Grapevine rupestris stem pitting virus: a new pathogen of grapevine in Serbia

AB  - Grapevine rupestris stem pitting-associated virus (GRSPaV), a member of the genus Foveavirus, is a commonly detected grapevine virus around the world. Despite the broad and global distribution of GRSPaV, no information on its presence in Serbia had been available before this study. In October 2018, 3-year-old grapevine plants of ‘Cabernet Sauvignon’ showing reduced growth and leaf reddening were observed in the vicinity of Krnjevo locality. The results of an mRT-PCR test showed that only samples prepared from cortical scrapings of mature symptomatic grapevine canes yielded an amplicon of 155 bp that confirmed the presence of GRSPaV. No amplification products were recorded in samples prepared from basal leaves or petioles. A serological assay confirmed the presence of GRSPaV in all tested mature cane samples. For further analyses, the CP (coat protein) genes of two selected isolates (VL8 and VL15) of naturally infected grapevine canes were partially sequenced and six sequence variants were determined, showing a certain variability among them. The constructed phylogenetic tree revealed the grouping of Serbian sequence variants into separate subgroups within the same phylogenetic lineage. This is the first report of GRSPaV present in Serbia. Further monitoring and increased phytosanitary control measures are needed to prevent potential spreading and introduction of new virus variants.

AB  - Aim of study: To report the occurrence of Pepino mosaic virus (PepMV) on tomato in Serbia and to genetically characterize Serbian PepMV isolates. Area of study: Tomato samples showing virus-like symptoms were collected in the Bogojevce locality (Jablanica District, Serbia). Material and methods: Collected tomato samples were assayed by DAS-ELISA using antisera against eight economically important or quarantine tomato viruses. Three selected isolates of naturally infected tomato plants were mechanically transmitted to tomato ‘Novosadski jabučar’ seedlings. For confirmation of PepMV infection, RT-PCR was performed using specific primers PepMV TGB F/PepMV UTR R. Maximum-likelihood phylogenetic tree was constructed with 47 complete CP gene sequences of PepMV to determine the genetic relationship of Serbian PepMV isolates with those from other parts of the world. Main results: The results of DAS-ELISA indicated the presence of PepMV in all tested samples. Mechanically inoculated ‘Novosadski jabučar’ seedlings expressed yellow spots and light and dark green patches, bubbling, and curled leaves. All tested tomato plants were RTPCR positive for the presence of PepMV. The CP sequence analysis revealed that the Serbian PepMV isolates were completely identical among themselves and shared the highest nucleotide identity of 95.1% (99.2% aa identity) with isolate from Spain (FJ263341). Phylogenetic analysis showed clustering of the Serbian PepMV isolates into CH2 strain, but they formed separate subgroup within CH2 strain. Research highlights: This is the first data of the presence of PepMV in protected tomato production in Serbia. Considering increased incidence and rapid spread in Europe, the presence of PepMV on tomato could therefore represent serious threat to this valuable crop in Serbia

Occurrence and molecular characterization of Impatiens necrotic spot tospovirus in ornamentals in Serbia

AB  - A subset of ornamentals grown in Serbia were surveyed for the presence of impatiens necrotic spot tospovirus (INSV) from 2008 to 2018. Out of 1578 samples tested for the presence of the virus using double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), INSV was detected in 26 Begonia sp. and four Tulipa sp. samples collected in 2008 and in 10 samples of each of Begonia sp., Impatiens walleriana and I. hawkeri 'SunPatiens' and 'New Guinea' collected in 2018. INSV was not detected in any samples collected during 2009-2017. For further confirmation, the nucleotide sequence of the INSV nucleocapsid (N) gene was obtained by reverse transcription polymerase chain reaction (RT-PCR) in two and four samples collected in 2008 and 2018, respectively. The success of mechanical transmission of INSV isolates from different hosts was variable depending on isolates. Serbian INSV isolates showed very low nucleotide diversity and they were closely related to the Chinese and Japanese isolates already reported in the literature, indicating that the Serbian isolates might be of Asian origin. Phylogenetic analysis and the median-joining haplotype network of the N gene sequences showed clustering of Serbian INSV isolates in the Asian clade into two different subclades with different recent common ancestors implying two independent introductions. The sporadic occurrence of INSV with high temporal distance is a likely consequence of virus spread through the international trade in ornamental planting material. Therefore, the establishment of the virus should be considered and prevented when introducing new plant material into the country. This is also the first report of the tulip as a natural host of INSV.

Characterization of Gnomoniopsis idaeicola, the Causal Agent of Canker and Wilting of Blackberry in Serbia

AB  - Blackberry cane diseases with the symptoms of necrosis, canker, and wilting are caused by several fungi worldwide. Surveys conducted from 2013 to 2016 in Serbia revealed the occurrence of Gnomoniopsis idaeicola, the causal agent of cane canker and wilting, which was found to be distributed in almost half of the surveyed orchards, in three blackberry cultivars, and with disease incidence of up to 80%. Wide distribution and high disease incidence suggest that G. idaeicola has been present in Serbia for some time. Out of 427 samples, a total of 65 G. idaeicola isolates were obtained (isolation rate of 34.19%). Reference isolates, originating from different localities, were conventionally and molecularly identified and characterized. G. idaeicola was detected in single and mixed infections with fungi from genera Paraconiothyrium, Colletotrichum, Diaporthe, Botryosphaeria, Botrytis, Septoria, Neofusicoccum, and Discostroma, and no diagnostically specific symptoms could be related directly to the G. idaeicola infection. In orchards solely infected with G. idaeicola, blackberry plant mortality was up to 40%, and yield loses were estimated at 50%. G. idaeicola isolates included in this study demonstrated intraspecies diversity in morphological, biological, pathogenic, and molecular features, which indicates that population in Serbia may be of different origin. This is the first record of a massive outbreak of G. idaeicola infection, illustrating its capability of harmful influence on blackberry production. This study represents the initial step in studying G. idaeicola as a new blackberry pathogen in Serbia, aiming at developing efficient control measures.

AB  - Brojnost virusa infektivnih za vinovu lozu stalno se povećava, tako da je do sada opisano više od 60 vrsta koji mogu da zaraze vinovu lozu. Paralelno sa globalnom ekspanzijom vinogradarske i vinarske industrije zapaženo je povećanje učestalosti i ekonomskog značaja oboljenja uvijenosti lišća vinove loze (Grapevine Leafroll Disease, GLD). GLD, osim što smanjuje vitalnost i dugovečnost vinove loze, značajno smanjuje prinos i kvalitet grožđa. Epidemiologija oboljenja je nedovoljno istražena zbog kompleksnosti ove bolesti, genetički i serološki različitih vrsta unutar kompleksa i različitosti simptoma kod crvenih i belih sorti vinove loze. Кompleks virusa prouzrokovača uvijenosti lišća vinove loze (Grapevine leafrollassociated viruses, GLRaVs) čini pet različitih virusa koji pripadaju familiji Closteroviridae: jedna vrsta koja pripada rodu Closterovirus (GLRaV-2), tri rodu Ampelovirus (GLRaV-1, GLRaV -3, GLRaV -4, i njegovi sojevi: GLRaV-4 soj 5, GLRaV-4 soj 6, GLRaV-4 soj 9, GLRaV-4 soj Pr, GLRaV-4 soj De i GLRaV-4 soj Car) i jedna vrsta iz roda Velarivirus (GLRaV-7). GLRaVs se prenose kalemljenjem i propagativnim materijalom (podloga i vijoka), a neke vrste i štitastim vašima. Virusi koji izazivaju uvijenost lišća se obično javljaju u mešanoj infekciji, a često su prisutni i u mešanoj infekciji sa drugim virusima vinove loze. Ranija istraživanja sprovedena u vinogorjima u Srbiji utvrdila su prisustvo GLRV-1, -2 i -3, pri čemu je GLRaV-3 bio najzastupljeniji. Ukoliko je širenje virusa unutar zasada putem vektora izraženo, krčenje ili uklanjanje zaraženih čokota, kontrola štitastih vaši i adekvatne sanitarne mere mogu redukovati nove infekcije.

AB  - Leek yellow stripe virus (LYSV) is one of the most frequent and important viruses in leek and garlic crops worldwide. In Serbia this virus is found both in leek and garlic, and often at high percentages. During two consecutive years, 2012 and 2013, a total 92 samples were collected from 11 inspected leek-, garlic- and onion-growing locations and they were analyzed for the presence of LYSV using DAS-ELISA. LYSV was detected in 31.5% of the tested samples. In 2012, the presence of LYSV was only detected in leek plants, and in 55.6% of the tested samples. During 2013, LYSV was detected in 85% of leek and 58.3% of garlic samples. In total, LYSV was detected in 56.4% of leek samples and 17.1% garlic samples. LYSV incidence was confirmed using RT-PCR with LYSV specific primers amplifying 1020 bp fragment representing coat protein and part of nuclear inclusion B genes. Molecular identification was confirmed by sequencing of two selected isolates, 181-13 (MG242625) from garlic and 298-13 (MG242624) from leek, and comparing them to the GenBank sequences of LYSV. Phylogenetic analysis of 55 sequences of LYSV from all over the world showed some correlation between host plant and geographical origin of the isolates, forming five separate clades. Two Serbian LYSV isolates fell into distant clades. The Serbian leek isolate 298-13 of LYSV belongs to clade B, while isolate 181-13 originating from garlic belongs in clade E.
AB  - Virus žute prugavosti praziluka (Leek yellow stripe virus, LYSV) je jedan od najčešćih i najznačajnijih virusa na praziluku i belom luku. U Srbiji je virus detektovan na obe kulture, i praziluku i belom luku, a često se javlja u visokim procentima. Tokom dve uzastopne godine (2013. i 2014.) pregledano je 11 lokaliteta gajenja praziluka, belog i crnog luka i sakupljena su 92 uzorka koja su DAS-ELISA metodom testirana na prisustvo LYSV. Prisustvo LYSV je dokazano u 31,5% testiranih uzoraka. Tokom 2012., prisustvo LYSV dokazano je samo u praziluku, u 55,6% testiranih uzoraka. Tokom 2013., LYSV je dokazan u 85% uzoraka praziluka i 58,3% uzoraka belog luka. Ukupno, prisustvo LYSV je dokazano u 56,4% uzoraka praziluka i 17,1% uzoraka belog luka. Filogenetske analize 55 sekvenci izolata LYSV iz različitih delova sveta ukazale su na delimičnu korelaciju između biljke domaćina i geografskog porekla izolata, formiranjem pet odvojenih grupa izolata u stablu. Dva izolata iz Srbije grupisala su se u udaljene grupe. Izolat iz Srbije iz praziluka 298-13 grupisao se u grupu B, dok se izolat iz belog luka 181-13 grupisao u grupu E.

AB  - Although natural infections of potato in Serbia have not been detected, two TSWV isolates from tobacco, with different biological features were compared based on NSm gene sequence analysis and they were used for mechanical and graft inoculations to evaluate the susceptibility of five popular potato cultivars: 'Riviera', 'Arnova', 'Curoda', 'Kondor' and 'Aladin'. Both TSWV isolates infected all tested potato cultivars. Statistical analyses showed that the rate of TSWV transmission from infected foliage to tubers and from infected tubers to progeny plants was affected by cultivars, while virus transmission efficiency was independent of investigated isolates. Different potato tissues used for sample preparation did not influence virus detection by ELISA, but composite sampling from six different tuber parts is recommended since it was the most reliable.

AB  - Proučavanjem pojave i rasprostranjenosti virusa mozaika krastavca u usevu lubenice u Srbiji tokom 2012. godine, prisustvo virusa utvrđeno je u pet od šest pregledanih lokaliteta. Prisustvo virusa ustanovljeno je u 24,05% serološki testiranih uzoraka (DAS-ELISA), a najzastupljeniji je bio na lokalitetu Silbaš. Tokom ovih istraživanja virus mozaika krastavca češće je bio prisutan u mešanim infekcijama sa ZYMV i/ili WMV (12,66%), dok je prisustvo pojedinačne infekcije utvrđeno u nešto nižem procentu, 11,39%. Mehaničkim inokulacijama N. glutinosa, dobijeno je pet izolata, od kojih su dva odabrana za dalju biološku karakterizaciju. Na osnovu tipa i jačine simptoma na test-biljkama, utvrđena je fenotipska varijabilnost ispitivanih izolata. Molekularna detekcija obavljena je amplifikacijom fragmenta dužine 871 bp kod svih ispitivanih izolata korišćenjem para prajmera CMVCPfwd/CPrev, koji omogućavaju umnožavanje gena za protein omotača i deo 5' i 3' UTR regiona. Analizom sekvenci kompletnog gena za protein omotača i rekonstrukcijom filogenetskog stabla, utvrđena je pripadnost izolata CMV iz lubenice iz Srbije podgrupi IA, koja obuhvata većinu izolata odabranih za filogenetsku analizu.

Na lokalitetu Gornji Tavankut, ZYMV je dokazan u 23,08% testiranih biljaka u pojedinačnim infekcijama, a na lokalitetu Silbaš u 35,29% testiranih biljaka u mešanim infekcijama sa virusom mozaika krastavca i virusom mozaika lubenice. ZYMV je uspešno mehanički prenesen sa prirodno zaraženih biljaka lubenice na Cucurbita pepo 'Ezra F1'. Molekularna detekcija obavljena je RT-PCR metodom umnožavanjem dela gena za nuklearne inkluzije, gena za protein omotača i dela 3' neprepisujućeg regiona, čime je potvrđena identifikacija izolata. Filogenetske analize pokazale su grupisanje izolata poreklom iz lubenice sa drugim izolatima iz Srbije i Centralne Evrope u okviru A-I podgrupe. Analize aminokiselinske sekvence N-terminalnog kraja CP gena, takođe su pokazale pripadnost izolata 550-11 centralnoevropskoj grani.

Epicoccum nigrum the new pathogen of sorghum seed in Serbia

AB  - Sixteen samples of sorghum seed (Sorghum bicolor (L.) Moench.) 'Alba', 'Gold', 'Prima' and 'Reform' were analyzed in the localities of Bački Petrovac and Čantavir in the period 2009-2011. Tipresence of species belonging to the genera Epicoccum, Fusarium, Alternaria, Aspergillus and Penicillium was established in single and mixed infections. From the infected sorghum seed, monosporial cultures identified as Epicoccum nigrum based on morphology, proved their pathogenicity on artificially inoculated sorghum seedlings. Molecular identification was performed by PCR and amplification of the ITS region of ribosomal DNA. Gene sequences of selected isolates 291-09 (JQ619838) and 315-09 (JQ619839) exhibited 99-100% nucleotide identity with the sequences of 31 isolates of E. nigrum deposited in the GenBank. It obtained results represent the first detailed characterization of E. nigrum in Serbia. The presence of a large number of phytopathogenic fungi on sorghum seed should be further investigated in order to clarify their relationships and relative significance.
AB  - U periodu 2009-2011. godine na lokalitetima Bački Petrovac i Čantavir prikupljeno je i analizirano 16 uzoraka zaraženog semena gajenog sirka (Sorghum bicolor (L.) Moench.) sorti Alba, Gold, Prima i Reform na prisustvo fitopatogenih gljiva. U pojedinačnim i mešanim zarazama ustanovljeno je prisustvo vrsta iz rodova Epicoccum, Fusarium, Alternaria, Aspergillus i Penicillium. Iz zaraženog semena izolovane su monosporne kulture i na osnovu morfoloških osobina identifikovane su kao Epicoccum nigrum. Patogenost izolata ove gljive potvrđena je pojavom simptoma na veštački inokulisanim sejancima sirka. Molekularna identifikacija obavljena je primenom PCR i amplifikacije ITS regiona ribozomalne DNK. Sekvence gena odabranih izolata 291-09 (JQ619838) i 315-09 (JQ619839) pokazale su 99-100% nukleotidne identičnosti sa sekvencama 31 izolata E. nigrum deponovanihu GenBank bazi podataka. Dobijeni rezultati predstavljaju prvu detaljnu karakterizaciju E. nigrum u Srbiji. Prisustvo većeg broja vrsta gljiva na semenu sirka zahteva dalja ispitivanja njihovih međusobnih odnosa i značaja.

AB  - Cucurbit species grown in the Vojvodina Province, Serbia, were surveyed for the incidence of Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), Cucumber mosaic virus (CMV), Squash mosaic virus (SqMV), Papaya ringspot virus (PRSV) and Tobacco ringspot virus (TRSV) from 2007 to 2009. Samples from more than 700 pumpkin, squash and bottle gourd plants with virus-like symptoms were analyzed by double-antibody sandwich (DAS)-ELISA. ZYMV, WMV and CMV were detected in 79.2, 32.2, and 12.8% of tested samples, respectively. WMV was prevalent in 2007 and ZYMV in 2008-09. Mixed infections were the most frequent type in 2007-08 in contrast to 2009 when single infection of ZYMV prevailed. ZYMV was the most widespread being found in 33 out of 39 inspected fields. Virus species identification was confirmed in selected samples by conventional reverse transcription-polymerase chain reaction (RT-PCR) and sequencing of their coat protein genes. By comparing the obtained virus isolate sequences with those available in GenBank, the identification of serologically detected viruses was confirmed. Phylogenetic analysis based on complete coat protein (CP) sequences highlighted that Serbian ZYMV isolates were closely related to other Central European ZYMV isolates. Finally, additional testing of ELISA-negative samples by RT-PCR using primers specific to six other mosaic viruses revealed the presence of Tomato spotted wilt virus (TSWV) in winter (Cucurbita maxima) and summer (C. pepo 'Beogradska') squash. This is the first report of TSWV natural occurrence on cucurbits in Serbia and on winter squash worldwide.
