AB  - Aim of study: To report the occurrence of Pepino mosaic virus (PepMV) on tomato in Serbia and to genetically characterize Serbian PepMV isolates. Area of study: Tomato samples showing virus-like symptoms were collected in the Bogojevce locality (Jablanica District, Serbia). Material and methods: Collected tomato samples were assayed by DAS-ELISA using antisera against eight economically important or quarantine tomato viruses. Three selected isolates of naturally infected tomato plants were mechanically transmitted to tomato ‘Novosadski jabučar’ seedlings. For confirmation of PepMV infection, RT-PCR was performed using specific primers PepMV TGB F/PepMV UTR R. Maximum-likelihood phylogenetic tree was constructed with 47 complete CP gene sequences of PepMV to determine the genetic relationship of Serbian PepMV isolates with those from other parts of the world. Main results: The results of DAS-ELISA indicated the presence of PepMV in all tested samples. Mechanically inoculated ‘Novosadski jabučar’ seedlings expressed yellow spots and light and dark green patches, bubbling, and curled leaves. All tested tomato plants were RTPCR positive for the presence of PepMV. The CP sequence analysis revealed that the Serbian PepMV isolates were completely identical among themselves and shared the highest nucleotide identity of 95.1% (99.2% aa identity) with isolate from Spain (FJ263341). Phylogenetic analysis showed clustering of the Serbian PepMV isolates into CH2 strain, but they formed separate subgroup within CH2 strain. Research highlights: This is the first data of the presence of PepMV in protected tomato production in Serbia. Considering increased incidence and rapid spread in Europe, the presence of PepMV on tomato could therefore represent serious threat to this valuable crop in Serbia

Occurrence and molecular characterization of Impatiens necrotic spot tospovirus in ornamentals in Serbia

AB  - A subset of ornamentals grown in Serbia were surveyed for the presence of impatiens necrotic spot tospovirus (INSV) from 2008 to 2018. Out of 1578 samples tested for the presence of the virus using double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), INSV was detected in 26 Begonia sp. and four Tulipa sp. samples collected in 2008 and in 10 samples of each of Begonia sp., Impatiens walleriana and I. hawkeri 'SunPatiens' and 'New Guinea' collected in 2018. INSV was not detected in any samples collected during 2009-2017. For further confirmation, the nucleotide sequence of the INSV nucleocapsid (N) gene was obtained by reverse transcription polymerase chain reaction (RT-PCR) in two and four samples collected in 2008 and 2018, respectively. The success of mechanical transmission of INSV isolates from different hosts was variable depending on isolates. Serbian INSV isolates showed very low nucleotide diversity and they were closely related to the Chinese and Japanese isolates already reported in the literature, indicating that the Serbian isolates might be of Asian origin. Phylogenetic analysis and the median-joining haplotype network of the N gene sequences showed clustering of Serbian INSV isolates in the Asian clade into two different subclades with different recent common ancestors implying two independent introductions. The sporadic occurrence of INSV with high temporal distance is a likely consequence of virus spread through the international trade in ornamental planting material. Therefore, the establishment of the virus should be considered and prevented when introducing new plant material into the country. This is also the first report of the tulip as a natural host of INSV.

Characterization of Gnomoniopsis idaeicola, the Causal Agent of Canker and Wilting of Blackberry in Serbia

AB  - Blackberry cane diseases with the symptoms of necrosis, canker, and wilting are caused by several fungi worldwide. Surveys conducted from 2013 to 2016 in Serbia revealed the occurrence of Gnomoniopsis idaeicola, the causal agent of cane canker and wilting, which was found to be distributed in almost half of the surveyed orchards, in three blackberry cultivars, and with disease incidence of up to 80%. Wide distribution and high disease incidence suggest that G. idaeicola has been present in Serbia for some time. Out of 427 samples, a total of 65 G. idaeicola isolates were obtained (isolation rate of 34.19%). Reference isolates, originating from different localities, were conventionally and molecularly identified and characterized. G. idaeicola was detected in single and mixed infections with fungi from genera Paraconiothyrium, Colletotrichum, Diaporthe, Botryosphaeria, Botrytis, Septoria, Neofusicoccum, and Discostroma, and no diagnostically specific symptoms could be related directly to the G. idaeicola infection. In orchards solely infected with G. idaeicola, blackberry plant mortality was up to 40%, and yield loses were estimated at 50%. G. idaeicola isolates included in this study demonstrated intraspecies diversity in morphological, biological, pathogenic, and molecular features, which indicates that population in Serbia may be of different origin. This is the first record of a massive outbreak of G. idaeicola infection, illustrating its capability of harmful influence on blackberry production. This study represents the initial step in studying G. idaeicola as a new blackberry pathogen in Serbia, aiming at developing efficient control measures.

Incidence and distribution of leek yellow stripe virus in allium crops in Serbia

AB  - Leek yellow stripe virus (LYSV) is one of the most frequent and important viruses in leek and garlic crops worldwide. In Serbia this virus is found both in leek and garlic, and often at high percentages. During two consecutive years, 2012 and 2013, a total 92 samples were collected from 11 inspected leek-, garlic- and onion-growing locations and they were analyzed for the presence of LYSV using DAS-ELISA. LYSV was detected in 31.5% of the tested samples. In 2012, the presence of LYSV was only detected in leek plants, and in 55.6% of the tested samples. During 2013, LYSV was detected in 85% of leek and 58.3% of garlic samples. In total, LYSV was detected in 56.4% of leek samples and 17.1% garlic samples. LYSV incidence was confirmed using RT-PCR with LYSV specific primers amplifying 1020 bp fragment representing coat protein and part of nuclear inclusion B genes. Molecular identification was confirmed by sequencing of two selected isolates, 181-13 (MG242625) from garlic and 298-13 (MG242624) from leek, and comparing them to the GenBank sequences of LYSV. Phylogenetic analysis of 55 sequences of LYSV from all over the world showed some correlation between host plant and geographical origin of the isolates, forming five separate clades. Two Serbian LYSV isolates fell into distant clades. The Serbian leek isolate 298-13 of LYSV belongs to clade B, while isolate 181-13 originating from garlic belongs in clade E.
AB  - Virus žute prugavosti praziluka (Leek yellow stripe virus, LYSV) je jedan od najčešćih i najznačajnijih virusa na praziluku i belom luku. U Srbiji je virus detektovan na obe kulture, i praziluku i belom luku, a često se javlja u visokim procentima. Tokom dve uzastopne godine (2013. i 2014.) pregledano je 11 lokaliteta gajenja praziluka, belog i crnog luka i sakupljena su 92 uzorka koja su DAS-ELISA metodom testirana na prisustvo LYSV. Prisustvo LYSV je dokazano u 31,5% testiranih uzoraka. Tokom 2012., prisustvo LYSV dokazano je samo u praziluku, u 55,6% testiranih uzoraka. AB  - Proučavanjem pojave i rasprostranjenosti virusa mozaika krastavca u usevu lubenice u Srbiji tokom 2012. godine, prisustvo virusa utvrđeno je u pet od šest pregledanih lokaliteta. Prisustvo virusa ustanovljeno je u 24,05% serološki testiranih uzoraka (DAS-ELISA), a najzastupljeniji je bio na lokalitetu Silbaš. Tokom ovih istraživanja virus mozaika krastavca češće je bio prisutan u mešanim infekcijama sa ZYMV i/ili WMV (12,66%), dok je prisustvo pojedinačne infekcije utvrđeno u nešto nižem procentu, 11,39%. Mehaničkim inokulacijama N. glutinosa, dobijeno je pet izolata, od kojih su dva odabrana za dalju biološku karakterizaciju. Na osnovu tipa i jačine simptoma na test-biljkama, utvrđena je fenotipska varijabilnost ispitivanih izolata. Molekularna detekcija obavljena je amplifikacijom fragmenta dužine 871 bp kod svih ispitivanih izolata korišćenjem para prajmera CMVCPfwd/CPrev, koji omogućavaju umnožavanje gena za protein omotača i deo 5' i 3' UTR regiona. Analizom sekvenci kompletnog gena za protein omotača i rekonstrukcijom filogenetskog stabla, utvrđena je pripadnost izolata CMV iz lubenice iz Srbije podgrupi IA, koja obuhvata većinu izolata odabranih za filogenetsku analizu.

AB  - Sixteen samples of sorghum seed (Sorghum bicolor (L.) Moench.) 'Alba', 'Gold', 'Prima' and 'Reform' were analyzed in the localities of Bački Petrovac and Čantavir in the period 2009-2011. From the infected sorghum seed, monosporial cultures identified as Epicoccum nigrum based on morphology, proved their pathogenicity on artificially inoculated sorghum seedlings. Molecular identification was performed by PCR and amplification of the ITS region of ribosomal DNA. Gene sequences of selected isolates 291-09 (JQ619838) and 315-09 (JQ619839) exhibited 99-100% nucleotide identity with the sequences of 31 isolates of E. nigrum deposited in the GenBank. It obtained results represent the first detailed characterization of E. nigrum in Serbia. The presence of a large number of phytopathogenic fungi on sorghum seed should be further investigated in order to clarify their relationships and relative significance.
AB  - U periodu 2009-2011. godine na lokalitetima Bački Petrovac i Čantavir prikupljeno je i analizirano 16 uzoraka zaraženog semena gajenog sirka (Sorghum bicolor (L.) Moench.) sorti Alba, Gold, Prima i Reform na prisustvo fitopatogenih gljiva. U pojedinačnim i mešanim zarazama ustanovljeno je prisustvo vrsta iz rodova Epicoccum, Fusarium, Alternaria, Aspergillus i Penicillium. AB  - Cucurbit species grown in the Vojvodina Province, Serbia, were surveyed for the incidence of Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), Cucumber mosaic virus (CMV), Squash mosaic virus (SqMV), Papaya ringspot virus (PRSV) and Tobacco ringspot virus (TRSV) from 2007 to 2009. Samples from more than 700 pumpkin, squash and bottle gourd plants with virus-like symptoms were analyzed by double-antibody sandwich (DAS)-ELISA. ZYMV, WMV and CMV were detected in 79.2, 32.2, and 12.8% of tested samples, respectively. WMV was prevalent in 2007 and ZYMV in 2008-09. Mixed infections were the most frequent type in 2007-08 in contrast to 2009 when single infection of ZYMV prevailed. ZYMV was the most widespread being found in 33 out of 39 inspected fields. Virus species identification was confirmed in selected samples by conventional reverse transcription-polymerase chain reaction (RT-PCR) and sequencing of their coat protein genes. By comparing the obtained virus isolate sequences with those available in GenBank, the identification of serologically detected viruses was confirmed. Phylogenetic analysis based on complete coat protein (CP) sequences highlighted that Serbian ZYMV isolates were closely related to other Central European ZYMV isolates. Finally, additional testing of ELISA-negative samples by RT-PCR using primers specific to six other mosaic viruses revealed the presence of Tomato spotted wilt virus (TSWV) in winter (Cucurbita maxima) and summer (C. pepo 'Beogradska') squash. This is the first report of TSWV natural occurrence on cucurbits in Serbia and on winter squash worldwide.

Presence and molecular characterization of alfalfa mosaic virus on tobacco in Serbia

AB  - Three-year investigation of the presence and distribution of tobacco viruses in Serbia revealed that Alfalfa mosaic virus (AMV) appeared every year with different frequency in tobacco crops. During 2008, the presence of AMV was detected in most of the tested samples (58.82%) and it was the second most common compared to all other viruses which presence was confirmed in Serbia. In 2006 and 2007, AMV was detected in a significantly lower percentage (2.80% and 13.64%, respectively). This study showed that Alfalfa mosaic virus was more commonly found in multiple infections with two, three or even four detected viruses. Single infections were detected only in 2006, in one tobacco field in the locality of Futog. During this investigation, a rapid and simple protocol was optimized and developed for molecular detection of AMV in tobacco leaves, using primers CPAMV1/CPAMV2 and commercially available kits for total RNA extraction as well as for RT-PCR (reverse transcription - polymerase chain reaction). Using RT-PCR and these primers that flank the AMV coat protein gene, a DNA fragment of 751 bp was amplified, sequenced, and compared with the sequences available in GenBank database. The sequence of isolate 196-08 (GenBank Acc. No. FJ527749) proved to be identical at the nucleotide level of 99 to 93% with those from other parts of the world. Phylogenetic analysis of 27 isolates based on 528 bp sequences of the coat protein gene did not show correlation of the isolates with their geographic origin or plant host and showed that these isolates fall into four molecular groups of strains. Serbian AMV isolate from tobacco belongs to group IV, the group that includes most of the isolates selected for phylogenetic analysis.

Plasmopara obducens: A new threat to the production of Impatiens Walleriana in Serbia

AB  - During 2010, Impatiens walleriana plants with symptoms of downy mildew were collected in a greenhouse in the vicinity of Mionica, Kolubara District. Disease incidence was extremely high, approaching 100%, and wilting and collapse of affected plants was very rapid, resulting in losses of more than 90%. White downy growth produced on the lower leaf surface consisted of hyaline, thin-walled sporangiophores with monopodial branching and numerous, ovoid and hyaline sporangia. Apical branchlets of sporangiophores were at right angles to the main axis, with no apical thickening. Pathogenicity tests included inoculation of young I. walleriana plants by spraying with a sporangial suspension, and downy mildew symptoms were observed after 13 to 15 days. The absence of well-defined spots on the infected impatiens leaves and straight sporangiophores indicated that the pathogen was P. obducens, which was further supported by molecular identification, the 5'-end of the nuclear DNA coding for the large ribosomal subunit (LSU rDNA) was amplified by PCR, using primers NL1 and NL4. A representative isolate, 28-10, was sequenced and phylogenetic analysis showed its grouping with other P. obducens isolates of different origin. Considering that impatiens downy mildew in Serbia is proved to be caused by P. obducens it is necessary to employ adequate phytosanitary measures to prevent further spread of the pathogen.
AB  - Tokom 2010. godine biljke Impatiens walleriana sa simptomima plamenjače sakupljene su u stakleniku u okolini Mionice, Kolubarski okrug. Učestalost oboljenja bila je izuzetno visoka, blizu 100%, a sušenje i propadanje zaraženih biljaka veoma brzo, što je dovelo do šteta većih od 90%. Bela prevlaka patogena koja se razvijala na naličju listova sastojala se od hijalinskih monopodijalno razgranatih sporangiofora sa tankim zidovima i brojnih, ovoidnih i hijalinskih sporangija. Vrhovi grana sporangiofora granali su se pod pravim uglom u odnosu na glavnu osu, bez vršnog zadebljanja. Testovi patogenosti uključili su inokulacije mladih biljaka I. walleriana prskanjem suspenzijom sporangija, a simptomi plamenjače razvili su se nakon 13-15 dana. Odsustvo definisanih pega na zaraženim listovima impatiensa i prisustvo pravih sporangiofora ukazalo je da je prouzrokovač P. obducens, što je dalje potvrđeno molekularnom identifikacijom. Primenom lančane reakcije polimeraze (polymerase chain reaction, PCR), pomoću prajmera NL1 i NL4, umnožen je kodirajući 5'-kraj DNA velike ribozomalne podjedinice (LSU rDNA). Amplifikovani produkt odabranog izolata 28-10 je sekvencioniran, a filogenetske analize pokazale su njegovo grupisanje sa ostalim izolatima P. obducens različitog porekla. Uzimajući u obzir da je ovim istraživanjima dokazano da je prouzrokovač plamenjače na Impatiens walleriana u Srbiji P. obducens, neophodno je preduzimanje odgovarajućih fitosanitarnih mera u cilju sprečavanja daljeg širenja patogena u našoj zemlji.

Optimal timing of fungicide applications for the control of Podosphaera leucotricha, the causal agent of peach rusty spot in Serbia

AB  - Peach rusty spot has regularly occurred in Serbia in recent years, causing significant economic losses in the production of late maturing cultivars (Summerset, Suncrest, Fayette and O'Henry). Concerns regarding the etiology of this disease have been solved using molecular identification and cross-pathogenicity tests, which have proved that peach rusty spot is caused by Podosphaera leucotricha. The usual recommended strategy for its control, by multiple treatments starting from petal fall phenophase, has not proved satisfactory in agro-ecological conditions in Serbia. After testing the fungicide efficacy, a three-year study was carried out to determine the precise treatment terms and make the control of P. leucotricha more efficient. The investigation was conducted in the conditions of natural infection in a Summerset cultivar orchard at the locality of Bela Crkva, during the period 2006-2008. Nine different variants of both single and mixed treatments were tested, covering six defined peach growth phenophases - from green top, until the phenophase of second fruit falling. In the conditions of high levels of natural infection in the control plots, in all three investigation years the most efficient was the variant in which kresoxim-methyl was applied three times: in green top, pink bud and full bloom phenophases (91.69 - 91.92%), followed by the variant of treatments in the phenophases of rose bud, full bloom and petal fall (86.3-87.87%) and the variant of two treatments, performed in the phenophases of pink bud and full bloom (79.3-83.09%). The other tested variants showed significantly lower efficiency. The results obtained could significantly contribute to better peach fruit production, without losses caused by peach rusty spot which is commercially very important disease in Serbia.
AB  - Rđasta mrežavost plodova breskve poslednjih godina u Srbiji redovno se javlja i u proizvodnji poznih sorti (Summerset, Suncrest, Fayette i O'Henry) nanoseći značajne ekonomske gubitke. Nedoumice u pogledu etiologije ove bolesti razrešene su na osnovu molekularne identifikacije i unakrsnih testova patogenosti čime je dokazano da rđastu mrežavost breskve izaziva Podosphaera leucotricha. Strategija kontrole P. leucotricha primenjena u svetu, po kojoj suzbijanje treba sprovoditi u višekratnim tretmanima od fenofaze precvetavanja, nije dala zadovoljavajuće rezultate u agroekološkim uslovima Srbije. Nakon izvršenih ispitivanja efikasnosti fungicida, obavljeno je trogodišnje istraživanje u cilju određivanja preciznih rokova tretiranja breskve radi uspostavljanja efikasne kontrole P. leucotricha. Ispitivanje je obavljeno u periodu od 2006. do 2008. godine, u zasadu breskve sorte Summerset, u uslovima prirodne zaraze na lokalitetu Bela Crkva. Testirano je devet različitih varijanti, koje su se sastojale iz kombinovanih i pojedinačnih tretiranja, tako da je pokriven period od šest definisanih fenofaza razvoja breskve, počevši od fenofaze zeleni vrh do fenofaze drugo opadanje plodova. AB  - In a survey to determine the presence of Phytophthora ramorum in Serbia, ornamentals from garden centers, nurseries, and private and public gardens, as well as imported plant material, were inspected. In total, 577 plant, soil, and potting media samples were tested using various detection methods: lateral flow diagnostic test, enzyme-linked immunosorbent assay, conventional polymerase chain reaction, and isolation, followed by identification based on growth characteristics in culture and morphological features. P. ramorum was not detected in any of the 162 soil or potting media tested by the baiting method. P. ramorum was detected in 12 Rhododendron samples from one private garden in Zemun (City of Belgrade District) exhibiting symptoms of leaf necrosis and blight and petiole necrosis, and in three samples of Pieris spp. from one garden center exhibiting symptoms of leaf necrosis. Eight Phytophthora isolates were obtained from the positive Rhododendron plants and three isolates from Pieris plants, and all were identified as P. ramorum on the basis of their uniform morphological and growth characteristics. P. ramorum conformation was also made by sequencing of the internal transcribed spacer regions for a single isolate taken from one infected rhododendron and one pieris plant. Serbian isolates were determined as A1 mating type, due to formation of a few typical sexual structures when crossed with the A2 mating type of P. cinnamomi and P. cryptogea. Pathogenicity test on non-wounded detached leaves of 19 popular ornamentals, as well as the most frequently imported ones, revealed that 10 host species were susceptible, including Robinia pseudoacacia, which is widely distributed in Serbia. During this study, Cotoneaster horizontalis and C. dammeri were determined to be new experimental hosts of P ramorum. This article provides evidence of P. ramorum introduction into Serbia. Although P. ramorum has not been detected in Serbian production nurseries, its presence outdoors might cause severe damages on susceptible common urban plants in public green and natural ecosystems.

Incidence and Distribution of Iris yellow spot virus on Onion in Serbia

AB  - In a survey to determine the presence and distribution of It-is yellows, spot virus (IYSV) in greenhouse ornamentals and onion field crops in 14 districts of Serbia as well as on imported ornamental plants, 1,574 samples were collected and analyzed by double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA). IYSV was not detected in nearly 1,200 plant samples collected from 39 genera of ornamentals grown in greenhouses in Serbia or imported from other countries during 2005 to 2007. The virus was detected in samples from an onion seed crop in the Sirig locality (South Backa District) that showed symptoms resembling those caused by IYSV and in samples without IYSV-Iike symptoms from an onion bulb crop in the Obrenovac locality (City of Belgrade District). Mechanical transmission of IYSV isolates was difficult, and only the isolate 605-SRB could infect four plant species, but not in all replications. No virus transmission could be demonstrated in 5,000 tested seeds originating from IYSV-infected onion crops. For further confirmation of IYSV, the nucleotide sequence of its nucleocapsid (NC) gene was obtained by reverse transcription-polymerase chain reaction (RT-PCR) in symptomatic onion samples as well as in symptomless leaves of Nicotiana benthamiana. Four previously developed primers were tested to determine their suitability for routine detection of Serbian IYSV isolates. Phylogenetic analysis showed clustering of isolates 605-SRB and 622-SRB from the onion seed crop and isolate 283-SRB front the onion bulb crop into two distant clades. The analysis indicated that Serbian isolates of IYSV do not share a recent common ancestor and that they represent two distinct lineages of IYSV in Serbia. Considering that onion is one of the most important and traditionally grown vegetable crops in Serbia, IYSV represents a potentially devastating pathogen in this country.
