Leaf spot disease on Philodendron scandens, Ficus carica and Actinidia deliciosa caused by Pseudomonas syringae pv. syringae in Serbia

AB  - During the spring of 2014 and 2015, a leaf spot disease was observed on heart-leaf philodendron (Philodendron scandens Koch & Sello) plants grown in a several house gardens near city of Jagodina, Moraviki region of Serbia. Diseased leaves were covered with watersoaked leaf spots with surrounded by yellow halos. Based on biochemical and molecular tests Pseudomonas syringae pv. syringae was identified as causing agent of bacterial leaf spot of heart-leaf philodendron and is in humid season accountable for appearence of disease. The genetic diversity within this pathogen isolated from different hosts in central Serbia, was studied to obtain insights into disease etiology. Therefore, kiwi fruit and fig tree leaves, with disease symptoms that were similar to those observed on heart-leaf philodendron plants, were collected and bacteria were isolated on NAS medium. Isolates were identified as P. syringae pv. syringae by detection of syrB gene, biochemical, pathogenicity tests and sequencing of gyrB gene. In cross inoculation tests on potted host plants, each strain induced leaf spot symptoms on heart-leaf philodendron and fig tree leaves, apart of the host of origin, and no significant symptoms in kiwi fruit leaves. Genomic profiles generated with rep-PCR analysis revealed that all strains originated from diseased heart-leaf philodendron, kiwi fruit and fig tree leaves had similiar DNA fingerprints. Overall, the data demonstrate P. syringae pv. syringae as a causing agent of leaf spot disease of heart-leaf philodendron and fig tree in Moraviki region of Serbia. This research is the first study that identified P. syringae pv. syringae associated with leaf spot disease on P. scandens, and also a starting point for investigation of the epidemiology of leaf spot disease on fruit trees that could be infected by the same bacterial strains.

Morpho-physiological study of Alternaria spp. isolates from celery

AB  - Genus Alternaria represents one of the most important pathogens of Apiaceae family. Alternariose symptoms may develop on foliage causing leaf spot, necrosis and eventually drying and black root. Celery plants with characteristic symptoms were collected during 2012 and 2013 in Serbia. The preliminary identification was done by analysis of morphological characteristics of conidia. After identification cultural characteristics, growth rate and spore morphology were examined and compared based on influence of ecological factors such as nutrient medium, temperature, pH value of the medium and light quality.
AB  - Vrste roda Alternaria predstavljaju jedne od najznačajnijih gljiva patogena familije Apiaceae. Promene koje izazivaju na biljkama domaćinima su mrka trulež korena, pegavost lišća, sušenje listova i lisnih preteljki. Tokom 2012. i 2013. godine na teritoriji Srbije, prikupljeni su uzorci celera sa karakterističnim simptomima pegavosti lišća i sušenja listova. Identifikacija izolata je izvršena analizom morfoloških karakteristika spora gljive. Nakon identifikacije izvršena su uporedna proučavanja uticaja različitih hranljivih podloga, temperatura, pH vrednosti podloga i kvaliteta svetlosti na morfološke karakteristike izolata, porast kolonija i morfologiju spora.

Antibacterial activities of some Bacillus spp. and Trichoderma harzianum against phytopathogenic bacteria

AB  - Biological control is an innovative, cost effective and eco-friendly approach for control of many plant diseases. Bacillus sp. and Trichoderma sp. were known for its mycoparasitic and antagonistic mechanism in the control of wide range of phytopathogenic diseases in many types of crops. This research is based on the antibacterial activities of some Bacillus spp. and Trichoderma harzianum in relation to the 10 selected phytopathogenic bacteria in vitro study. The results indicated that among 52 strains of Bacillus spp. presented against phytopathogenic bacteria, only 6 of them showed inhibition but only against Xanthomonas genera. Other Bacillus spp. strains weren't effective against tested phytopathogenic bacteria. Results of testing of T. harzianum efficacy against the growth of selected phytopathogenic bacteria showed that this fungi could be effective to strains from genera Pseudomonas and Xanthomonas but for other tested genera was ineffective. In further work, all of this trial need to be supported by evaluation of antimicrobial activity in vivo.
AB  - Biološka kontrola predstavlja inovativni, isplativ, ekološki pristup za suzbijanje mnogih biljnih bolesti. Vrste roda Bacillus i Trichoderma su poznati po svom mikoparazitskom i antagonističkom mehanizmu suzbijanja širokog spektra fitopatogenih prouzrokovača bolesti kod brojnih poljoprivrednih kultura. U ovom radu proučavano je baktericidno delovanje nekih vrsta roda Bacillus, kao i gljive Trichoderma harzianum na 10 odabranih fitopatogenih bakterija u in vitro uslovima. Prema dobijenim rezultatima, od ukupno 52 testirana izolata iz roda Bacillus, samo 6 je inhibiralo porast fitopatogenih bakterija i to samo iz roda Xanthomonas. Rezultati ispitivanja efikasnosti gljive T. harzianum su ukazali na mogućnost delovanja ove gljive na bakterije iz roda Pseudomonas i Xanthomonas, ali ne i na druge rodove fitopatogenih bakterija testirane u ovom radu. Dalja istraživanja treba bazirati na proučavanjima antimikrobne aktivnosti testiranih agenasa prema fitopatogenim bakterijama u in vivo ogledima.
