AB  - The present study examined the effects of Candidatus Phytoplasma solani infection on antioxidative metabolism in leaves and roots of carrot (Daucus carota L.). Disease symptoms appeared at the end of June in the form of the chlorosis on some of the leaves, which became intensely red one week later, while the previously healthy leaves from the same branch becme chlorotic. A few days later, all leaves from the infected leaf branch were intensely red. Infected plants also had slower growth compared to the healthy ones with fewer leaf branches developed. The roots of infected plants were less developed, seared, or gummy with or without brown-colored root hair. The presence of the pathogen was detected by sequencing the 16S rRNA. National Center for Biotechnology Information (NCBI) BLAST analyses of the obtained sequence revealed 100% identity of tested strain with deposited Ca. Phytoplasma solani strains from various countries and hosts, all belonging to the "stolbur" group (16SrXII-A). Identity of 99.74% was found when the tested Serbian strain (MF503627) was compared with the reference stolbur strain STOL11 (AF248959). The oxidative damage of membranes in carrot cells was accompanied by a decrease in the content of photosynthetic pigments. Furthermore, for the determination of specific scavenging properties of the extracts, in vitro antioxidant assay was performed. In phytoplasma-infected carrot leaves, there was a greater reduction in the level of glutathione content (GSH); however; flavonoids and anthocyanidins seem to be responsible for the accompanied increased antioxidative capacity against hydroxyl radical and hydrogen peroxide.

Allelic profile of Serbian Xanthomonas campestris pv. campestris isolates from cabbage

AB  - Xanthomonas campestris pv. campestris (Xcc), the causal agent of black rot disease of cabbage (Brassica oleracea var. capitata L.), is one of the most important bacteria which affect proper cabbage growth, leading to head weight and quality losses and thereby drastically reducing its marketing value. The pathogen is genetically diverse, which is evident from the presence of eleven races worldwide and more than thirty combinations of allelic profiles. Therefore, this study aimed to determine the allelic profiles of Serbian cabbage Xcc strains obtained in 2014. The analysis was done on three selected Xcc strains whose DNA was first amplified using polymerase chain reaction (PCR) with four housekeeping genes - P-XdnaK, fyuA, gyrB, and rpoD, then sequenced, and the obtained sequences were finally used to determine allelic profiles. Allelic profiles were determined by comparison with 33 Xcc strains obtained from different hosts and regions, whose allelic profiles had been determined previously. A non-redundant database (NRDB) from the pubMLST was used for allelic profile determination and Phyloviz software for constructing a minimum spanning tree. The obtained allelic profile of all Serbian Xcc cabbage strains was 1, 3, 1, 1 for the P-X-dnaK, fyuA, gyrB and rpoD genes, respectively. This profile is assigned as sequence type 2 (ST2) and it coincides with a Portuguese B. oleracea Xcc strain, CPBF 213, originating from B. oleracea var. costata. No connection between sequence type (ST) and the host was detected.
AB  - Xanthomonas campestris pv. campestris (Xcc), prouzrokovač crne truleži kupusa (Brassica oleracea var. capitata L.) svrstava se među najvažnije fitopatogene bakterije koje utiču na pravilno razviće kupusa, dovodeći do gubitka težine glavice i njenog kvaliteta i na taj način drastično smanjuje njegovu ekonomsku vrednost. Ovaj patogen je genetički heterogen, što se ogleda kroz prisustvo dokazanih jedanaest rasa i više od trideset kombinacija alelskih profila širom sveta. Zbog svega navedenog, cilj ovog rada je determinacija alelskih profila Xcc sojeva poreklom sa kupusa prikupljenih 2014. godine. Analiza je vršena kod tri reprezentativna Xcc soja čija je DNK amplifikovana primenom lančane reakcije polimeraze (PCR) sa četiri konzervativna gena - dnaK, fyuA, gyrB i rpoD, a zatim sekvencirana i korišćena za determinaciju alelskih profila. Alelski profili su određivani poređenjem sa 33 Xcc soja izolovana sa različitih domaćina i regiona, čiji su alelski profili prethodno utvrđeni. Dobijeni alelski profil za sve Xcc sojeve sa kupusa iz Srbije je 1, 3, 1, 1 za gene dnaK, fyuA, gyrB i rpoD, redom. Ovaj profil je označen kao tip sekvence 2 (ST2) i podudara se sa portugalskim B. oleracea Xcc sojem CPBF 213 poreklom sa B. oleracea var. costata. Veza između tipa sekvence (ST) i biljke domaćina nije pronađena.

Foliar pathogens of sweet and sour cherry in Serbia

AB  - In recent years, sweet and sour cherry production in Serbia has increased. Under Serbian agroecological conditions, pathogens causing leaf diseases threaten the success of sweet and sour cherry production. In the period 2012-2019, the health status of cherries was monitored in more than 30 locations. Depending on the production system, the following leaf pathogens were identified: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina and Pseudomonas syringae pv. syringae and morsprunorum race 1. Leaf pathogens caused premature defoliation, which adversely affected bud formation for the next growing season and increased susceptibility to freezing. Therefore, attention should be focused on correct leaf pathogen identification, and proper selection, application and rotation of fungicides.
AB  - Poslednjih godina proizvodnja trešnje i višnje u Srbiji je povećana. U našim agroekološkim uslovima, patogeni lista ugrožavaju proizvodnju ovih perspektivnih voćnih vrsta. U periodu 2012-2019. godine praćeno je zdravstveno stanje trešnje i višnje, na više od 30 lokaliteta. avisno od sistema proizvodnje identifikovani su sledeći patogeni lista: Blumeriella jaapii, Wilsonomyces carpophilus, Mycosphaerella cerasella, Phoma prunicola, Podosphaera clandestina, Pseudomonas siringae pv. syringae i morsprunorum rasa 1. Patogeni lista izazivaju prevremenu defolijaciju, što negativno utiče na formiranje pupoljaka za narednu vegetaciju i povećanu osetljivost na izmrzavanje. Stoga pažnju treba usmeriti na ispravnu identifikaciju patogena, pravilan izbor i rotaciju fungicida različitog mehanizma delovanja, kao i optimalno vreme za njihovu primenu.

AB  - Bacterial leaf spot caused by the plant pathogenic bacterium Pseudomonas syringae pv. coriandricola (Psc) was observed on carrot, parsnip, and parsley grown on a vegetable farm in the Vojvodina Province of Serbia. Nonfluorescent bacterial colonies were isolated from diseased leaves and characterized using different molecular techniques. Repetitive element PCR fingerprinting with five oligonucleotide primers (BOX, ERIC, GTG5, REP, and SERE) and the randomly amplified polymorphic DNA-PCR with the M13 primer revealed identical fingerprint patterns for all tested strains. Multilocus sequence analysis of four housekeeping genes (gapA, gltA, gyrB, and rpoD) showed a high degree (99.8 to 100%) of homology with sequences of Psc strains deposited in the Plant-Associated Microbes Database and NCBI database. The tested strains caused bacterial leaf spot symptoms on all three host plants. Host-strain specificity was not found in cross-pathogenicity tests, but the plant response (peroxidase induction and chlorophyll bleaching) was more pronounced in carrot and parsley than in parsnip.

Repetitive element PCR fingerprinting with five oligonucleotide primers (BOX, ERIC, GTG5, REP, and SERE) and the randomly amplified polymorphic DNA-PCR with the M13 primer revealed identical fingerprint patterns for all tested strains. Multilocus sequence analysis of four housekeeping genes (gapA, gltA, gyrB, and rpoD) showed a high degree (99.8 to 100%) of homology with sequences of Psc strains deposited in the Plant-Associated Microbes Database and NCBI database. The tested strains caused bacterial leaf spot symptoms on all three host plants. Host-strain specificity was not found in cross-pathogenicity tests, but the plant response (peroxidase induction and chlorophyll bleaching) was more pronounced in carrot and parsley than in parsnip.",

Interactions between CLE combinations were mostly indifferent. The biocontrol potential of CLEs, mixtures of CLEs, and cell culture of B. amyloliquefaciens SS-12.6 was tested on sugar beet plants inoculated with P. syringae pv. aptata P53. The best result in inhibiting the appearance of tissue necrosis (up to 92%) was achieved with B. amyloliquefaciens SS-12.6 cell culture. Conclusion This work demonstrated significant biocontrol potential of the CLE and cell culture of B. amyloliquefaciens SS-12.6 which successfully suppress leaf spot disease severity on sugar beet plants. Significance and Impact of the Study The findings of biocontrol of sugar beet emerging pathogen will contribute to growers in terms of alternative disease control management. This study represents first assessment of biological control of P. syringae pv. aptata.

AB  - Four Gram-negative, rod-shaped pectinolytic bacterial strains designated as 2M, 9M, DPMP599 and DPMP600 were subjected to polyphasic analyses that revealed their distinctiveness from the other Pectobacterium species. Strains 2M and 9M were isolated from Calla lily bulbs cultivated in Central Poland. DPMP599 and DPMP600 strains were isolated from Calla lily leaves from plants grown in Serbia. Phylogenetic analyses based on nine housekeeping genes (gapA, gyrA, icdA, pgi, proA, recA, recN, rpoA, and rpoS), as well as phylogeny based on the 381 most conserved universal proteins confirmed that Pectobacterium zantedeschiae strains were distantly related to the other Pectobacterium, and indicated Pectobacterium atrosepticum, Pectobacterium betavasculorum, Pectobacterium parmentieri and Pectobacterium wasabiae as the closest relatives. Moreover, the analysis revealed that Pectobacterium zantedeschiae strains are not akin to Pectobacterium aroidearum strains, which were likewise isolated from Calla lily. The genome sequencing of the strains 2M, 9M and DPMP600 and their comparison with whole genome sequences of other Pectobacterium type strains confirmed their distinctiveness and separate species status within the genus based on parameters of in silico DNA–DNA hybridization and average nucleotide identity (ANI) values. The MALDI-TOF MS proteomic profile supported the proposition of delineation of the P. zantedeschiae and additionally confirmed the individuality of the studied strains. Based on of all of these data, it is proposed that the strains 2M, 9M, DPMP599, and DPMP600 isolated from Calla lily, previously assigned as P. atrosepticum should be reclassified as Pectobacterium zantedeschiae sp. nov. with the strain 9M T (PCM2893 = DSM105717 = IFB9009) as the type strain.

AB  - This paper presents data on the dynamic of B. aeneus populations in winter oilseed rape in the downriver section of the Danube in Serbia, which were acquired by yellow water traps and a beating method. Their susceptibility to several insecticides of different classes (organophosphates, pyrethroids and neonicotinoids) (adult vial test and dipping test) was tested over two seasons (2009 and 2010). B. aeneus populations developing under the agroecological conditions that exist in Serbia were monitored to detect the moment of OSR infestation during its sensitive growth stages. Adults were counted, and the data revealed that they infest winter OSR crops during the stem elongation growth stage (BBCH 30-31), reaching a population peak at the green-yellow bud stage (BBCH 57-59), and exceeding the economic threshold, while the populations decreased substantially during the subsequent stage (BBCH 61-69). Laboratory test results did not confirm any changes in B. aeneus susceptibility/resistance to pyrethroid, organophosphate and neonicotinoid insecticides. Migration of B. aeneus populations was monitored as a way of developing predictive models for estimation of infestation severity and the timing of pest outbreaks under agroecological conditions existing in Serbia. As resistance to pyrethroids has been confirmed in B. aeneus populations in many European countries, their susceptibility will be further monitored.

A blight disease on highbush blueberry associated with Macrophomina phaseolina in Serbia

AB  - Unusual blight-like symptoms appeared on highbush blueberry plants in Serbia during August 2015 and infected plants showed browning and reddening of leaves, drying of foliage and brown discolouration of internal vascular stem tissues. The objective of this study was to isolate and confirm a causal agent of the disease. Five diseased blueberry plants (2-year-old), with visible brown discolouration in the wood, were collected for isolation on potato dextrose agar (PDA). Morphological analysis of the selected fungal isolates showed the presence of abundant black, round to oblong, or irregularly shaped microsclerotia immersed in the PDA. Dark, globose pycnidia formed on water agar with an initially hyaline, granular content and single-celled conidia, indicating the presence of plant pathogenic fungus Macrophomina phaseolina associated with symptomatic plant tissues. Pathogenicity was confirmed on potted blueberry plants based on the initial symptoms of leaves turning yellowish to brown at the leaf edges, followed by the defoliation of leaves of the inoculated stems. Discolouration of vascular tissues was also observed on transverse sections of inoculated stems. The pathogen M. phaseolina was confirmed using molecular analysis of the ITS1-5.8S-ITS2 region of rDNA and a part of the TEF-1 gene region. This is the first report of M. phaseolina causing a blight disease on highbush blueberry in Serbia. The study should help in elucidating disease symptomatology and provide information on the risk which this fungus could pose in blueberry production.

A preliminary study of antibacterial activity of thirty essential oils against several important plant pathogenic bacteria

AB  - Numerous scientific research studies all over the world have addressed the problem of agriculture in the 21st century as being particularly sensitive to climate change, which has caused phytopathogenic bacteria to spread. Therefore, there is a clear and urgent need to contain this kind of risk in agricultural production (both conventional and organic farming). The objective of this study was to determine the antibacterial activity of 30 essential oils (EOs) against three harmful plant pathogenic bacteria of agricultural importance, Erwinia amylovora, Xanthomonas campestris pv. campestris and Pseudomonas syringae pv. syringae. The study included in vitro testing, using an agar-diffusion assay. The EOs of Ceylon cinnamon (leaf and bark), oregano, clove bud and palmarosa revealed antibacterial activity against the test bacteria, and the maximum mean inhibition zone diameters of 35 mm was found against E. amylovora and X. campestris pv. campestris (highly sensitive reaction), while it was smaller in the case of P. syringae pv. syringae, from 18.25-26.25 mm (sensitive to very sensitive reaction). Maximum diameter of the zone of inhibition (35 mm) was obtained using basil and peppermint against E. amylovora, and rosemary, blue gum and camphor tree against X. campestris pv. campestris. Not a single EO inhibited P. syringae pv. syringae with the resulting total diameter zone of 35 mm, and this test bacteria was resultingly classified as the least susceptible bacterium of the three tested. EOs of lemongrass, aniseed, ylang ylang, silver fir, lemon, dwarf mountain pine, bay laurel and scots pine caused sensitive reaction of the tested bacteria. Peppermint, black cumin, Indian frankincense, bergamot orange, common juniper, bitter orange and neem produced variable reactions from total to weakly or no inhibition at all. Weakly activity was found in niaouli and Atlas cedar. Eastern red cedar, patchouli, Indian sandalwood and ginger caused no reaction of any of the test bacteria. The results offer a basis for further work based on in vivo testing for the purpose of developing ‘natural pesticides’ for control of phytopathogenic bacteria, thus giving a significant contribution to reducing yield losses in agriculture and sustainable development.
AB  - Brojna naučna istraživanja širom sveta potvrđuju da je poljoprivreda u 21. veku posebno osetljiva na klimatske promene koje su uzrok širenja fitopatogenih bakterija. Stoga je jasna hitna potreba za ublažavanjem ovog rizika u poljoprivrednoj proizvodnji (u konvencionalnoj i organskoj poljoprivredi). Cilj ovog rada je određivanje antibakterijske aktivnosti 30 etarskih ulja prema tri ekonomski značajne fitopatogene bakterije, Erwinia amylovora, Xanthomonas campestris pv. campestris i Pseudomonas syringae pv. syringae. Istraživanja su vršena u in vitro uslovima, korišćenjem agar-difuzne metode. Etarska ulja pravog cimeta (od lista i kore), origana, zatim karanfilića i palmaroze, su pokazala antibakterijsku aktivnost prema testiranim sojevima bakterija, ostvarujući zone inhibicije maksimum prečnika 35 mm dobijene u slučaju E. amylovora i X. campestris pv. campestris (visoko osetljiva reakcija), a u slučaju P. syringae pv. syringae manju, od 18.25-26.25 mm (osetljiva do vrlo osetljiva reakcija). Maksimalni prečnik inhibicione zone (35 mm) je takođe dobijen primenom ulja bosiljka i pitome nane prema E. amylovora i ruzmarina, eukaliptusa i ravensare prema X. campestris pv. campestris. Kod P. syringae pv. syringae ni u jednom slučaju primene ulja nije postignut maksimalan prečnik inhibicije od 35 mm, na osnovu čega je ova bakterija svrstana kao slabije osetljiva. Etarska ulja limun trave, anisa, ilang-ilanga, evropske jele, limuna, planinskog bora, lovora i belog bora su rezultirala osetljivom reakcijom testiranih sojeva bakterija. Pitoma nana, čurukot, tamjan, begramot, kleka, gorka pomorandža i nim su izazvali varijabilnu reakciju, od potpune inhibicije, do slabe ili čak i bez inhibicije. Slaba aktivnost je ostvarena kod niaoulija i atlaskog kedra. Sve tri testirane bakterije nisu pokazale reakciju prema virdžinijskoj kleki, pačuliju, sandalovini i đumbiru. Rezultati dobijeni u ovom radu daju osnovu za dalja istraživanja in vivo, sa svrhom razvoja 'prirodnih pesticida' koji se mogu primeniti za suzbijanje fitopatogenih bakterija, čime se daje značajan doprinos u smanjenju gubitaka prinosa u poljoprivredi i održivom razvoju.

Genetic diversity and pathogenicity of Pseudomonas syringae pv. aptata isolated from sugar beet

AB  - Pseudomonas syringae pv. aptata is the causal agent of bacterial leaf spot disease of sugar beet (Beta vulgaris). During 2013, 250 samples were collected from leaf lesions with typical symptoms of bacterial leaf spot in commercial fields of sugar beet in Serbia, and 104 isolates of P.syringae pv. aptata were obtained. Identification and characterization was performed using biochemical, molecular and pathogenicity tests. Identification included LOPAT tests and positive reactions using primers Papt2F and Papt1R specific for P.syringae pv. aptata. Repetitive (rep) sequence-based PCR typing with ERIC, REP and BOX primers revealed high genetic variability among isolates and distinguished 25 groups of different fingerprinting profiles. Pulse-field gel electrophoresis (PFGE) and multilocus sequence analysis (MLSA) of representative isolates showed higher genetic variability than in rep-PCR analysis and distinguished three and four major genetic clusters, respectively. A pathogenicity test performed with 25 representative isolates on four cultivars of sugar beet confirmed the occurrence of leaf spot disease and showed correlation between the most aggressive isolates and the genetic clusters obtained in MLSA. All these findings point to the existence of several lines of P.syringae pv. aptata infection in Serbia that are genetically and pathologically different.

Evaluation of cherry cultivar susceptibility to bacterial canker and leaf spot disease

AB  - In this study, the susceptibility of different sweet (19) and sour cherry (3) cultivars (cvs.) to the bacterial pathogens Pseudomonas syringae pv. syringae and P. s. pv. morsprunorum race 1 causing bacterial canker (sensu stricto) and leaf spot was evaluated. Susceptibility to bacterial canker was examined by artificial inoculations of 1-year-old rooted trees, which revealed susceptibility of all cvs. to both P. syringae pathovars. Susceptibility level differed among cvs.; sweet cherry cvs. Katalin, Linda, Summit, New Star and Bigarreau Burlat were highly susceptible, sour cherry cvs. erdi botermo and sweet cherry Drogan's Yellow, Carmen, Germersdorfer and Noir de Meched were susceptible. Sour cherry cvs. Spanska and ujfehertoi furtos, as well as sweet cherry cv. Rita were found to be less susceptible to bacterial canker. Susceptibility to bacterial leaf spot disease was evaluated on naturally infected (field conditions) and artificially inoculated leaves (laboratory conditions). Severe symptoms of bacterial spot with shot hole appearance were observed in cherry leaves at the beginning of May under field conditions. In all collected leaf samples, presence of P. s. pv. morsprunorum race 1 was determined. Sweet cherries Valery Chkalov, Summit, Bigarreau Burlat and New Star were highly susceptible to this bacterium. No symptoms of bacterial spot were found on sweet cherries Solomary gomboly, Linda, Germersdorfer, Hedelfinger, Katalin, Margit, Carmen, Aida, Sunburst, Alex, Peter, Rita and Kavics and sour cherry erdi botermo. In laboratory conditions, susceptibility of the tested cvs. varied and mostly depended on the inoculation method. Nonetheless, sweet cherry cvs. Carmen and Margit and sour cherry erdi botermo exhibited a certain level of resistance.

Biochemical changes in Oenothera biennis plants infected by 'Candidatus Phytoplasma solani'

AB  - The aim of the present paper was to study the response of Oenothera biennis L. to 'Candidatus Phytoplasma solani' (Stolbur) infection by analyzing total sugars, polyphenols, photosynthetic pigments content and the antioxidant capacity in leaves and roots of healthy and infected plants. The infection caused a significant increase in peroxidation of lipids, phenylalanine ammonia-lyase activity, total sugar, polyphenols and anthocyanins content (2.8, 2.6, 1.8, 1.4, 6.8 fold, respectively), as well as a decrease in photosynthetic pigments (2-6 fold) and total flavonoids (1.5 fold) in the leaves of Oe. biennis. Changes in these parameters were insignificant in the roots except for the total polyphenols content that was 2.7 times higher in the infected ones. Reduced gluthatione content in both tested organs was not affected by the infection (3.7 and 1.7 mu mol/g fresh weight of leaves and roots, respectively). The elevated content of total sugars, flavonoids and polyphenols, as well as the reduction of photosynthetic pigments and anthocyanins in infected plants are indicative of changes in the metabolism of Oe. biennis affected by the Stolbur phytoplasma. In addition to reduction of chlorophyll and carotenoids, the phytoplasma accelerated leaf senescence. Plants responded to the infection via enhanced superoxide anion scavenging, even though this reaction did not prevent, apparently, membrane damage in analysed leaves. abstract = "The aim of the present paper was to study the response of Oenothera biennis L. to 'Candidatus Phytoplasma solani' (Stolbur) infection by analyzing total sugars, polyphenols, photosynthetic pigments content and the antioxidant capacity in leaves and roots of healthy and infected plants. The infection caused a significant increase in peroxidation of lipids, phenylalanine ammonia-lyase activity, total sugar, polyphenols and anthocyanins content (2.8, 2.6, 1.8, 1.4, 6.8 fold, respectively), as well as a decrease in photosynthetic pigments (2-6 fold) and total flavonoids (1.5 fold) in the leaves of Oe. biennis. Changes in these parameters were insignificant in the roots except for the total polyphenols content that was 2.7 times higher in the infected ones. Reduced gluthatione content in both tested organs was not affected by the infection (3.7 and 1.7 mu mol/g fresh weight of leaves and roots, respectively). The elevated content of total sugars, flavonoids and polyphenols, as well as the reduction of photosynthetic pigments and anthocyanins in infected plants are indicative of changes in the metabolism of Oe. biennis affected by the Stolbur phytoplasma. In addition to reduction of chlorophyll and carotenoids, the phytoplasma accelerated leaf senescence. Plants responded to the infection via enhanced superoxide anion scavenging, even though this reaction did not prevent, apparently, membrane damage in analysed leaves. AB  - This study presents the results of a multiple-year evaluation (1991-1996; 2005-2007) of susceptibility of more than 100 apple cultivars to Venturia inaequalis, Podosphaera leucotricha and Erwinia amylovora under agroecological conditions existing in Serbia. Some of the most popular cultivars were found highly susceptible to V. inaequalis (Cripps Pink, Mutsu, Gloster 69, Wellspur); while Golden Delicious, Richared, Gala, Čačanska pozna, Čadel and Jonagold were susceptible; Idared, Granny Smith and Jonathan moderately susceptible; Lord Lamburne and London Pepping, as well as several autochthonous cultivars were moderately resistant; and a group of resistant cultivars included Prima, Priscilla, Williams Pride, Dayton, Enterprise, Gold Rush, Golden Orange, many of the Re-cultivars (Germany), as well as Baujade, Selena, Dukát, Produkta, Topaz, some older cvs. (Worcester Pearmain, Merton Worcester, James Grieve, Akane, Astilish, Astrachan Red and Discovery), some Co-op selections (USA) and NS hybrids (Serbia). Regarding powdery mildew, Idared and Jonathan were highly susceptible; Gala, Akane, Jonagold, Priscilla, Mutsu, Čačanska pozna, Golden Delicious, Granny Smith, Čadel and GoldRush were susceptible; Wellspur, Astrachan Red, Richared, Jonadel, Dayton and several autochthonous cultivars were moderately susceptible; Lord Lamburne, Astlisch, Prima, Champagne Reinette, Discovery and many autochthonous cultivars were moderately resistant; while most Re-cvs. (Germany), several cultivars from the Czech Republic, some selections from the USA and UK and most NS hybrids (Serbia) were resistant. Also, some cultivars showed variable susceptibility depending on location (Williams Pride, Gloster 69, Baujade and Produkta). E. amylovora was observed only in 2007 and at relatively low intensity (up to 12% infection). The highest disease severity was observed on cv. Elstar, then Granny Smith, Idared and Jonagored; while the lowest was found on Red Chief and Hapke apple trees.

Characterization of Pseudomonas syringae pv. syringae, Causal Agent of Citrus Blast of Mandarin in Montenegro

AB  - Citrus blast caused by bacterium Pseudomonas syringae is a very important disease of citrus occuring in many areas of the world, but with few data about genetic structure of the pathogen involved. Considering the above fact, this study reports genetic characterization of 43 P syringae isolates obtained from plant tissue displaying citrus blast symptoms on mandarin (Citrus reticulata) in Montenegro, using multilocus sequence analysis of gyrB, rpoD, and gapl gene sequences. Gene sequences from a collection of 54 reference pathotype strains of P syringae from the Plant Associated and Environmental Microbes Database (PAMDB) was used to establish a genetic relationship with our isolates obtained from mandarin. Phylogenetic analyses of gyrB, rpoD, and gapl gene sequences showed that P syringae pv. syringae causes citrus blast in mandarin in Montenegro, and belongs to genomospecies 1. Genetic homogeneity of isolates suggested that the Montenegrian population might be clonal which indicates a possible common source of infection. These findings may assist in further epidemiological studies of this pathogen and for determining mandarin breeding strategies for P syringae control.

Chlorine dioxide as a disinfectant for Ralstonia solanacearum control in water, storage and equipment

AB  - Brown rot or bacterial wilt caused by bacterium Ralstonia solanacearum is the main limiting factor in potato production. Quarantine measures are necessary to avoid spread of disease to disease-free areas. R. solanacearum has been shown to contaminate watercourses from which crop irrigation is then prohibited causing further potential losses in yield and quality. The bacteria also spread via surfaces that diseased seed potatoes come into contact with. This study showed bactericidal activity of chlorine dioxide (CIO2) on R. solanacearum for disinfection of water, surface and equipment. The results showed that CIO2 solution at concentration of 2 ppm at 30 minutes of exposure time had bactericidal effect for disinfection of water. For surface and equipment disinfection, concentration of 50 ppm showed total efficacy at 30 min and 5 sec exposure time, respectively. Results suggest that use of CIO2 as a disinfectant has a potential for control of brown rot pathogen in water, storage and equipment.
AB  - Mrka trulež ili bakteriozna uvelost krompira prozrokovana bakterijom Ralstonia solanacearum ograničavajući je faktor uspešne proizvodnje krompira. Sprovođenje karantinskih mera zaštite je neophodno kako bi se izbeglo širenje bakterije u regione u kojima bolest nije prisutna. S obzirom da R. solanacearum može kontaminirati vodene tokove i izvore koji služe za navodnjavanje useva, zabrana korišćenja dovodi do dodatnih potencijalnih gubitaka u prinosu i kvalitetu krompira. Bakterija takođe kontaminira površine sa kojima zaraženi semenski krompir dolazi u dodir. U ovom radu prikazana je baktericidna aktivnost hlor-dioksida (CIO2) na bakteriju R. solanacearum u cilju dezinfekcije vode, površine i opreme. Dobijeni rezultati pokazuju da CIO2 ispoljava baktericidni efekat za dezinfekciju vode u koncentraciji od 2 ppm nakon 30 minuta ekspozicije, za dezinfekciju površine u koncentraciji 50 ppm nakon 30 minuta ekspozicije i za dezinfekciju opreme u koncentraciji 50 ppm nakon 5 sekundi ekspozicije. Occurrence, identification and phylogenetic analysis of Fusarium proliferatum on bean seed (Phaseolus vulgaris L.) in Serbia

AB  - During the routine quality control analysis on bean seeds in 2015, Fusarium fungal infection was observed on an average of 17% of the bean seed. The objective of this paper was isolation and identification of Fusarium sp. based on the pathogen's morphological and molecular characteristics. Morphological identification of Fusarium isolates was performed on PDA and CLA. DNA of 14 Fusarium sp. isolates was extracted directly from the mycelium (~ 100 mg wet weight), with a Dneasy Plant Mini Kit (Qiagen, Hilden, Germany). Following DNA extraction, the translation elongation factor 1-alpha region was amplified by PCR with the primer pair EF1 and EF2. An amplicon of 700 bp was amplified in all tested isolates. Identification of one isolate was performed by sequencing the translation elongation factor EF-1a gene. Completed morphological and molecular characteristics of isolates, as well as the results of sequencing confirmed that Fusarium proliferatum was the causal agent of bean seed rot.
AB  - Tokom rutinske kontrole kvaliteta semena pasulja (Phaseolus vulgaris L.) u 2015. godini ustanovljeno je prisustvo Fusarium sp. u visokom procentu, u proseku 17%. Cilj ovog rada bio je izolacija i identifikacija prouzrokovača truleži semena pasulja, na osnovu morfoloških i molekularnih karakteristika patogena. Odgajivačke karakteristike 14 odabranih izolata ocenenjene su na PDA i CLA podlogama. Ekstrakcija DNK 14 odabranih izolata obavljena je direktno iz 100 mg sveže micelije, korišćenjem Dneasy Plant Mini Kit (Qiagen, Hilden, Nemačka). Amplifikacija DNK obavljena je pomoću PCR korišćenjem para prajmera EF1 i EF2. U svim proučavanim izolatima formirani su amplikoni veličine 700 bp. Identifikacija jednog odabranog izolata izvršena je sekvenciranjem translacionog faktora EF-1 a gena. Na osnovu dobijenih rezultata ustanovljeno je da je prouzrokovač truleži semena pasulja Fusarium proliferatum.

Characterization of Xanthomonas hortorum pv. pelargonii Isolated from Geranium in Serbia

AB  - Geranium leaves and stems with symptoms of bacterial blight were collected from commercial greenhouses during the last decade in Serbia. In total, 17 isolates with colony morphology typical for the genus Xanthomonas were characterized with pathogenicity, biochemical, serological, and molecular assays. All 17 isolates reacted positive in a polymerase chain reaction (PCR) using XcpM1 and XcpM2 primers specific for Xanthomonas hortorum pv. pelargonii. In pathogenicity tests on Pelargonium zonale (leaf and stem inoculation), all isolates caused typical symptoms on leaves starting 2 days after inoculation as sunken, water-soaked, irregular lesions, and 6 to 8 days after inoculation on stems as necrotic lesions also showing yellow exudate. Symptoms resulted in general wilting of inoculated plants 20 days after inoculation. Selected phenotypic tests indicated that all isolates showed the same results as described for the bacterium X. hortorum pv. pelargonii. Repetitive sequence-based PCR typing using BOX and ERIC revealed that all isolates showed two fingerprinting profiles but (GTG)(5) and REP did not reveal differences. Multilocus sequence typing of partial sequences of rpoD, dnaK, fyuA, and gyrB genes of tested isolates and sequences obtained from GenBank of Xanthomonas pathovar pathotype strains did not reveal genetic variability among the isolates, showing the same gene sequence pattern.

AB  - The purpose of the present study was to investigate the genetic diversity of X. arboricola pv. juglandis strains in Serbia. This bacterium is the causal agent of walnut blight and is also associated with apical necrosis of immature walnut fruits. Although walnut blight is long known and widespread in Serbia, a systematic strain diversity study of Xanthomonas arboricola pv. juglandis strains from different regions in Serbia has not been performed. The objectives of this work were to examine the molecular diversity and its possible biological significance of 59 isolates of X. arboricola pv. juglandis collected from different geographic locations in Serbia. Genomic variability was assessed by using repetitive PCR, SpeI macrorestriction analysis of genomic DNAs by pulsed-field gel electrophoresis (PFGE) and partial sequencing of the gyrB gene. Molecular analyses showed substantial genetic diversity among strains and existence of diverse populations of X. arboricola pv. juglandis in Serbia.
