The effect of IAA producing Bacillus sp. Q3 strain on marshmallow seed germination

AB  - Marshmallow (Althea officinalis) is one of the important medicinal plants in Serbia. One of the bigest problem in growing of marshmallow is poor seeds germination. Rhyzospheric bacteria able to produced plant growth stimulating hormones can inpruve seed germinations and decreace seed infections with patogens. IAA production by Bacillus sp. Q3 strain estimate in this investigation ranged from 3.76-10.62 μgml-1. The application of soil bacteria as the antagonists to the growth of pathogenic fungi, indicated that IAA producing Bacillus sp. strain Q3 demonstrated not only a high level of antagonism towards the seed mycoflora, but significantly increased the germination rate of the marshmallow seeds. Soaking marshmarrow seeds for 24 h in the 105 CFU ml-1 of investigated strain Bacillus sp. strain Q3 increased the 55.1% germination and decreased the percentage of the seed infection by the fungus Alternaria alternata, compared to the control (32%). Strong increasing percent germination of marshmallow's seeds from 26.9-55.1% and decreasing seed infection with phytopathogen Alternaria alternate, as predominant marshmallow seeds pathogen, can recommend this strain for seed protection and as PGPR.

AB  - The presence and distribution of grapevine phytoplasma Flavescence doree (FD) was investigated in the eight important viticultural areas of Serbia from 2005 to 2007. Overall 272 vineyards were investigated in 67 vineyard localities. Results are presented through phytoplasma infection FD C by locality and grapevine cultivars. The presence of FD stain C was detected in seven of nine investigated areas. Phytoplasma FD was detected in the six grapevine cultivars - Plovdina, Frankovka, Zupski bojadiser, Smederevka, Italian Riesling and Black Burgundy, of eight tested cultivars. The cultivar Plovdina appeared to be extremely sensitive to FD phytoplasma. Phytoplasma presence in grapevine is checked by PCR and RFLP. The RFLP patterns obtained from the Serbian grapevine samples confirmed that all detected FD strains belong to 16SrV-C subgroupe of phytoplasma.

Screening of antagonistic activity of microorganisms against Colletotrichum acutatum and Colletotrichum gloeosporioides

AB  - The antagonistic activities of five biocontrol agents: Trichoderma harzianum, Gliocladium roseum, Bacillus subtilis, Streptomyces noursei and Streptomyces natalensis, were tested in vitro against Colletotrichum acutatum and Colletotrichum gloeosporioides, the causal agents of anthracnose disease in fruit crops. The microbial antagonists inhibited mycelial growth in the dual culture assay and conidial germination of Colletotrichum isolates. The two Streptomyces species exhibited the strongest antagonism against isolates of C. acutatum and C. gloeosporioides. Microscopic examination showed that the most common mode of action was antibiosis. The results of this study identify T. harzianum, G. roseum, B. subtilis, S. natalensis and S. noursei as promising biological control agents for further testing against anthracnose disease in fruits.
AB  - Antagonističko delovanje pet agenasa biološke zaštite: Trichoderma harzianum, Gliocladium roseum, Bacillus subtilis, Streptomyces noursei i Streptomyces natalensis, testirano je in vitro na Colletotrichum acutatum i Colletotrichum gloeosporioides, prouzrokovače antraknoze plodova. Svi mikrobni antagonisti inhibiraju porast micelije u testovima dvojne kultivacije i klijavost konidija Colletotrichum izolata. Dve vrste roda Streptomyces ispoljavaju najjači stepen antagonizma prema izolatima C. acutatum i C. gloeosporioides. Mikroskopsko ispitivanje pokazuje da je antibioza najčešći način delovanja antagonista. T. harzianum, G. roseum, B. subtilis, S. natalensis i S. noursei su rezultatima ovih istraživanja identifikovani kao biološki agensi koji se mogu uspešno uključiti u buduća testiranja u cilju suzbijanja antraknoze plodova.

AB  - Factors associated with conidial germination and germ tube length of Colletrichum gloeosporioides, a Polystigma rubrum subsp. rubrum stromata mycoparasite, have been studied in vitro. Maximal germination was obtained at 20°C and pH ranging from 6-7. An incubation period of 3-8 prior to germination, depending on temperature, was required. Germination and germ tube length decreased when conidia were exposed to sunlight or if the concentration of conidia in suspension increased. There was poor germination (2.5% after 24 h) at high relative humidity (near 100%). Stromata fragments added into conidial suspension increased germination and germ tube length. Conidia discharged from acervuli on parasited stromata kept germination viability for more than 4 months, while germination of those placed on glass slides was negligible after 4 weeks.
