AB - Sugar beet (Beta vulgaris L.) has been grown for centuries as one of the most important agricultural plants worldwide for the purpose of white crystal sugar production. A large number of pathogens are present on the sugar beet, causing a large number of various symptoms and damaging production in a big scale. Plant diseases and pests are present on leaves and roots, causing damage to the above- and below-ground parts. Rhizoctonia solani Kuhn. is one such disease that attacks the sugar beet and is widespread throughout the world. R. solani is a soil-borne pathogen that survives in soil, where its life cycle is maintained for a long time period. It grows in the temperate and continental climate zone, making it a very important pathogen with respect to this crop due to its favourable condition. It overwinters in the soil for many years in the form of sclerotia or mycelium. The first symptoms appear on the upper part of sugar beet plants; usually symptomatic plants appear in bordered zones, often present in certain parts of the field with a clearly bordered belt. The infected plants have lost their turgor, and most of the leaf rosette gets laid on the ground. In the cross section, the central part of the root is initially healthy, while the disease is primarily observed on the rim of the root. The most important and only fully effective approach in controlling R. solani is the application of fungicides from SDHI and QoI groups at the appropriate time of application.

AB - Sugar beet is listed within the top ten most important crops in the world. The paleobotanic data suggest the sea beet was grown in the ancient times, while the beets with swollen roots were cultivated in the Middle Ages in Europe. Sugar beet cyst nematode Heterodera schachtii is an invasive organism causing high economic loss to sugar beet worldwide. The fundamental steps in control of harmful organisms in plant protection and food safety are grounded on rapid detection of the causative agent and its proper identification. The prompt reaction before obvious symptoms occur can prevent devastating consequences. To confirm the identity of an invasive organism, the process demands the combination of identification techniques, such as morphology and molecular characterization. The phylogeography of available H. schachtii populations, based on matching historical data with phylogenetic analyses of the ITS rRNA region pinpointed a possible place of origin of the European H. schachtii populations. A next step is the determination of natural antagonists of nematodes. Due to the long persistence of the parasite in soil, cysts harbor a large number of bacteria and fungi, the presence of which can lead to cyst death and population decline. Bacteria, fungi and other antagonists are an inevitable part of the soil ecosystem and are also part of those mechanisms in nature that limit the excessive number of invasive organisms and return ecological system to its stable equilibrium, acceptable for the existence of humans as well as micro and macrobiota on Earth.

Antagonistička aktivnost 6 autohtonih bakterijskih izolata (Bacillus subtilis Z3, Bacillus mojavensis 5B2, Bacillus mojavensis R6, Bacillus firmus S5, Bacillus proteolyticus 7E i Bacillus thuringiensis 109 /18 1.) ispitivana je metodom dvojne kultivacije.

U ovom istraživanju, korišćeno je 12 bakterijskih sojeva roda Bacillus (Bacillus velezensis 31/18 RK 1-1, Bacillus amyloliquefaciens 31/18 RK 1-2, Bacillus licheniformis 31/18 RS-9, Bacillus mojavensis 31/18 5b2, Bacillus aerius 92/17 7, Bacillus marisflavi 6E, Bacillus pumilus W27-2, Bacillus macroides W29-5, Bacillus megaterium S1, Bacillus thuringiensis BCE, Bacillus amyloliquefaciens B002, Bacillus proteolyticus 7E).

AB - Cercospora leaf spot (CLS) caused by Cercospora beticola occurs annually in Serbia causing severe yield losses of sugar beet, which requires intensive use of fungicides. In recent years we have observed unsatisfactory control of CLS originating from northwestern Serbia. Frequency of C. beticola populations resistant to Quinone outside inhibitors (QoI) was 81% (51/63 isolates), 98% (62/63) to sterol-demethylation inbibitors (DMI) and 54% (34/63) to methyl-2-benzimidazole carbamate fungicides (MBC). The genetic basis underlying the resistance was tested by characterizing the cob, CYP51 and -tubulin genes, associated with resistance to QoI, DMI and MBC fungicides, respectively. Isolates that were resistant to QoI fungicides had the G143A mutation in the cob gene. Characterization of the CYP51 gene revealed seven diverse haplotypes; however, no correlation with sensitivity or resistance to DMI fungicides could be identified. Resistance to MBC fungicides was associated with the presence of the E198A mutation in the -tubulin gene of all resistant isolates. From a total of 63 isolates originating from sugar beet fields of northwestern Serbia, 62 isolates showed resistance to multiple modes of action. Three multi-resistant phenotypes were identified: MR1 (N = 29) - resistant to QoI and DMI fungicides (QoI-R and DMI-R) but sensitive to MBC fungicides (MBC-S); MR2 (N = 11, QoI-S, DMI-R and MBC-R); and MR3 (N = 22), resistant to all three groups of fungicides (QoI-R, DMI-R and MBC-R). This is the first report of C. beticola resistance to QoI fungicides in Serbia. This study revealed development of multi-resistance of C. beticola isolates to MBC, DMI and QoI fungicides, which represents the first record of this phenomenon in C. beticola populations.

AB - In May 2014, during a cool, humid spring period, bacterial spot symptoms were observed for the first time on pea (Pisum sativum L.) in a home garden in the Bačka region of Vojvodina Province in Serbia. Commercial pea is grown on 11,533 ha, as well as in small vegetable farms in Serbia, with average yields of 4 t/ha. Observed spots on leaves were necrotic and progressed inward from the margins. Disease incidence approached 10 to 30%. Bacteria consistently isolated from margins of leaf lesions macerated in sterile phosphate buffered saline (PBS, pH 7.4) formed round, shiny, white, 2- to 3-mm in diameter colonies on nutrient agar with 5% (w/v) sucrose (NAS) and a green fluorescent pigment on King’s medium B (KB). A collection of 20 strains selected for further study was strictly aerobic, gram-negative; positive for levan and tobacco hypersensitive response and negative for oxidase, arginine dihydrolase, and potato soft rot (LOPAT group Ia). Strains were characterized by repetitive (rep)-PCR, sequence typing, and pathogenicity. To determine genetic heterogenicity among the tested strains, rep-PCR genetic fingerprinting was conducted using the REP, ERIC, and BOX primers (Louws et al. 1994). Profile analyses yielded identical banding patterns for all tested strains. To identify three representative strains by sequence typing, the housekeeping genes gyrB and gltA were amplified (Ferrante and Scortichini 2010). Sequences were deposited in GenBank under accession numbers KP188575 to 77 for gyrB and KP188578 to 80 for gltA. BLAST analysis showed that the regions were 100% homologous for the partial gltA gene of the reference strain P. syringae pv. syringae CFBP 4702 (KF937504) and 99% homologous for the partial gyrB gene of the reference strain P. syringae pv. syringae PD 2021 (KJ158894). Pathogenicity of the three tested strains was tested by using lesion tests and stem inoculation (Mazarei and Kerr 1990). For all tests, a reference strain P. syringae pv. syringae GSPB 1142 and PBS served as a positive and negative controls, respectively. In lesion tests, young bean pods and pale yellowish-green lemon fruits were inoculated by placing drops of bacterial suspension (with 108 CFU/ml) on the surface and pricking lightly through the drops with a sterile needle. Fruits and pods were incubated at 25°C for 7 days. Tested strains produced small reddish-brown depressed lesions on bean pods and dark brown depressed lesions on lemon fruits. For stem inoculation, pea cv. Mali provansalac plantlets were grown from seed for 10 to 14 days. Bacteria grown on KB for 24 h were scraped from the surface with a sterile needle and stabbed into the main stem at the youngest two nodes. Inoculation resulted in extensive necrosis and collapse with stunting on the plants 10 days after inoculation. Control strain GSPB 1142 developed similar symptoms to those of the strains tested. No symptoms were observed on plants inoculated with PBS. The bacterium was reisolated from lesions of test plants for the tree tested strains and confirmed to be the same bacterium using LOPAT tests and rep-PCR genetic fingerprinting. To our knowledge, this is the first report of P. syringae pv. syringae causing bacterial brown spot of pea in Serbia. The information of this species as a pea pathogen is still limited but is considered to be one of the limiting factor in pea cropping (Martín-Sanz et al. 2011).

AB - The emergence of Cercospora beticola populations that are resistant to benzimidazoles (MBC) and demethylation-inhibiting fungicides (DMI) has been recently reported in Serbia and has resulted in a reduced efficacy of fungicides in controlling Cercospora leaf spot (CLS). Between 2008 and 2011, using a discriminatory concentration method in sugar beet fields in two separate regions of Serbia, we determined that 93.3%-98.6% of collected C. bed cola isolates were resistant to MBCs, whereas 6.2%-42.4% were resistant to DMI fungicides. At the same localities, field trials were conducted to investigate the impact of resistant C. beticola populations on disease management From the MBC group of fungicides, both thiophanate methyl and carbendazim failed to suppress the spread of CLS at both of the tested localities. Between 2008 and 2010, DMI fungicides expressed moderate efficacy at a South Banat locality (79.8%-84.6%) whether they were applied individually (flutriafol, epoxiconazole) or in combination with MBCs (epoxiconazole/carbendazim, thiophanate-methyl/epoxiconazole). The frequency of resistant isolates in these test trials ranged from 6.2% to 10.9%. In 2011, at the same locality, conditions were observed to change in favor of the occurrence of resistant populations, which comprised up to 18.7% of the population, at the expense of DMI efficacy in CLS management At a Srem locality, the frequency of C. beticola isolates that were resistant to DMIs was high during all four years of field testing (30.5%-42.4%), and the efficacy of these fungicides ranged from 48.4% to 68.0%. A combination of DMI and a protective chlorothalonil had a stable, moderate impact on disease management regardless of the frequency of DMI resistance, whereas a combination of the cyproconazole DMI with trifloxystrobin from the strobilurin group of fungicides expressed the highest efficacy. High correlation coefficient values (r = 0.87) indicated how strongly the frequencies of resistant populations affected disease severity in the trial plots that were treated with carbendazim and thiophanate methyl, as well as in the plots that were treated with flutriafol and epoxiconazole (r = 0.98). In shift sensitivity trials, MBC-resistant C. beticola isolates were found at equally high frequencies both before and after the treatments, indicating a complete loss of efficacy in CLS control. This test revealed the significant impact of multiple DMI applications in terms of increasing the frequency of resistant C. beticola populations following treatments with flutriafol, epoxiconazole, epoxiconazole/carbendazim, thiophanate-methyl/epoxiconazole and flutriafol/chlorothalonil, except when used in combination with trifloxystrobin, in which case CLS suppression was substantially high. AB - In the recent years, several studies have shown that calcium salts may have potential as environmentally compatible, nontoxic fungicides for controlling postharvest fungal pathogens. Therefore, the objective of this study was to evaluate and compare the effects of calcium chloride and calcium hydroxide on in vitro mycelial growth, spore germination and germ tube growth of Colletotrichum acutatum, C. gloeosporioides, Alternaria alternata, and Penicillium expansum. The obtained results showed that the fungal isolates grew similarly or stimulated in the presence of 1 and 1.5% calcium salts compared to the control. After seven days of incubation, reduction of mycelial growth was observed only on PDA supplemented with 2% calcium salts. Calcium chloride and calcium hydroxide at 1.5% and 2.0% concentrations significantly decreased spore germination and germ tube growth of all fungal isolates. The results of this study show that the tested calcium salts can be used as a alternative treatment against postharvest fungal pathogens C. acutatum, C. gloeosporioides, A. alternata and P. expansum.
Kalcijum hlorid i kalcijum hidroksid u koncentracijama od 1,5 i 2,0% značajno smanjuju klijavost konidija i rast klicinih cevi svih ispitivanih izolata gljiva.

AB - Raspberry cane decay disease are the major problem in its production by reducing yield and lifetime of orchards. This disease cause a several disease-causing agents, and the most important is Didymella applanata, causal agent of raspberry spur blight. With regard to the biology of the pathogen fungicide use postharvest is not efficient enough because pathogen can infect raspberry far earlier. Therefore, the aim of this study is testing the efficacy of newer organic fungicides applied before and after the harvest raspberries and mutual comparison thereof on biological efficacy. The results show that the tested fungicides exhibited very high efficacy against D. apllanata on the raspberry leaves and the canes (from 83.0 - 96.5%).
AB - Prouzrokovači 'kompleksa sušenja' maline predstavljaju najveći problem u njenoj proizvodnji smanjujući prinose i životni vek zasada.

AB - C. apii is one of the most damaging plant pathogenic fungi of celery. During the 2012, samples of celery with typical symptoms of leaf blight were collected on the territory of Veliko Grdaište and their identification is performed using morphological and molecular methods. The pathogenicity test was conducted on healthy plants and all tested isolates caused typical symptoms after inoculation. Conidiophores arising from the stromata formed dense fascicles or single, straight to slightly curved, brown. Conidia are solitary, hyaline and septate (5-13 septa), cylindrical to obclavate, or straight to slightly curved, obtuse at the apex. Truncated and thickened at the base. Fragments of the expected length for the internal transcribed spacer (ITS1/ITS4), calmodulin (CAL-228F/CAL2Rd) and histone (CYLH3F/ CYLH3R) gene were amplified and the obtained sequences showed 100% identity with the C. apii sequences deposited in GenBank.

abstract = "C. apii is one of the most damaging plant pathogenic fungi of celery. Fragments of the expected length for the internal transcribed spacer (ITS1/ITS4), calmodulin (CAL-228F/CAL2Rd) and histone (CYLH3F/ CYLH3R) gene were amplified and the obtained sequences showed 100% identity with the C. apii sequences deposited in GenBank., C. apii predstavlja jednu od najštetnijih fitopatogenih gljiva celera.

AB - Investigation of the effect Alternaria alternata on seed germination of wheat were tested on three varieties of Victory, Renaissance and NS 40 S. Seed germination is the ISTA rules, in order to elucidate the effect of primary and secondary infection pathogen A.alternata on seed natural and conditioned seeds were placed tests with seeds untreated, treated with NaOCl and fungicide Tycoon included. The high rate of seed germination was found in all three experiments. The correlation coefficient between secondary infection of seedlings with A.alternata and germination of wheat was -0.69 to -0.76, depending on the variety, indicating that the rate of infection has no significant effect on seed germination.

AB - Morphological and cultural characteristics of Monilinia laxa isolates were studied on PDA and MEA media. The appearance of colonies (color, texture, rosettes, rosettes with black arcs), as well as conidial sze were identified. Growth of mycelium was monitored on both media, while sporulation was followed on PDA medium. Isolates formed aeriform (puffy) mycelium, with significant variations in color on different mediums, but variations in color are present within isolates grown on same medium. On MEA medium isolates are somewhat brighter then isolates grown on PDA medium. All isolates are more or less rosette, but not all of them form black arcs. Conidial size is also characteristic that is variable compared to isolates and medium. Mycelial growth is slightly lower on MEA medium.

AB - Determination of breeding and morphological characteristics of C. beticola isolates was performed on PDA, MEA i CDA mediums. Determination of morphological characteristics were evaluated by monitoring mycelial growth, texture, color, edge and color of beneath of mycelium. Isolates growth on different temperatures was monitored on PDA medium on 10°C, 15°C, 20°C, 25°C, 30°C, 35°C and 40°C. There was a significant increase in variability of the isolates on PDA, MEA and CDA mediums, but there is a variability in the growth of different isolates of the same medium too. Isolates expressed the variation in color and the width of the edge zone during growth on different substrates. The mycelium texture was cottony on all surfaces, while the beneath was brown to dark-olive colour. At temperatures of 10°C and 40°C isolates of C. beticola did not develop mycelium, and the highest increase was recorded at 25°C.
AB - Utvrđivanje morfoloških i odgajivačkih karakteristika izolata C. beticola izvedeno je na KDA, MEA i CDA podlozi. Utvrđivanje morfoloških karakteristika obuhvatilo je praćenje porasta micelije, teksture, boje, ivične zone i boje naličja micelije.

abstract = "Determination of breeding and morphological characteristics of C. beticola isolates was performed on PDA, MEA i CDA mediums. At temperatures of 10°C and 40°C isolates of C. beticola did not develop mycelium, and the highest increase was recorded at 25°C., Utvrđivanje morfoloških i odgajivačkih karakteristika izolata C. beticola izvedeno je na KDA, MEA i CDA podlozi. Utvrđivanje morfoloških karakteristika obuhvatilo je praćenje porasta micelije, teksture, boje, ivične zone i boje naličja micelije.
