AB  - Sunflower (Helianthus annuus L.) is one of the most important oil crops worldwide, including Serbia, where it is grown on about 200,000 ha with an average yield of 3 t/ha. Botrytis cinerea Pers. (1794) regularly occurs on sunflower seeds associated with the phenology of the host plant. However, sunflower seed is often affected by the Rhizopus spp., which causes similar symptoms on seed and seedlings as B. cinerea, and it is important to reliably differentiate those species based on morphological and molecular characteristics. The method used in this study for the detection of B. cinerea is described by ISTA method 7-003. The main task of this study was to isolate the causal agent of grey mould and rot of sunflower seed and seedlings. Infected seeds were covered with overgrown dark-grey mycelia with sporulation. In order to isolate the pathogen, the infected seed was transferred onto potato dextrose agar (PDA) and incubated for 7 days at 20°C. For morphological identification, 11 isolates were single-spored and subcultured onto PDA. Strains formed straight, noddy conidiophores, branched at the top and resembling a cluster with conidia. Conidia were ovoid or elliptical avg. 11.6 × 7.5 µm in size.

Biochemical changes in Oenothera biennis plants infected by 'Candidatus Phytoplasma solani'

AB  - The aim of the present paper was to study the response of Oenothera biennis L. to 'Candidatus Phytoplasma solani' (Stolbur) infection by analyzing total sugars, polyphenols, photosynthetic pigments content and the antioxidant capacity in leaves and roots of healthy and infected plants. The infection caused a significant increase in peroxidation of lipids, phenylalanine ammonia-lyase activity, total sugar, polyphenols and anthocyanins content (2.8, 2.6, 1.8, 1.4, 6.8 fold, respectively), as well as a decrease in photosynthetic pigments (2-6 fold) and total flavonoids (1.5 fold) in the leaves of Oe. biennis. Changes in these parameters were insignificant in the roots except for the total polyphenols content that was 2.7 times higher in the infected ones. Reduced gluthatione content in both tested organs was not affected by the infection (3.7 and 1.7 mu mol/g fresh weight of leaves and roots, respectively). The elevated content of total sugars, flavonoids and polyphenols, as well as the reduction of photosynthetic pigments and anthocyanins in infected plants are indicative of changes in the metabolism of Oe. biennis affected by the Stolbur phytoplasma. In addition to reduction of chlorophyll and carotenoids, the phytoplasma accelerated leaf senescence. Plants responded to the infection via enhanced superoxide anion scavenging, even though this reaction did not prevent, apparently, membrane damage in analysed leaves. This investigation presents new data on the effect of a phytoplasma infection on its host.

Resistance evaluation of different inbreds and hybrids of sunflower to broomrape (Orobanche cumana wallr.)

AB  - Resistance to broomrape of different inbred lines, hybrids and hybrid combinations of sunflower was investigated. The trial for this experiment, which was set at 6 locations on territory of Bačka, included set of differential genotypes which, according to literature, posess genes for resistance to races A-E, 152 inbred and 121 restorer lines developed at the Institute of Field and Vegetable Crops in Novi Sad, 72 crosses between these lines and 15 Russian and Ukrainian varieties. Differential genotypes behaved identically in all test sites. Domestic mother lines, restorer lines, newly developed hybrids as well as hybrid combinations showed resistance to present races of broomrape in all test sites in Vojvodina. Determination of resistance genes requires analysis of crossing inbred lines and differential lines. Results of the experiment performed in Romania and Turkey, with part of the experimental material, shows that these genotypes behaved differently in terms of resistance to broomrape in different conditions for experiments. The results require further separation on the basis of resistance to different races of the parasite by introducing new differential line.
AB  - Ispitivana je otpornost na volovod različitih inbred linija, hibrida i hibridnih kombinacija suncokreta. Ogled za ovaj eksperiment, koji je postavljen na 6 lokacija na teritoriji Bačke, uključio je set diferencijalnih genotipova koji, prema literaturi, imaju gene za otpornost prema rasama A-E, 152 i 121 samooplodne restorer linije stvorene u Institutu za ratarstvo i povrtarstvo, Novi Sad, 72 ukrštanja ovih linija i 15 ruskih i ukrajinskih sorti. Diferencijalni genotipovi su se ponašali identično u svim ispitivanim lokacijama. Domaće linije majke, restorer linije, novorazvijeni hibridi, kao i hibridne kombinacije su pokazali otpornost na prisutne rase volovoda u svim ispitivanim lokacijama na teritoriji Vojvodine. Određivanje gena otpornosti zahteva analizu ukrštanja inbred linija i diferencijalnih linija. Rezultati ogleda izvedenog u Rumuniji i Turskoj sa delom eksperimentalnog materijala pokazuju da su se ovi genotipovi različito ponašali u pogledu otpornosti na volovod u različitim uslovima izvođenja ogleda. Rezultati nalažu dalje razdvajanje na osnovu otpornosti na različite rase volovoda uvođenjem novih diferencijalnih linija.

Prisustvo i karakterizacija virusa žutog mozaika cukinija u usevu lubenice u Srbiji

Na lokalitetu Gornji Tavankut, ZYMV je dokazan u 23,08% testiranih biljaka u pojedinačnim infekcijama, a na lokalitetu Silbaš u 35,29% testiranih biljaka u mešanim infekcijama sa virusom mozaika krastavca i virusom mozaika lubenice. ZYMV je uspešno mehanički prenesen sa prirodno zaraženih biljaka lubenice na Cucurbita pepo 'Ezra F1'. Molekularna detekcija obavljena je RT-PCR metodom umnožavanjem dela gena za nuklearne inkluzije, gena za protein omotača i dela 3' neprepisujućeg regiona, čime je potvrđena identifikacija izolata. Filogenetske analize pokazale su grupisanje izolata poreklom iz lubenice sa drugim izolatima iz Srbije i Centralne Evrope u okviru A-I podgrupe. Analize aminokiselinske sekvence N-terminalnog kraja CP gena, takođe su pokazale pripadnost izolata 550-11 centralnoevropskoj grani.

Characterization of sirodesmins isolated from the phytopathogenic fungus Leptosphaeria maculans

AB  - The pathogenicity of phytopathogenic fungi is associated with phytotoxins, especially with their chemical nature and quantity. Sirodesmins are phytotoxins from the epipolythiodioxopiperazines group, produced by the fungus Leptosphaeria maculans, which are a cause of blackleg and stem canker in oilseed rape (Brassica napus L.). The aim of this work was to obtain a detailed chemical profile of sirodesmins in five fungal isolates (four from Vojvodina, Serbia, and one from the Centre for Agricultural Research, Rothamsted, UK). Sirodesmins showing different phytotoxicity on treated cotyledons of cv. Quinta were separated and detected by thin layer chromatography in all analysed isolates (L.m, C-3, St-5 and S-11) except K-113, which neither contained sirodesmin congeners nor did it exhibit activity. By use of high performance liquid chromatography coupled with tandem mass spectrometer, it was possible to identify total of 10 sirodesmins, together with their precursor-phomamide. It was found that the dominant epipolythiodioxopiperazines of the investigated L. maculans isolates were sirodesmin PL, sirodesmin C, and their de-acetylated derivatives.
AB  - Patogenost fitopatogenih gljiva povezana je sa fitotoksinima, a naročito sa njihovom hemijskom prirodom i količinom. Sirodezmini su fitotoksini iz grupe epipolitiodioksopiperazina, koje proizvodi gljiva Leptosphaeria maculans, uzročnik suve truleži korena i raka stabla uljane repice. Cilj ovog rada bila je detaljna hemijska karakterizacija sirodezmina u pet izolata gljiva (četiri iz Vojvodine i jedan iz Velike Britanije, Centar za poljoprivredna istraživanja, Rothamsted). Kod svih ispitivanih izolata (L. maculans, C-3, St-3, S-11), osim K-113 (koji nije sadržao sirodezmine niti pokazivao aktivnost) tankoslojnom hromatografijom su razdvojeni i detektovani sirodezmini koji su pokazali različitu fitotoksičnost na tretiranim kotiledonima sorte Quinta. Primenom tečne hromatografije visoke efikasnosti, kuplovane sa tandemskim masenim spektrometrom, bilo je moguće identifikovati ukupno 10 sirodezmina, kao i njihov prekursor - fomamid. Non-persistently aphid-borne viruses infecting pumpkin and squash in Serbia and partial characterization of Zucchini yellow mosaic virus isolates

AB  - Cucurbit species grown in the Vojvodina Province, Serbia, were surveyed for the incidence of Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), Cucumber mosaic virus (CMV), Squash mosaic virus (SqMV), Papaya ringspot virus (PRSV) and Tobacco ringspot virus (TRSV) from 2007 to 2009. Samples from more than 700 pumpkin, squash and bottle gourd plants with virus-like symptoms were analyzed by double-antibody sandwich (DAS)-ELISA. ZYMV, WMV and CMV were detected in 79.2, 32.2, and 12.8% of tested samples, respectively. WMV was prevalent in 2007 and ZYMV in 2008-09. Mixed infections were the most frequent type in 2007-08 in contrast to 2009 when single infection of ZYMV prevailed. ZYMV was the most widespread being found in 33 out of 39 inspected fields. Virus species identification was confirmed in selected samples by conventional reverse transcription-polymerase chain reaction (RT-PCR) and sequencing of their coat protein genes. By comparing the obtained virus isolate sequences with those available in GenBank, the identification of serologically detected viruses was confirmed. Phylogenetic analysis based on complete coat protein (CP) sequences highlighted that Serbian ZYMV isolates were closely related to other Central European ZYMV isolates. Finally, additional testing of ELISA-negative samples by RT-PCR using primers specific to six other mosaic viruses revealed the presence of Tomato spotted wilt virus (TSWV) in winter (Cucurbita maxima) and summer (C. pepo 'Beogradska') squash. This is the first report of TSWV natural occurrence on cucurbits in Serbia and on winter squash worldwide.

Status of tobacco viruses in Serbia and molecular characterization of tomato spotted wilt virus isolates

AB  - In a four-year survey to determine the presence and distribution of viruses in tobacco crops at 17 localities of the Vojvodina Province and Central Serbia, 380 samples were collected and analyzed by DAS-ELISA. Out of the seven viruses tested, tomato spotted wilt virus (TSWV), potato virus Y (PVY), tobacco mosaic virus (TMV), cucumber mosaic virus (CMV), and alfalfa mosaic virus (AMV) were detected in 37.9, 33.4, 28.7, 23.9, and 15.5% of the total tested samples, respectively. TSWV was the most frequently found virus at the localities of Central Serbia, while PVY and CMV were the most frequent viruses in the Vojvodina Province. Single infections were prevalent in years 2005–2007 and the most frequent were those of PVY. A triple combination of those viruses was most frequent mixed infection type in 2008. The presence of all five detected viruses was confirmed in selected ELISA-positive samples by RT-PCR and sequencing. The comparisons of obtained virus isolate sequences with those available in NCBI, confirmed the authenticity of serologically detected viruses. Phylogenetic analysis based on partial nucleocapsid gene sequences revealed a joint clustering of Serbian, Bulgarian and Montenegrin TSWV isolates into one geographic subpopulation, which was distinct from the other subpopulation of TSWV isolates from the rest of the European countries. The high incidence of viruses in Serbian tobacco crops highlights the importance of enhancing farmers knowledge towards better implementation of control strategies for preventing serious losses.

Characterization of cucumber mosaic virus originating from cucurbits in Serbia

AB  - Cucumber mosaic virus (CMV) is considered one of the most economically important plant viruses and has a worldwide distribution and a very wide host range including plants from family Cucurbitaceae. In Serbia, on cucurbits CMV was detected in single and mixed infections with Zucchini yellow mosaic virus (ZYMV) and Watermelon mosaic virus (WMV). Viruses, including CMV, are constantly present in cucurbit crops, but their frequency changes by year and locality. Surveys and sample collections were conducted in cucurbit crops in the period from 2008 to 2009 at 15 localities in Vojvodina province, and sample testing was carried out using the DAS-ELISA method and commercially available antisera for six economically most important cucurbit viruses. In 2008, a total of 51 samples were collected from 13 cucurbit crops of oilseed pumpkin Olinka variety, squash, and bottle gourd and CMV was detected in a total of 55% of tested samples with symptoms of viral infection. The most common infectious type was mixed infection with ZYMV and WMV (35.3%), and then mixed infection with ZYMV (17.7%) and WMV (2%). A total of 599 symptomatic samples of oilseed pumpkin Olinka variety, zucchini squash varieties Beogradska and Tosca, squash, and winter squash were collected in 15 cucurbits crops in 2009. CMV was present in 4.4% of total collected samples, in single infections in 1.3%, and in mixed with WMV or ZYMV in 1.3%, and 1.8%. Five CMV isolates were obtained by mechanical inoculations of N. glutinosa and one of them was selected for further biological characterization. Test plants which were described to be hosts of CMV expressed symptoms characteristic for those caused by CMV after inoculations by isolate 115-08. CMV specific primers Au1u/Au2d were used to amplify an 850 bp fragment using RT-PCR method. Amplified fragment encodes the entire viral coat protein (CP) gene and partial 5' and 3' UTRs of two selected CMV isolates. Amplified fragments were sequenced and deposited in the NCBI, where they were assigned accession numbers, HM065510 (115-08) and HM065509 (151-08). The sequences of CMV isolates from Serbia shared the highest nucleotide and amino acid identity with isolates from subgroup IA, from 99.5 to 97.4% and 99.1 to 97.4%, and the lowest identities were with the subgroup II isolates from 66.9 to 64, 5%, from 75.8 to 74.1%.
