štetnih nematoda koji se intenzivno sprovodi svake godine kao i fitosanitarnom regulativom, utvrđuje se prisustvo ovih organizama na teritoriji Republike Srbije i u uzorcima biljnog materijala. Globodera pallida i G. rostochiensis su najznačajniji paraziti krompira kod nas a njihovo prisustvo se prati od 2005. odnosno 2000. god. Morfologija odnosno ispitivanje morfoloških karakteristika različitih populacija cistolikih nematoda krompira daje podatke o njihovoj međusobnoj sličnosti (razlikama) unutar vrsta, između vrsta i sličnost sa tipskim populacijama. U radu su dati morfološki opisi cisti i larvi drugog stepena dve populacije G. rostochiensis pronadjene na lokalitetima Rujevac i Tabanovići u 2012. godini.

AB  - Determination of breeding and morphological characteristics of C. beticola isolates was performed on PDA, MEA i CDA mediums. Determination of morphological characteristics were evaluated by monitoring mycelial growth, texture, color, edge and color of beneath of mycelium. Isolates growth on different temperatures was monitored on PDA medium on 10°C, 15°C, 20°C, 25°C, 30°C, 35°C and 40°C. There was a significant increase in variability of the isolates on PDA, MEA and CDA mediums, but there is a variability in the growth of different isolates of the same medium too. Isolates expressed the variation in color and the width of the edge zone during growth on different substrates. The mycelium texture was cottony on all surfaces, while the beneath was brown to dark-olive colour. At temperatures of 10°C and 40°C isolates of C. beticola did not develop mycelium, and the highest increase was recorded at 25°C.
AB  - Utvrđivanje morfoloških i odgajivačkih karakteristika izolata C. beticola izvedeno je na KDA, MEA i CDA podlozi. Utvrđivanje morfoloških karakteristika obuhvatilo je praćenje porasta micelije, teksture, boje, ivične zone i boje naličja micelije. Porast izolata na različitim temperaturama praćen je na KDA podlozi na temperaturama 10°C, 15°C, 20°C, 25°C, 30°C, 35°C i 40°C. Utvrđena je značajna varijabilnost porasta izolata na KDA, MEA i CDA podlozi, a variabilnost postoji i u porastu različitih izolata na istoj podlozi. Izolati su ispoljili i variranje u boji i širini ivične zone tokom porasta na različitim podlogama. Tekstura micelije bila je pamučasta na svim podlogama, dok je boja sa naličja bila mrka do tamno-maslinasta. Na temperaturama 10°C i 40°C izolati C. beticola nisu razvijali miceliju, a najintenzivniji porast je ostvaren na 25°C.

Porast izolata na različitim temperaturama praćen je na KDA podlozi na temperaturama 10°C, 15°C, 20°C, 25°C, 30°C, 35°C i 40°C. Utvrđena je značajna varijabilnost porasta izolata na KDA, MEA i CDA podlozi, a variabilnost postoji i u porastu različitih izolata na istoj podlozi. Izolati su ispoljili i variranje u boji i širini ivične zone tokom porasta na različitim podlogama. Tekstura micelije bila je pamučasta na svim podlogama, dok je boja sa naličja bila mrka do tamno-maslinasta. Na temperaturama 10°C i 40°C izolati C. beticola nisu razvijali miceliju, a najintenzivniji porast je ostvaren na 25°C.",

AB  - Plant diseases caused by phytopathogenic fungi are one of the major problems in orchard plants in Serbia. To avoid the use of chemical agents in high percent, biological control is becoming promising method in plant disease management. It has been showed that apple is host of Venturia inaequalis, a very aggressive phytopathogenic fungus known for production scab. In search for ecological ways for prevention and reduction of disease symptoms, biological control with indigenous fluorescent Pseudomonas isolates has been offered as a possible solution. The aim of this study was to examine antifungal activity of different indigenous fluorescent Pseudomonas isolates (Q16, B25 and PS2) against phytopathogenic fungus V. inaequalis which had infected apple. Indigenous Pseudomonas isolates showed in vitro inhibition of fungal growth 13.85% to 54.36 % on WA and 11.37 % to 58.33% on PDA. These isolates should be further analyzed in order to classify them as promising group of biocontrol agents against diseases of apple scab caused by V. inaequalis.
AB  - Biljne bolesti prouzrokovane fitopatogenim gljivicama su jedan od glavnih problema u voćnjacima u Srbiji. Biološka kontrola patogena sve više se koristi u upravljanju biljnim bolestima kako bi se izbeglo korišćenje hemijskih agenasa u velikoj meri. Jabuka kao veoma značajna voćarska kultura je domaćin veoma agresivne fitopatogene gljive Venuria inaequalis, prouzrokovača čađave krastavosti ploda. U potrazi za ekološkim načinima za sprečavanje pojave i smanjenje simptoma bolesti, biološka kontrola autohtonim izolatima iz roda Pseudomonas je potencijalno moguće rešenje. Cilj ovog istraživanja bio je da se ispita antifungalna aktivnost različitih autohtonih izolata Pseudomonas (P16, B25 i PS2) protiv fitopatogene gljive V.inaequalis koja zaražava jabuku. Autohtoni Pseudomonas izolati su u in vitro uslovima pokazali inhibiciju rasta kolonija gljivice od 13.85% do 54.36% na WA podlozi i od 11.37% do 58.33% na PDA. Ove izolate treba dalje analizirati kao grupu bioloških agenasa koja može imati veliki potencijal u suzbijanju V.inaequalis, prouzrokovača čađave krastavosti jabuke.

AB  - Plants of alfalfa (Medicago sativa) exhibiting general stunting, proliferation and phyllody associated with leaf yellowing and reddening were observed in three localities of Central Serbia. Phytoplasma strains belonging to 16SrIII-B and 16SrXII-A groups were detected and identified by RFLP and sequence analysis of 16S rDNA. Stolbur phytoplasma tuf gene RFLP analysis showed the presence of the TufAY-b-type phytoplasma subgroup in 80% of symptomatic samples. This is the first report of 16SrIII-B and 16SrXII-A phytoplasma groups affecting alfalfa in Serbia.

AB  - During the late summer of 2007, a severe phytoplasma-like disease was observed for the first time in blackberry plants (Rubus fruticosus), commercial cv. Čačanska beztrna. Redness and downward rolling of leaves were symptoms observed in three localities in Central Serbia. The presence of Stolbur phytoplasma, belonging to the taxonomic subgroup 16SrXII-A, in diseased samples was confirmed by the PCR - RFLP analysis of 16S rDNA genes and elongation factor Tu (tuf) gene. A sequence analysis of the tuf gene confirmed homology with phytoplasmas stolbur tuf-type II detected previously in Italian grapevines and red clovers in the Czech Republic. This is the first report of Stolbur phytoplasma 16SrXII-A group tuf-type II on blackberries in Serbia.
AB  - Simptomi nalik fitoplazmi prvi put su uočeni tokom leta 2007 g. na kupini (Rubus fruticosus) sorte Čačanska beztrna. Crvenilo i uvrtanje listova kao najčešći simptomi primećeni su na 3 lokaliteta u centralnoj Srbiji. Prisustvo Stolbur fitoplazme, koja pripada 16SrXII-A taksonomskoj podgrupi, utvrđeno je PCR - RFLP analizom 16S rDNK gena i gena za elongacioni faktor Tu (tuf) u uzorcima sa izraženim simptomima. Given the large number of these units there was a need to unite together with one author registers and keywords to help users search. The units are arranged in chronological order. Stacking units is done in accordance to bibliographic rules, meaning that the works of three authors are linked to the first author, and where there are more than three authors, the unit takes on the title. On this occasion, given the bibliography of papers published in the period 2001-2010. year, the number of 235-274, with 112 bibliographical.
AB  - Časopis 'Zaštita bilja', čiji je vlasnik i osnivač Institut za zaštitu bilja i životnu sredinu, osnovan je 1950. godine. Do sada su odštampana 274 broja. Časopis se bavi objavljivanjem naučnih radova o bolestima i štetočinama u poljoprivredi i šumarstvu, rezultatima ispitivanja pesticida i njihovim načinima delovanja. Radovi se štampaju na srpskom ili engleskom jeziku. Časopis se distribuira pretplatom ili razmenom u 43 zemlje sveta. Časopis 'Zaštita bilja' izlazi preko 60 godina tokom kojih je objavljeno 2394 bibliografske jedinice. AB  - A special resistance to sharka (Plum Pox Virus, PPV) has been discovered and studied in plum trees. That resistance is characterized by the absence of sharka symptoms and PPV, too, in some of primary branches of plum trees otherwise diseased by sharka. Symptoms appearance was observed and PPV presence was checked serologically by DAS-ELISA test as well as by grafting transmission. That resistance mainly appears in a portion of the tree crown, i.e. on some primary branches of young plum trees and it is diminishing in following years. That distinct resistance to sharka (PPV) was found in trees of Čačanska rodna, Stanley and Crvena ranka cvs. In selected plum trees in the parts with such resistance no sharka symptoms appeared nor PPV was proved. The absence of sharka symptoms as well as PPV in a portion of a plum tree otherwise diseased by sharka is considered as a sign of resistance. For such resistance the name 'Sectorial Resistance' is proposed. More and detail research on sectorial resistance of plum to PPV are in progress.
AB  - Otkrivena je i istraživana jedna posebna otpornost šljive na virus šarke šljive (VŠŠ, Plum pox virus - PPV) za koju se predlaže naziv 'sektorijalna otpornost'. Ta otpornost se ispoljava u delu krune, odnosno na pojedinim skeletnim granama obolelih stabala. U tom delu krune, odnosno na skeletnim granama koje pokazuju sektorijalnu otpornost nema simptoma oboljenja, niti je prisutan VŠŠ. Odsustvo VŠŠ proveravano je serološki DAS-ELISA testom i kalemljenjem. Sektorijalna otpornost je nađena kod stabala Čačanska rodna, Stenley-a i Crvene ranke. Sektorijalna otpornost šljive na VŠŠ je u negativnoj korelaciji sa starošću stabala na kojima se javlja. Broj skeletnih grana sa sektorijalnom otpornošću kod obolelih stabala se smanjuje iz godine u godinu, a povećava broj skeletnih grana sa simptomima oboljenja. U toku su dalja i svestranija proučavanja te novootkrivene otpornosti šljive prema VŠŠ.

AB  - Grapevine cv. Plovdina is a traditional local variety that is very sensitive to flavescence doree (FD) disease. In this paper, we present the results of top-grafting FD-infected cv. Plovdina plants with scions originating from healthy mother plants of the same cultivar using the "green grafting" method. The first grapevine yellows (GY) symptoms, consisting of yellowing of the Plovdina leaves, have been observed on scions 20 days after grafting. Our results show that GY symptoms were due to phytoplasma and not the viral infection, since grapevine leafroll associated viruses-1, 2 and 3 (GLRaV-1, 2, 3) could not be detected in the scions. Twenty days after-grafting, phytoplasma FD was detected in all grafted scions. Our results indicate that grapevine cv. Plovdina is very suitable as an indicator plant for Flavescence doree disease.

Complete nucleotide sequence and genome organisation of grapevine Bulgarian latent virus

AB  - The complete genome sequence of grapevine Bulgarian latent virus (GBLV) has been determined. RNA-1 (7,452 nt in length) contains a single ORF of 6,285 nt, encoding a polyprotein with conserved motifs characteristic of the viral protease cofactor (Prot-cofact), the NTP-binding protein (NTP), the cysteine-like protease (Cyst-Prot) and the RNA-dependent RNA polymerase (RdRp) of members of the order Picornavirales and show high aa sequence identity with blackcurrant reversion virus (BRV, 64%). RNA-2 (5,821 nt) contains a single ORF of 4,500 nt, encoding a polyprotein in which the conserved motifs of the movement protein (MP) and coat protein (CP) have been identified. The GBLV CP aa sequence shows highest homology with that of blueberry leaf mottle virus (BLMoV, 68%). Both RNAs have a poly(A) tail and a NCR at the 3' and 5' termini, respectively. The results of this study confirm the classification of GBLV as a member of a distinct species in subgroup C of the genus Nepovirus.
