Epidemiology studies of Pseudomonas syringae pathovars associated with bacterial canker on the sweet cherry in Serbia

AB  - This study was conducted to examine the epidemiological characteristics of two Pseudomonas syringae pvs. – syringae (Pss) and morsprunorum race 1 (Psm) on the sweet cherry in both field and laboratory conditions. Cycled inoculations of  one-year-old branches indicated that  the sweet cherry became sensitive to Psm race 1 earlier in  the season (October) compared to Pss (November). The most severe infections occurred in the dormancy period (November), while the necroses formed in January and March were less in their length. Inoculations of the two-/three-year-old branches performed in November resulted in necrosis, with those induced by Pss being more aggressive. The January inoculations, however, resulted in less formed necroses in length. Laboratory tests performed on excised sweet cherry branches confirmed the results yielded by the field experiments, indicating that the dormancy period poses the greatest risk for the P. syringae pvs. infection. The isolation of Pss and Psm bacteria from naturally infected sweet cherry samples (cankers) during the summer indicated that the bacteria that persists in cankers in low numbers became active during autumn. Significant Pss and Psm race 1 epiphytic bacterial populations were noted during the spring, but decreased in the summer and increased again in the autumn, indicating the presence of inoculum reservoirs. The greater understanding of the control strategies aimed at the epidemiological factors should, thus, facilitate better disease outbreak management.

Evaluation of cherry cultivar susceptibility to bacterial canker and leaf spot disease

AB  - In this study, the susceptibility of different sweet (19) and sour cherry (3) cultivars (cvs.) to the bacterial pathogens Pseudomonas syringae pv. syringae and P. s. pv. morsprunorum race 1 causing bacterial canker (sensu stricto) and leaf spot was evaluated. Susceptibility to bacterial canker was examined by artificial inoculations of 1-year-old rooted trees, which revealed susceptibility of all cvs. to both P. syringae pathovars. Susceptibility level differed among cvs.; sweet cherry cvs. Katalin, Linda, Summit, New Star and Bigarreau Burlat were highly susceptible, sour cherry cvs. erdi botermo and sweet cherry Drogan's Yellow, Carmen, Germersdorfer and Noir de Meched were susceptible. Sour cherry cvs. Spanska and ujfehertoi furtos, as well as sweet cherry cv. Rita were found to be less susceptible to bacterial canker. Susceptibility to bacterial leaf spot disease was evaluated on naturally infected (field conditions) and artificially inoculated leaves (laboratory conditions). Severe symptoms of bacterial spot with shot hole appearance were observed in cherry leaves at the beginning of May under field conditions. In all collected leaf samples, presence of P. s. pv. morsprunorum race 1 was determined. Sweet cherries Valery Chkalov, Summit, Bigarreau Burlat and New Star were highly susceptible to this bacterium. No symptoms of bacterial spot were found on sweet cherries Solomary gomboly, Linda, Germersdorfer, Hedelfinger, Katalin, Margit, Carmen, Aida, Sunburst, Alex, Peter, Rita and Kavics and sour cherry erdi botermo. In laboratory conditions, susceptibility of the tested cvs. varied and mostly depended on the inoculation method. Nonetheless, sweet cherry cvs. Carmen and Margit and sour cherry erdi botermo exhibited a certain level of resistance.

AB  - This study presents the results of a multiple-year evaluation (1991-1996; 2005-2007) of susceptibility of more than 100 apple cultivars to Venturia inaequalis, Podosphaera leucotricha and Erwinia amylovora under agroecological conditions existing in Serbia. Some of the most popular cultivars were found highly susceptible to V. inaequalis (Cripps Pink, Mutsu, Gloster 69, Wellspur); while Golden Delicious, Richared, Gala, Čačanska pozna, Čadel and Jonagold were susceptible; Idared, Granny Smith and Jonathan moderately susceptible; Lord Lamburne and London Pepping, as well as several autochthonous cultivars were moderately resistant; and a group of resistant cultivars included Prima, Priscilla, Williams Pride, Dayton, Enterprise, Gold Rush, Golden Orange, many of the Re-cultivars (Germany), as well as Baujade, Selena, Dukát, Produkta, Topaz, some older cvs. (Worcester Pearmain, Merton Worcester, James Grieve, Akane, Astilish, Astrachan Red and Discovery), some Co-op selections (USA) and NS hybrids (Serbia). Regarding powdery mildew, Idared and Jonathan were highly susceptible; Gala, Akane, Jonagold, Priscilla, Mutsu, Čačanska pozna, Golden Delicious, Granny Smith, Čadel and GoldRush were susceptible; Wellspur, Astrachan Red, Richared, Jonadel, Dayton and several autochthonous cultivars were moderately susceptible; Lord Lamburne, Astlisch, Prima, Champagne Reinette, Discovery and many autochthonous cultivars were moderately resistant; while most Re-cvs. (Germany), several cultivars from the Czech Republic, some selections from the USA and UK and most NS hybrids (Serbia) were resistant. Also, some cultivars showed variable susceptibility depending on location (Williams Pride, Gloster 69, Baujade and Produkta). E. amylovora was observed only in 2007 and at relatively low intensity (up to 12% infection). The highest disease severity was observed on cv. Elstar, then Granny Smith, Idared and Jonagored; while the lowest was found on Red Chief and Hapke apple trees.
AB  - Ovaj rad prikazuje rezultate višegodišnjeg proučavanja (1991-1996; 2005-2007) osetljivosti više od 100 sorti jabuke prema patogenima Venturia inaequalis, Podosphaera leucotricha i Erwinia amylovora u agroekološkim uslovima Srbije. Etiology of bacterial canker on young sweet cherry trees in Serbia

AB  - Bacterial canker of sweet cherries caused by Pseudomonas syringae was studied in the Vojvodina Province (region of Serbia) with the aim of better understanding disease etiology and epidemiology. During the last decade, bacterial canker has become a recurrent problem, especially in young plantations characterized by intensive production and lack of effective control measures. Among the 24 isolates collected from two localities (Selenc. a and Gornji Tavankut), classical bacteriology tests, pathogenicity, as well as genetic methods based on PCR for syrB gene and DNA sequencing with four housekeeping genes (MLSA), were performed allowing identification of 10 isolates as P. syringae pv. syringae and 14 isolates as P. syringae pv. morsprunorum race 1. In both pathovars, genetic groups were homogenous, confirming earlier research findings and contributing to the extant knowledge of its geographic distribution.

Characterization of Xanthomonas hortorum pv. pelargonii Isolated from Geranium in Serbia

AB  - Geranium leaves and stems with symptoms of bacterial blight were collected from commercial greenhouses during the last decade in Serbia. In total, 17 isolates with colony morphology typical for the genus Xanthomonas were characterized with pathogenicity, biochemical, serological, and molecular assays. All 17 isolates reacted positive in a polymerase chain reaction (PCR) using XcpM1 and XcpM2 primers specific for Xanthomonas hortorum pv. pelargonii. In pathogenicity tests on Pelargonium zonale (leaf and stem inoculation), all isolates caused typical symptoms on leaves starting 2 days after inoculation as sunken, water-soaked, irregular lesions, and 6 to 8 days after inoculation on stems as necrotic lesions also showing yellow exudate. Symptoms resulted in general wilting of inoculated plants 20 days after inoculation. Selected phenotypic tests indicated that all isolates showed the same results as described for the bacterium X. hortorum pv. pelargonii. Repetitive sequence-based PCR typing using BOX and ERIC revealed that all isolates showed two fingerprinting profiles but (GTG)(5) and REP did not reveal differences. Multilocus sequence typing of partial sequences of rpoD, dnaK, fyuA, and gyrB genes of tested isolates and sequences obtained from GenBank of Xanthomonas pathovar pathotype strains did not reveal genetic variability among the isolates, showing the same gene sequence pattern.

Morphological characteristics of isolates of Colletotrichum spp.: Causal agents of anthracnose

AB  - In the morphological studies were included isolates of Colletotrichum spp. originating from pear, apple and sour cherry fruits, as well as reference strains of C. acutatum (CBS 294.67) and C. gloeosporioides (CBS 516.97). The colony characteristics, appearance, shape and size of conidioma, appressoria, and conidia of isolates of Colletotrichum spp. were studied. The four morphological groups of colonies on PDA were observed, confirming the great phenotypic variability of isolates of Colletotrichum spp. Most isolates from apple and pear fruits (KC-6, KC-9, KC-12, JC-5, JC-7, JC-9), formed the first morphological group. The isolates from sour cherry (VC-3, VC-5, VC-7, VC-9) are separated in the fourth morphological group. The size and shape of conidia from these isolates fit the description of C. gloe­osporioides. The chromogenic isolates from pear (KC-21, KC-23, KC-82) formed the second morphological group, and in the third morphological group is isolate (JC-4) from apple fruit. The morphology of conidia from these isolates fit the description of C. acutatum. During this research were evident that all isolates formed conidioma - acervuli. The tested cultures of Colletotrichum spp. did not form teleomorpf. Because of outstanding variability, most of the studied characteristics cannot be independently applied as taxonomic criteria. Obtai­ned results showed that, for Colletotrichum species classification, at least two taxonomic criteria should be used: morphological features combined with molecular identification.
AB  - U morfološka ispitivanja su uključeni izolati Colletotrichum spp. poreklom sa plodova kruške, jabuke i višnje, kao i referentni sojevi C. acutatum (CBS 294.67) i C. gloeosporioides (CBS 516.97). Proučavanjem njihovih morfoloških karakteristika, dobijeni su podaci o izgledu ispitivanih kolonija, odlikama plodonosnog tela, apresorija, veličini i obliku konidija. Na podlozi KDA moguće je razlikovati četiri morfološke grupe kolonija, što potvrđu­je fenotipsku varijabilnost proučavanih izolata Colletotrichum spp. Većina izolata sa ploda kruške i jabuke (KC-6, KC-9, KC-12, JC-5, JC-7, JC-9) obrazuje prvu morfološku grupu. Izolati sa ploda višnje (VC-3, VC-5, VC-7, VC-9), su izdvojeni u četvrtu morfološku grupu. Veličina i oblik konidija ovih izolata odgovara opisu vrste C. gloeosporioides. AB  - Fennel (Foeniculum vulgare Mill.) is aromatic plant from Apiaceae family. Fennel fruits (Foeniculi fructus) usually contain 2-6% of essential oil, with trans-anethole which is main component with 60-90%. The fruits are widely used in medicine and nutrition, but also in organic agricultural production. In the world and in our country it was found a large number of diseases, including fungal which are more abundant, than bacterial and phytoplasmas on the fennel. It was determined and the appearance of insects and parasitic flower dodder (Cuscuta sp.).
AB  - Komorač (Foeniculum vulgare Mill.) je aromatična biljka iz familije Apiaceae. Plodovi komorača (Foeniculi fructus) obično sadrže 2-6% etarskog ulja, čija je glavna komponenta trans-anetol koji čini 60-90%. Imaju široku upotrebu u medicini i ishrani, ali i u poljoprivrednoj proizvodnji. Na komoraču je u svetu i kod nas zabeležen veliki broj bolesti, među kojima su gljivične najbrojnije, ali se javljaju i bakterioze i fitoplazme. Utvrđena je i pojava insekata i parazitne cvetnice Cuscuta sp.

A method for the rapid detection and identification of halo blight pathogen on common bean

AB  - A diagnostic method based on nested-PCR, followed by ELISA and conventional bacteriology tests, for the rapid and reliable detection of halo blight pathogen Pseudomonas savastanoi pv. phaseolicola (Psp) collected from infected bean leaves and seeds is described. Psp formed white, small and flat colonies on nutrient agar medium, creamy white, flat and circular on Milk-Tween agar medium and light yellow, convex and shiny on modified sucrose peptone agar medium. Eighteen Gram-negative, catalase-positive and oxidase-negative strains were subjected to nested PCR with primers P 5.1/P 3.1 and P 5.2/P 3.2, which directed the amplification of the 450 bp target DNA fragment in all tested strains. According to the results of DAS-and PTA-ELISA with respect to reactivity to specific antibodies, all analyzed strains belonged to Psp bacterium. Pathogenicity was tested on bean pods and cotyledon leaves, on which greasy spots were formed. Psp did not cause hypersensitive reaction on the leaves of tobacco and geranium. Strains produced levan, fluorescent pigment, oxidative metabolism of glucose, did not reduce nitrate, did not produce indole and H2S, did not hydrolyze starch, gelatin and esculin; they produced acid from glucose, mannose, sucrose and glycerol, and did not produce acid from maltose, starch, esculin, dulcite, sorbitol, inositol and erythritol.

According to the results of DAS-and PTA-ELISA with respect to reactivity to specific antibodies, all analyzed strains belonged to Psp bacterium. Pathogenicity was tested on bean pods and cotyledon leaves, on which greasy spots were formed. Psp did not cause hypersensitive reaction on the leaves of tobacco and geranium. Strains produced levan, fluorescent pigment, oxidative metabolism of glucose, did not reduce nitrate, did not produce indole and H2S, did not hydrolyze starch, gelatin and esculin; they produced acid from glucose, mannose, sucrose and glycerol, and did not produce acid from maltose, starch, esculin, dulcite, sorbitol, inositol and erythritol.",

AB  - During 2010, black rot symptoms were observed in oilseed rape plants grown in a commercial plot in Serbia. Ten bacterial isolates obtained from diseased plants, and identified as Xanthomonas campestris pv. campestris (Xcc) based on pathogenicity, physiological and biochemical tests, PTA-ELISA and 16S rDNA sequences analysis, were investigated in detail. Strains were characterized by comparing them by rep-PCR fingerprints using ERIC and (GTG)(5) primers. The 16S rDNA sequences of strains TUr1 and TUr6 were deposited in GenBank under accession Nos. KF057196 and KF057197, respectively. Phylogenetic analysis of the 16S regions showed high similarity level for oilseed rape representative strains and Xcc strains of different origin isolated from kale, cabbage and broccoli.

AB  - Halo blight, caused by Pseudomonas savastanoi pv. phaseolicola (Psp), is considered to be an important bacterial disease on common bean (Phaseolus vulgaris L.) in Serbia. Use of pathogen-free seeds is one of the most effective control measures against this disease. The aim of this study was to evaluate a detection method for Psp on untreated common bean seeds (23 genotypes) from commercial crops grown within Serbia. Detection of this pathogen was made by plating onto the modified sucrose peptone (MSP) and Milk Tween (MT) semi-selective mediums from soaked whole common bean seed. Colonies growing on the MSP medium were light yellow, convex and shiny, whereas on the MT medium, they were creamy white, flat and circular. The pathogenicity of the obtained strains was confirmed by the inoculation of germinated bean seed. The isolates recovered from the seed assay were further confirmed to be Psp by using both Enzyme-linked immunosorbent assay (ELISA) and Polymerase Chain Reaction (Nested-PCR) detection methodologies. The International Seed Testing Association (ISTA) method selected for this work was found to be effective in detecting the presence of Psp in common bean seed. The bacterium Psp was detected in only two of the 23 seed samples analyzed by this method, which shows that the bacterium is not widespread in Serbia.

Identification and Differentiation of Erwinia amylovora Using Fatty Acid Analysis and BIOLOG

AB  - A collection of 41 Erwinia amylovora strains isolated from 8 plant species at 13 locations in Serbia and one location in Montenegro was tested and their relationship was determined based on carbohydrate metabolism and fatty acid composition. Metabolism of carbon compounds was tested using the Biolog GN MicroPlate system. After 24 h of incubation, the resulting metabolic fingerprints were recorded and compared with the Biolog GN database. Fatty acid composition was analyzed by Microbial Identification System and compared with the data from the MIDI database. Our results showed that all strains tested were identified as E. amylovora, with similarity index (SI) between 0.62 and 0.99, using BIOLOG. Based on fatty acid composition, 39 strains were identified as E. amylovora, with SI between 0.61 and 0.92. More than 14 fatty acids were detected, eight of which were present in all strains with more than 1% of the total named peak area. Based on quantitative and qualitative content of fatty acid composition, the isolates were clustered into three groups: alpha, beta and gamma (Euclidian distance  lt = 6). Cluster analysis revealed that groups beta and gamma consists of only strains isolated from north Serbia, whereas all strains isolated from central or south Serbia belong to group alpha.

Evaluation of possibilities of Venturia inaequlis control by ecologically acceptable products

AB  - Possibility of V. inaequalis control on apple variety Idared was evaluated by ecologically acceptable products such as inorganic fungicides and acidified clays. The obtained results were compared with common, chemical protection. Year 2009 was one of the unfavorable for V. inaequalis, and 2010 was extremely favorable for apple scab development. Accordingly, in 2009 intensity of infection on apple leaves and fruits in control variant was low (leaves 15,4-34,9%; fruits 0-23,8%). In 2010 infection intensity on leaves and fruits was high (leaves 81,2%; fruits 100%). During period 2009-2010, in these different environmental conditions, common chemical protection showed high efficacy (leaves 96,2-100%; fruits 97,5-100%). High efficacy level in control of V. inaequalis on leaves of 87,4-99,7% was achieved also in variants in which copper products were applied in low concentration of 0,05%. It should be noted that in the most critical period, in April and May of 2010, the treatments were carried out in 5-6 days interval. Copper products showed differing efficacy on fruits (48,5-90,2%). In 2009 which is considered as one of the unfavorable for V. inaequalis development, product Ulmasud B applied alone on leaves was less efficiacy (51,8-58,6%), than on fruits (87,8%). In the same year, efficacy of this product in combination with sulfur, was better (leaves 71,4-76,6%; fruits 86,6). In 2010, which was especially rainy and favourable for spreading the disease, efficiacy of mention combination of products didn't satisfy (leaves 44,8%; fruits 41,0%). In the system of organic production, during vegetation period, program of apple protection in control of V. inaequalis should be supplemented by use of low concentrations of copper products (0,05%) and by various alternative products of different action mechanisms, such as mineral clays, sulfur products and etc. These researches should be continued.
AB  - Ispitivana je mogućnost suzbijanja V. inaequalis na jabuci (ajdared) ekološki prihvatljivim preparatima (neorganski fungicidi i kisela glina). Dobijeni rezultati su poređeni sa uobičajenom, hemijskom zaštitom. Godina 2009. je bila jedna od najnepovoljnijih za V. inaequalis, a 2010. jedna od najpovoljnijih za širenje ovog patogena na jabuci. Shodno tome, u 2009. godini (tri lokaliteta) intenzitet zaraze u kontroli na listovima i plodovima jabuke bio je nizak (listovi 15,4-34,9%; plodovi 0-23,8%). U 2010. godini intenzitet zaraze na listovima i plodovima je bio visok (listovi 81,2%; plodovi 100%). Pri navedenim različitim uslovima zaraze, efikasnost uobičajene, hemijske zaštite, u obe godine je bila na visokom nivou (listovi 96,2-100%; plodovi 97,5-100%). Visoka efikasnost u suzbijanju V. inaequalis na listovima (87,4-99,7%), postignuta je i u varijantama sa primenom bakarnih preparata u niskoj koncentraciji (0,05%), uz napomenu da su u 2010. godini tretmani u najkritičnijem periodu izvođeni u intervalu 5-6 dana. Efikasnost bakarnih preparata na plodovima je bila različita (48,5-90,2%). U 2009. godini, koja je bila jedna od nepovoljnijih za širenje crne pegavosti na jabuci, preparat Ulmasud B primenjen samostalno, ispoljio je znatno slabiju efikasnost na listovima (51,9-58,6%), u odnosu na plodove (87,8%). Iste godine, ovaj preparat u kombinaciji sa sumpornim preparatima, obezbedio je bolju efikasnost (listovi 71,4-76,6%; plodovi 86,6%). Suprotno tome, u 2010. godini, koja je bila izrazito povoljna za širenje ovog patogena, efikasnost navedene kombinacije preparata nije bila zadovoljavajuća (listovi 44,8%; plodovi 41,0%). U sistemu organske proizvodnje, program zaštite jabuke od V. inaequalis tokom vegetacije treba dopuniti korišćenjem preparata na bazi bakra (u niskoj koncentraciji) i raznih alternativnih preparata (na bazi mineralnih glina, sumpornih preparata i sl.). Ova ispitivanja bi trebalo nastaviti.

abstract = "Possibility of V. inaequalis control on apple variety Idared was evaluated by ecologically acceptable products such as inorganic fungicides and acidified clays. The obtained results were compared with common, chemical protection. Year 2009 was one of the unfavorable for V. inaequalis, and 2010 was extremely favorable for apple scab development. Accordingly, in 2009 intensity of infection on apple leaves and fruits in control variant was low (leaves 15,4-34,9%; fruits 0-23,8%). In 2010 infection intensity on leaves and fruits was high (leaves 81,2%; fruits 100%). During period 2009-2010, in these different environmental conditions, common chemical protection showed high efficacy (leaves 96,2-100%; fruits 97,5-100%). High efficacy level in control of V. inaequalis on leaves of 87,4-99,7% was achieved also in variants in which copper products were applied in low concentration of 0,05%. It should be noted that in the most critical period, in April and May of 2010, the treatments were carried out in 5-6 days interval. Copper products showed differing efficacy on fruits (48,5-90,2%). In 2009 which is considered as one of the unfavorable for V. inaequalis development, product Ulmasud B applied alone on leaves was less efficiacy (51,8-58,6%), than on fruits (87,8%). In the same year, efficacy of this product in combination with sulfur, was better (leaves 71,4-76,6%; fruits 86,6). In 2010, which was especially rainy and favourable for spreading the disease, efficiacy of mention combination of products didn't satisfy (leaves 44,8%; fruits 41,0%). In the system of organic production, during vegetation period, program of apple protection in control of V. inaequalis should be supplemented by use of low concentrations of copper products (0,05%) and by various alternative products of different action mechanisms, such as mineral clays, sulfur products and etc. These researches should be continued., Ispitivana je mogućnost suzbijanja V. inaequalis na jabuci (ajdared) ekološki prihvatljivim preparatima (neorganski fungicidi i kisela glina). Dobijeni rezultati su poređeni sa uobičajenom, hemijskom zaštitom. Godina 2009. je bila jedna od najnepovoljnijih za V. inaequalis, a 2010. jedna od najpovoljnijih za širenje ovog patogena na jabuci. Shodno tome, u 2009. godini (tri lokaliteta) intenzitet zaraze u kontroli na listovima i plodovima jabuke bio je nizak (listovi 15,4-34,9%; plodovi 0-23,8%). U 2010. godini intenzitet zaraze na listovima i plodovima je bio visok (listovi 81,2%; plodovi 100%). Pri navedenim različitim uslovima zaraze, efikasnost uobičajene, hemijske zaštite, u obe godine je bila na visokom nivou (listovi 96,2-100%; plodovi 97,5-100%). Visoka efikasnost u suzbijanju V. inaequalis na listovima (87,4-99,7%), postignuta je i u varijantama sa primenom bakarnih preparata u niskoj koncentraciji (0,05%), uz napomenu da su u 2010. godini tretmani u najkritičnijem periodu izvođeni u intervalu 5-6 dana. Efikasnost bakarnih preparata na plodovima je bila različita (48,5-90,2%). U 2009. godini, koja je bila jedna od nepovoljnijih za širenje crne pegavosti na jabuci, preparat Ulmasud B primenjen samostalno, ispoljio je znatno slabiju efikasnost na listovima (51,9-58,6%), u odnosu na plodove (87,8%). Iste godine, ovaj preparat u kombinaciji sa sumpornim preparatima, obezbedio je bolju efikasnost (listovi 71,4-76,6%; plodovi 86,6%). Suprotno tome, u 2010. godini, koja je bila izrazito povoljna za širenje ovog patogena, efikasnost navedene kombinacije preparata nije bila zadovoljavajuća (listovi 44,8%; plodovi 41,0%). U sistemu organske proizvodnje, program zaštite jabuke od V. inaequalis tokom vegetacije treba dopuniti korišćenjem preparata na bazi bakra (u niskoj koncentraciji) i raznih alternativnih preparata (na bazi mineralnih glina, sumpornih preparata i sl.). Ova ispitivanja bi trebalo nastaviti.",

The two Streptomyces species exhibited the strongest antagonism against isolates of C. acutatum and C. gloeosporioides. Microscopic examination showed that the most common mode of action was antibiosis. The results of this study identify T. harzianum, G. roseum, B. subtilis, S. natalensis and S. noursei as promising biological control agents for further testing against anthracnose disease in fruits.
AB  - Antagonističko delovanje pet agenasa biološke zaštite: Trichoderma harzianum, Gliocladium roseum, Bacillus subtilis, Streptomyces noursei i Streptomyces natalensis, testirano je in vitro na Colletotrichum acutatum i Colletotrichum gloeosporioides, prouzrokovače antraknoze plodova. Svi mikrobni antagonisti inhibiraju porast micelije u testovima dvojne kultivacije i klijavost konidija Colletotrichum izolata. Dve vrste roda Streptomyces ispoljavaju najjači stepen antagonizma prema izolatima C. acutatum i C. gloeosporioides. Mikroskopsko ispitivanje pokazuje da je antibioza najčešći način delovanja antagonista. T. harzianum, G. roseum, B. subtilis, S. natalensis i S. noursei su rezultatima ovih istraživanja identifikovani kao biološki agensi koji se mogu uspešno uključiti u buduća testiranja u cilju suzbijanja antraknoze plodova.

Morphological and molecular identification of Colletotrichum acutatum from tomato fruit

AB  - Colletotrichum gloeosporioides, Colletotrichum acutatum, Colletotrichum coccodes, and Colletotrichum dematium are the four main species of Colletotrichum that cause tomato anthracnose. In Serbia, the occurrence of anthracnose on tomato fruit has been recorded during the last several years. Typical fruit symptoms include dark, sunken, and circular lesion with orange conidial masses. Pathogen isolates were obtained from a diseased tomato fruits, on PDA medium forming a white to gray colonies. The cultures developed black acervuli around the center of the colony. Conidia were hyaline, aseptate, and fusiform or rarely cylindrical. Appressoria were smooth, simple, clavate to ovate, and varied from light to dark brown. Pathogenicity tests with representative isolates were conducted on symptomless, detached tomato fruits. All tested isolates caused anthracnose lesions on tomato fruit after 7 days of incubation. Koch's postulates were fulfilled by reisolation from inoculated tomato fruits. PCR analysis (using species-specific primer pair, CaInt2/ ITS4) of genomic DNA from tomato isolates resulted in an amplification product of 490 bp, specific for C. acutatum, further confirming the identity of the pathogen. Based on morphological and molecular characteristics, the isolates from tomato fruit were determined as C. acutatum.
AB  - Četiri glavna prouzrokovača antraknoze paradajza iz roda Colletotrichum su: Colletotrichum gloeosporioides, Colletotrichum acutatum, Colletotrichum coccodes i Colletotrichum dematium. U Srbiji je tokom poslednjih godina zabeležena pojava antraknoze ploda paradajza. Tipični simptomi na plodu su tamne, ulegnute, kružne lezije sa masom narandžastih konidija. Sa obolelih plodova paradajza dobijeni su izolati patogena koji na KDA podlozi obrazuju bele do sivo obojene kolonije. Acervuli crne boje formiraju se u kulturi oko centra kolonije. Konidije su hialinske, neseptirane, fusiformne ili ređe cilindrične. Apresorije su glatke, jednostavne, okruglastog ili oblika izdužene palice, svetlo do tamno braon boje. Test patogenosti je obavljen sa reprezentativnim izolatima, na odabranim, zdravim plodovima. Svi ispitivani izolati prouzrokuju antraknozne lezije na plodu paradajza 7 dana nakon inokulacije. Detection and identification of Xanthomonas axonopodis pv. phaseoli on bean seed collected in Serbia

AB  - Laboratory assays were conducted to detect and identify Xanthomonas axonopodis pv. phaseoli on bean seeds collected from naturally contaminated commercial crops. Semi-selective media, viz. Xanthomonas campestris pv. phaseoli agar and milk tween agar were used to detect the bacterium from whole bean seed extract. Colonies of the bacterium were yellow, mucoid and convex on these media and zones of hydrolysis formed around them. The pathogenicity of the strains was confirmed by stem inoculation of young bean plants. Identification of the strains was proved by using rapid methods (ELISA and PCR). The results showed the presence of seed-borne X. axonopodis pv. phaseoli in 20 out of the 23 assayed bean cultivars that are commercially grown in Serbia. These results indicate that the seed of the bean cultivars grown in our country is highly infected by the bacterium.

Distribution and characterization of phytopathogenic bacteria on commercial bean crop in Vojvodina

AB  - The present paper discusses the occurrence and severity of bacterial bean diseases in the Serbian province of Vojvodina. Leaf samples of diseased bean plants were collected from a number of locations in the province. All the samples from all the locations tested positive for Xanthomonas axonopodis pv. phaseoli, indicating that the bacterium is widespread on commercial plots and gardens planted to bean in Vojvodina. The bacterium Pseudomonas savastanoi pv. phaseolicola was isolated from a smaller number of samples, which shows that it is not widespread on bean in the province. Identification of the isolates was confirmed using the classic phytobacterial methods (for the pathogenic, morphological, breeding and biochemical-physiological characteristics of the isolates) as well as the rapid modern methods (ELISA for serological and PCR for molecular properties).
AB  - U radu su proučavane bakterioze pasulja, njihovo prisustvo i intenzitet pojave na teritoriji Vojvodine. Izolacijama iz prikupljenih obolelih uzoraka pasulja (list) na standardne hranljive podloge, dobijene su kolonije Xanthomonas axonopodis pv. phaseoli iz svih ispitivanih uzoraka i lokaliteta, što ukazuje na dominantno prisustvo ove bakterije na proizvodnim parcelama i okućnicama pasulja kod nas. Bakterija Pseudomonas savastanoi pv. phaseolicola je izolovana kod manjeg broja uzoraka, što pokazuje da nije široko rasprostranjena u našim uslovima gajenja pasulja. Identifikacija izolata ovih bakterija je dokazana korišćenjem klasičnih fitobakterioloških (patogene, morfološke, odgajivačke i biohemijsko-fiziološke odlike izolata) i brzih, savremenih metoda (serološke-ELISA i molekularne-PCR).

Rezultati dobijeni tokom rada pokazuju da su poluselektivne podloge MT (Milk Tween Agar) i MSP (Modified Sucrose Peptone Agar) pogodne za izolaciju ove bakterije. Patogenost odabranih izolata dokazana je na kotiledonim listovima pasulja. Primenom ELISA testova i PCR, potvrđeno je da svi ispitivani izolati i reizolati pripadaju bakteriji P.s.pv. phaseolicola.

AB  - Bacterial blight is a disease of soybean appearing regularly with high intensity in Serbia. Isolation of pathogen was done using beef extract (MPA) and the medium enriched with sucrose (NSA) from infected soybean leaves of several varieties grown in Vojvodina province, during 2005.
Only the representative isolates were chosen for analyses. Pathogenicity of the isolates including the reference strain (National Collection of Plant Pathogenic Bacteria, United Kingdom-NCPPB 3318) was determined by inoculating soybean plants at cotyledon stage (cv. Balkan), by dipping and spraying with bacterial suspensions (conc. 108 CFU/ml). Pathogenicity of isolates was also confirmed on tobacco plants (HR positive reaction).
Cultural, biochemical and physiological characteristics were tested according to methods cited by Lelliot and Stead (1987), Klement et al. (1990), Schaad (1980). The colonies are chosen by fluorescence on KB agar, and by the LOPAT tests (levan production, oxidase reaction, potato rot, arginine dihydrolase production and tobacco hypersensitivity: +, −, −, −, +). The studied isolates of bacterium did not hydrolyze starch, gelatine and aesculin. They did not reduce nitrate but metabolized glucose oxidatively and were catalase positive. All the isolates produced acid from glucose, galactose, xylose, arabinose, manose, sucrose, rafinose and inositol.
Bacterium identification was confirmed by a serological method (agglutination tests by Express Kit – NEOGEN Europe Ltd., Scotland, UK), with appropriate antibodies of Pseudomonas syringae pv. glycinea. The studied isolates formed whitish sediment in the case of positive reaction.
Differential soybean sortiment: Acme, Chippewa, Flambeau, Harosoy, Lindarin, Merit and Norchief was used to determine which physiological race the isolates belonged to. Young plants were inoculated in two ways: by rubbing leaves with sterile cotton swab dipped in an aqueous bacterial suspension (Alvarez et al., 1995), and by spraying leaves under pressure (Cross et al., 1966; Balaž et al., 1990; Prom and Venette, 1997).
Based on the results of these studies we concluded that strains which cause blight of soybeans obtained from Vojvodina, belong to Pseudomonas syringae pv. glycinea, race 4.

AB  - Ekološki najprihvatljiviji put ostvarenja ciljeva integralne proizvodnje voća je stvaranje i uvođenje u proizvodnju novih sorti koje imaju genetsku otpornost prema patogenima. To znači da biljka produktima svoga metabolizma sprečava patogena da izvrši infekciju i da se uspešno razvija. Takav pristup zaštitu biljaka hemijskim putem svodi na vrlo malu meru, proizvodnja postaje jeftinija, hranidbena vrednost plodova visoka a životna sredina čistiji. Predmet istraživanja su kontinentalne voćne vrste: jabuka, breskva, kajsija, orah, leska i vinova loza.

AB  - Twelve isolates of Phomopsis sp. were obtained from the branches and the trunk of plums (Prunus domestica L) with decay symptoms in Valjevo, Ljig Koceljeva and Ub vicinity during 2004-2006. Morphological, pathogenic and growing characteristics were studied. Pathogen caused tissue necrosis of branches around the inoculate seats, and wrinkling and watering of plum fruits. All media were suitable for pathogen development, except prune agar. The best growth of isolates was at medium pH 5,5. The optimal temperature for growth and germination of pycnidiospores was 25°C.
AB  - Dvanaest izolata Phomopsis sp. dobijeno je sa grana i stabla šljive (Prunus domestica L) sa simptomima propadanja u okolini Valjeva, Ljiga, Koceljeve i Uba tokom 2004-2006. godine. Proučene su morfološke, patogene i odgajivačke odlike ovih izolata. Prvi simptomi uočavaju se u vreme cvetanja i listanja šljive ukoliko se infekcija desila u blizini pupoljaka i osnove lisnih peteljki. U tom slučaju listovi su sitniji i hlorotični. Kasnije i listovi i cvetovi nekrotiraju i suše se. Tek formirani mladi izdanci oboljevaju postaju nekrotični i propadaju. Na granama i stablu formiraju se ulegnute nekrotične pege, koje se brzo šire i u okviru kojih kora uzdužno puca, tako da se formiraju rak-rane. Nekroza se širi na drvenasti deo, gde bivaju zahvaćeni sprovodni sudovi, što dovodi do uvenuća i izumiranja grana. Patogen se dobro razvija na svim podlogama, osim na podlozi od suvih šljiva. Podloge čija je pH 5,5 pokazale su se kao najbolje za porast patogena. Temperature od 25°C su najpovoljnije za razvoj patogena i klijavost konidija.
