AB  - Flavescence dorée (FD) is a European quarantine grapevine disease transmitted by the Deltocephalinae leafhopper Scaphoideus titanus. Whereas this vector had been introduced from North America, the possible European origin of FD phytoplasma needed to be challenged and correlated with ecological and genetic drivers of FD emergence. For that purpose, a survey of genetic diversity of these phytoplasmas in grapevines, S. titanus, black alders, alder leafhoppers and clematis were conducted in five European countries. Out of 132 map genotypes, only 11 were associated to FD outbreaks, three were detected in clematis, whereas 127 were detected in alder trees, alder leafhoppers or in grapevines out of FD outbreaks. Most of the alder trees were found infected, including 8% with FD genotypes M6, M38 and M50, also present in alders neighboring FD-free vineyards and vineyard-free areas. The Macropsinae Oncopsis alni could transmit genotypes unable to achieve transmission by S. titanus, while the Deltocephalinae Allygus spp. and Orientus ishidae transmitted M38 and M50 that proved to be compatible with S. titanus. Variability of vmpA and vmpB adhesin-like genes clearly discriminated 3 genetic clusters. Cluster Vmp-I grouped genotypes only transmitted by O. alni, while clusters Vmp-II and -III grouped genotypes transmitted by Deltocephalinae leafhoppers. Interestingly, adhesin repeated domains evolved independently in cluster Vmp-I, whereas in clusters Vmp-II and–III showed recent duplications. Latex beads coated with various ratio of VmpA of clusters II and I, showed that cluster II VmpA promoted enhanced adhesion to the Deltocephalinae Euscelidius variegatus epithelial cells and were better retained in both E. variegatus and S. titanus midguts. Our data demonstrate that most FD phytoplasmas are endemic to European alders. Their emergence as grapevine epidemic pathogens appeared restricted to some genetic variants pre-existing in alders, whose compatibility to S. titanus correlates with different vmp gene sequences and VmpA binding properties.

Occurrence and molecular characterization of Impatiens necrotic spot tospovirus in ornamentals in Serbia

AB  - A subset of ornamentals grown in Serbia were surveyed for the presence of impatiens necrotic spot tospovirus (INSV) from 2008 to 2018. Out of 1578 samples tested for the presence of the virus using double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), INSV was detected in 26 Begonia sp. and four Tulipa sp. samples collected in 2008 and in 10 samples of each of Begonia sp., Impatiens walleriana and I. hawkeri 'SunPatiens' and 'New Guinea' collected in 2018. INSV was not detected in any samples collected during 2009-2017. For further confirmation, the nucleotide sequence of the INSV nucleocapsid (N) gene was obtained by reverse transcription polymerase chain reaction (RT-PCR) in two and four samples collected in 2008 and 2018, respectively. The success of mechanical transmission of INSV isolates from different hosts was variable depending on isolates. Serbian INSV isolates showed very low nucleotide diversity and they were closely related to the Chinese and Japanese isolates already reported in the literature, indicating that the Serbian isolates might be of Asian origin. Phylogenetic analysis and the median-joining haplotype network of the N gene sequences showed clustering of Serbian INSV isolates in the Asian clade into two different subclades with different recent common ancestors implying two independent introductions. The sporadic occurrence of INSV with high temporal distance is a likely consequence of virus spread through the international trade in ornamental planting material. Therefore, the establishment of the virus should be considered and prevented when introducing new plant material into the country. This is also the first report of the tulip as a natural host of INSV.

the subfamily Aphidiinae, due to their low economic importance, as they do not parasitize pests
of cultivated plants. Consequently, data about this group are scarce, including its taxonomy and
phylogeny. In the present study, we explore inter- and intraspecific genetic variation of Monoctonina
species, including genera Monoctonus Haliday 1833, Monoctonia Starý 1962, Falciconus Mackauer
1959 and Harkeria Cameron 1900. We employ two molecular markers, the barcode region of the
mitochondrial cytochrome c oxidase subunit I (COI) and the D2 region of the 28S nuclear gene
(28S rDNA), to analyze genetic structuring and phylogeny of all available Monoctonina species,
and combine them with morphological data for an integrative approach. We report one new species,
and three potentially new species which can be formally described when further specimens are
available. Analysis of phylogenetic relationships within the subtribe shows a basal position for the
genera Falciconus and Monoctonia, and the close relatedness of Harkeria and Monoctonus.

Diversity of phytoplasmas identified in the polyphagous leafhopper Euscelis incisus (Cicadellidae, Deltocephalinae) in Serbia: pathogen inventory, epidemiological significance and vectoring potential

Two generations per year allows this species to come into contact with a plethora of different phytoplasmas. We first investigated the interactions of E. incisus with phytoplasmas in different ecosystems as a bioindicator of phytoplasma diversity. Among the 510 analyzed E. incisus specimens collected from eight localities in Serbia, 53 (10%) tested positive for diverse ‘Ca. Phytoplasma’ taxa. Restriction fragment length polymorphism (RFLP) analysis and sequence comparison based on the F2nR2 fragment of the 16S rRNA gene revealed the presence of six 16S phytoplasma ribosomal groups and eight subgroups in E. incisus (16SrI-R, 16SrI-F, 16SrII-E, 16SrIII-B, 16SrIX-C, 16SrIX-E, 16SrXI-G and 16SrXII-A). The most prevalent was the 16SrXII-A phytoplasma group, represented by two multilocus stolbur phytoplasma genotypes: STOLg and Rqg31g. While the 16SrXII-A phytoplasmas were identical to the previously recorded isolates from Serbia, the 16SrI-F and 16SrI-R subgroups closely related to the previously documented 16SrI-B and 16SrI-C subgroups were recorded for the first time in Serbia. In addition, records of the 16SrIX and 16SrXI groups represent the first findings of these phytoplasma groups in Serbia. Transmission trials of ‘Ca. P. solani’ were performed with naturally infected E. incisus adults. The leafhoppers successfully transmitted stolbur phytoplasma to exposed Catharanthus roseus plants, indicating their role as a natural vector.

Multipotentnost cikade Euscelis incisus (Hemiptera, Cicadellidae, Deltocephalinae) u prenošenju i epidemiologiji fitoplazmi ('Candidatus Phytoplasma')

AB  - Euscelis incisus je polifagna vrsta, koja je široko rasprostranjena i česta u različitim agroekosistemima. Ima dve generacije godišnje koje joj omogućavaju da uspešno ostvari kontakt sa različitim grupama i vrstama fitoplazmi. Od ukupno 510 analiziranih primeraka E. incisus sakupljenih na 8 lokaliteta u Srbiji, 53 (10.4%) je bilo pozitivno na prisustvo različitih fitoplazmi. RFLP analizom F2nR2 fragmenta 16S rRNK regiona, utvrđeno je prisustvo šest 16S ribozomalnih grupa i osam podgrupa fitoplazmi u jedinkama E. incisus (16SrI-R, 16SrI-F, 16SrII-E, 16SrIII-B, 16SrIX-C, 16SrIX-E, 16SrXI-G i 16SrXII-A). Najzastupljenija je bila 16SrXII-A podgrupa prisutna u jedinkama E. incisus sa dva multilokusna genotipa: STOLg i Rqg31g. Fitoplazme 16SrIX i 16SrXI grupe, kao i dve podgrupe unutar Aster yellows fitoplazme, 16SrI-F i 16SrI-R zabeležene su po prvi put u Srbiji. Fitoplazme 16SrII-E, 16SrIII-B, 16SrIX-C, 16SrIX-E i 16SrXI-G grupe predstavljaju prve nalaze ovih patogena u jedinkama E. incisus. U biljnom materijalu je prvi put registrovano prisustvo 16SrIII grupe fitoplazmi u biljkama Cirsium vulgare, Carduus acanthoides, Lathyrus tuberosus i Lathyrus aphaca, dok je prisustvo 16SrIX grupe fitoplazmi utvrđeno prvi put u biljkama Cichorium intybus i Knautia arvensis u Srbiji. Eksperimenti prenošenja fitoplazmi pomoću prirodno inficiranih populacija vrste E. incisus pokazali su da je ova vrsta uspešan prirodni vektor u prenošenju fitoplazmi 16SrIII i 16SrXII grupe. The vector status of the deltocephalinae leafhopper Neoaliturus fenestratus harbouring ‘Candidatus Phytoplasma solani’ (“stolbur”) was tested in two separate transmission trials with seedlings of lettuce and carrot. In both tests, the leafhopper successfully transmitted the “stolbur” phytoplasma to the exposed plants, which expressed partial leaf discoloration and rolling and rapid plant desiccation. Molecular characterization of the stamp gene encoding the antigenic membrane protein determined the presence of three “stolbur” phytoplasma strains in field collected N. fenestratus and in the insect inoculated plants. The same genotypes of phytoplasma shared by the vector samples and the exposed lettuce and carrot seedlings confirm the ability of N. fenestratus to transmit the “stolbur” phytoplasma to the tested plants. These findings indicate a possible role of N. fenestratus in the epidemiology of the “stolbur” phytoplasma in vegetable crops.

AB  - Surveys for the presence of symptoms typical for the “stolbur” disease were conducted during 2015 and 2016 in pepper growing areas of Serbia. ‘Candidatus Phytoplasma solani’ detection was performed by Stol11 primer amplification. Characterisation was done by multi-locus typing (MLST) of tuf, secY, vmp1 and stamp genes. The disease was observed in pepper in all the surveyed locations except in south Serbia, whith a low incidence (3-5%). Overall, 13 MLST genotypes were distinguished, and the most common were Rqg31/Rqg50, Rpm35/M5 and STOL.

Characterization of Gnomoniopsis idaeicola, the Causal Agent of Canker and Wilting of Blackberry in Serbia

AB  - Blackberry cane diseases with the symptoms of necrosis, canker, and wilting are caused by several fungi worldwide. Surveys conducted from 2013 to 2016 in Serbia revealed the occurrence of Gnomoniopsis idaeicola, the causal agent of cane canker and wilting, which was found to be distributed in almost half of the surveyed orchards, in three blackberry cultivars, and with disease incidence of up to 80%. Wide distribution and high disease incidence suggest that G. idaeicola has been present in Serbia for some time. Out of 427 samples, a total of 65 G. idaeicola isolates were obtained (isolation rate of 34.19%). Reference isolates, originating from different localities, were conventionally and molecularly identified and characterized. G. idaeicola was detected in single and mixed infections with fungi from genera Paraconiothyrium, Colletotrichum, Diaporthe, Botryosphaeria, Botrytis, Septoria, Neofusicoccum, and Discostroma, and no diagnostically specific symptoms could be related directly to the G. idaeicola infection. In orchards solely infected with G. idaeicola, blackberry plant mortality was up to 40%, and yield loses were estimated at 50%. G. idaeicola isolates included in this study demonstrated intraspecies diversity in morphological, biological, pathogenic, and molecular features, which indicates that population in Serbia may be of different origin. This is the first record of a massive outbreak of G. idaeicola infection, illustrating its capability of harmful influence on blackberry production. This study represents the initial step in studying G. idaeicola as a new blackberry pathogen in Serbia, aiming at developing efficient control measures.

AB  - A new vagrant eriophyoid mite species of the archaic genus Pentasetacus (Schliesske 1985), P. novozelandicus n. sp., is described with the aid of conventional microscopy, confocal laser scanning microscopy and scanning electron microscopy. It was found on Araucaria heterophylla, which is an araucarian that is endemic to Norfolk Island and introduced to New Zealand. Partial sequences of mitochondrial barcode COI gene and D1–D2 domains of nuclear rDNA of two pentasetacid mites, P. araucariae (MK903025 and MK898944) and P. novozelandicus n. sp. (MK903024 and MK898943) are provided. Molecular phylogenetic analyses of full-length D1–D2 eriophyoid sequences, including GenBank sequences and newly generated sequences of pentasetacids, confirmed the monophyly of Pentasetacidae but failed to resolve the basal phylogeny of Eriophyoidea. This may be because the D1–D2 domains of 28S are hypervariable in Eriophyoidea. Moreover, in pentasetacids D1–D2 sequences are about 20% shorter than in other eriophyoids, and thus harder to align. Two types of anal lobes are described in Eriophyoidea: (1) Eriophyidae s.l. and Phytoptidae s.l. have bilaterally symmetric lobes; (2) pentasetacids have non-divided lobes. abstract = "A new vagrant eriophyoid mite species of the archaic genus Pentasetacus (Schliesske 1985), P. novozelandicus n. sp., is described with the aid of conventional microscopy, confocal laser scanning microscopy and scanning electron microscopy. It was found on Araucaria heterophylla, which is an araucarian that is endemic to Norfolk Island and introduced to New Zealand. Partial sequences of mitochondrial barcode COI gene and D1–D2 domains of nuclear rDNA of two pentasetacid mites, P. araucariae (MK903025 and MK898944) and P. novozelandicus n. sp. (MK903024 and MK898943) are provided. Molecular phylogenetic analyses of full-length D1–D2 eriophyoid sequences, including GenBank sequences and newly generated sequences of pentasetacids, confirmed the monophyly of Pentasetacidae but failed to resolve the basal phylogeny of Eriophyoidea. This may be because the D1–D2 domains of 28S are hypervariable in Eriophyoidea. Moreover, in pentasetacids D1–D2 sequences are about 20% shorter than in other eriophyoids, and thus harder to align. Two types of anal lobes are described in Eriophyoidea: (1) Eriophyidae s.l. and Phytoptidae s.l. have bilaterally symmetric lobes; (2) pentasetacids have non-divided lobes. It is known that these species are broadly distributed, parasitizing various aphid hosts and showing great biological and ecological diversity. On the other hand, this group of endoparasitoids is understudied and was thought to be represented by a single species in Europe, viz., Paralipsis enervis (Nees). However, recent description of two new species indicated the possibility of cryptic speciation and recognition of additional Paralipsis species in Europe. In this research, Paralipsis specimens collected during the last 60 years from eight European countries, as well as one sample from Morocco, were subjected to molecular and morphological characterization. Newly designed genus-specific degenerative primers successfully targeted short overlapping fragments of COI of the mitochondrial DNA. Molecular analyses showed clear separation of four independent lineages, two of which are the known species P. enervis and P. tibiator, while two new species are described here, viz., P. brachycaudi Tomanovic & Stary, sp. n. and P. rugosa Tomanovic & Stary, sp. n. No clear specialization of the taxa to a strict root aphid host has been determined. The recognized mitochondrial lineages were distinct one from another, but with a substantial within-lineage divergence rate, clearly indicating the complexity of this group of parasitoids, on which further research is required in order to clarify the factors triggering their genetic differentiation. We reviewed literature data and new records of Paralipsis enervis aphid host associations and distributions. A key for the identification of all known Paralipsis species is provided and illustrated.

Resource allocation in response to herbivory and gall formation in Linaria vulgaris

AB  - Trehalose and its precursor, trehalose 6-phosphate (T6P), are essential regulators of plant response to abiotic and biotic stress. Here we used the specific host-insect interaction between Linaria vulgaris (Plantaginaceae) and stem-galling weevil, Rhinusa pilosa (Mecinini, Curculionidae) with the aim to distinguish carbohydrate allocation patterns in response to herbivory, gall formation (G1, 24 h after oviposition), and gall development (G2, 7 days after oviposition) under controlled conditions. The hypothesis is that herbivory and galling induce distinct responses in both leaves and stems, and that shifts in carbon allocations are regulated by signaling sugars. Systemic response to herbivory was accumulation of T6P and maltose. The main feature of G1 in the stems was accumulation of trehalose, accompanied by increased T6P, turanose and glucose content, oppositely to the leaves. In G2, galls had 3-folds higher weight than controls, with further accumulation of fructose, glucose, turanose, and total water-insoluble carbohydrates (TIC), while the sucrose/hexose ratio decreased. Analysis of fast chlorophyll fluorescence kinetic (OJIP) transients in G2 showed a slight decrease in quantum yield of electron transport flux from Q(A) to Q(B), and towards photosystem I acceptor side, correlated with the decreased content of photosynthetic pigments and hexoses accumulation. Redistribution of photosynthates, and accumulation of T6P were induced in response to herbivory, indicating its signaling role. The results support the hypothesis that R. pilosa can induce plant reprogramming towards the accumulation of beneficial carbohydrates in developing gall by mechanisms which include both T6P and trehalose.

AB  - Bois noir (BN), a grapevine yellows disease induced by Candidatus Phytoplasma solani', is spreading and diversifying in several wine-producing regions of Europe, causing increased yield losses and economic concerns. The planthopper Hyalesthes obsoletus (Hemiptera: Cixiidae) is a major vector of the pathogen that causes BN disease in grapevine. Recent studies suggest that epidemiology of BN is affected by the interaction of specific host races of H. obsoletus with weedy host plants that serve as reservoirs for the pathogen. Since BN is common in grape-growing regions of Serbia, a survey was conducted during 2015-2016 to evaluate the host-plant association and vector role of H. obsoletus in central and eastern regions. We tested populations of H. obsoletus associated with Convolvulus arvensis (field bindweed) a known host for the vector and pathogen and Crepis foetida (stinking hawk's-beard) a newly identified host in southeastern Europe. Molecular phylogenetic analyses of H. obsoletus collected from C. arvensis and C. foetida using the mtDNA COI marker determined that the two populations had a genetic divergence of 1.4%. Transmission trials confirmed the vector role of C. arvensis-associated H. obsoletus and identified C. foetida-associated populations as a new and efficient host-specialized vector of BN. Each of the two host-specialized vector populations transmitted Ca. P. solani' to its original host (C. arvensis or C. foetida) and to grapevine cv. Chardonnay. Multilocus genotyping of the transmitted phytoplasma isolates based on tuf/stamp/vmp1 Ca. P. solani'-specific loci indicated pathogen segregation according to the host-plant association of the vector. All isolates transmitted by C. foetida-associated H. obsoletus were of the STOL genotype (tuf-b/STOL/V2-TA), while the C. arvensis-associated population transmitted several genotypes: tuf-b/Rqg50/V4, tuf-b/Rqg50/V14, tuf-b/Rqg31/V14 and tuf-b/Vv24/V14. This study provides evidence of a new independent BN epidemiological cycle of tuf-b Ca. P. solani' strain in the Balkan region, sourced by C. foetida and transmitted by its associated H. obsoletus population.

Evolution of host plant use and diversification in a species complex of parasitic weevils (Coleoptera: Curculionidae)

AB  - Weevils (Coleoptera: Curculionoidea) represent one of the most diverse groups of organisms on Earth; interactions with their host plants have been recognized to play a central role in their remarkable diversity, yet the exact mechanisms and factors still remain poorly understood. Using phylogenetic comparative analyses, here we investigate the evolution of host use and its possible role in diversification processes of Rhinusa and Gymnetron, two closely related groups of weevils that feed and develop inside plant tissues of hosts within the families Scrophulariaceae and Plantaginaceae. We found strong evidence for phylogenetic conservatism of host use at the plant family level, most likely due to substantial differences in the chemical composition of hosts, reducing the probability of shifts between host families. In contrast, the use of different plant organs represents a more labile ecological trait and ecological niche expansion that allows a finer partitioning of resources. Rhinusa and Gymnetron weevils initially specialized on plants within Scrophulariaceae and then shifted to the closely related Plantaginaceae; likewise, a gall inducing behavior evolved from non-galler weevils, possibly in response to resource competition, as galls facilitate larval development by providing enhanced nutrition and a favorable microhabitat. Results from trait-dependent diversification analyses suggest that both use of hosts within Plantaginaceae and parasitism on fruits and seed capsules are associated with enhanced diversification of Rhinusa and Gymnetron via low extinction rates. Our study provides quantitative evidence and insights on the ecological factors that can promote diversification in phytophagous insects that feed and develop inside plant tissues.

Using phylogenetic comparative analyses, here we investigate the evolution of host use and its possible role in diversification processes of Rhinusa and Gymnetron, two closely related groups of weevils that feed and develop inside plant tissues of hosts within the families Scrophulariaceae and Plantaginaceae. We found strong evidence for phylogenetic conservatism of host use at the plant family level, most likely due to substantial differences in the chemical composition of hosts, reducing the probability of shifts between host families. In contrast, the use of different plant organs represents a more labile ecological trait and ecological niche expansion that allows a finer partitioning of resources. Rhinusa and Gymnetron weevils initially specialized on plants within Scrophulariaceae and then shifted to the closely related Plantaginaceae; likewise, a gall inducing behavior evolved from non-galler weevils, possibly in response to resource competition, as galls facilitate larval development by providing enhanced nutrition and a favorable microhabitat. AB  - A new vagrant eriophyoid mite species, Setoptus tsugivagus n. sp. Chetverikov (Eriophyoidea, Phytoptidae, Nalepellinae, Nalepellini), is described from the needles of the western hemlock, Tsuga heterophylla (Rafinesque) Sargent (Pinaceae) in Vancouver, Canada. The new species can be distinguished from all other members of Setoptus by a distinct pattern of several short longitudinal ridges on the posterior half of the prodorsal shield. Elements of the anal secretory apparatus (ASA) were observed in adults of S. tsugivagus n. sp., suggesting that the ASA is present in both major phylogenetic lineages of Eriophyoidea (Eriophyidae s. l. and Phytoptidae s. l.). Therefore, this structure could be a synapomorphy for all Eriophyoidea. We briefly discuss the function and morphological variety of the ASA in Eriophyoidea. D2 28S rDNA sequences of four nalepelline species were obtained: Boczekella reticulata Bagnyuk 1987 (GenBank accession number MK124605), Nalepella tsugifoliae Keifer 1953 (MK124606), Setoptus pini Boczek, 1964 (MK124607), and S. tsugivagus n. sp. (MK124608). Molecular phylogenetic analyses of D2 28S rDNA sequences of the mites of the subfamily Nalepellinae confirmed monophyly of the tribe Nalepellini and retrieved Setoptus and Nalepella as polyphyletic. Additionally, our data indicate that 28S rDNA sequence KF782472.1, previously reported to be Trisetacus ehmanni Keifer 1963 by Li et al. (2014), belongs to another species of the genus Trisetacus, possibly T. quadrisetus (Thomas), and the sequence KF782471.1, previously reported to be Trisetacus sp., might belong to T. juniperinus (Nalepa). We also discuss the systematics of nalepellines and their host association with conifers.

AB  - Supplementary morphological descriptions of five Cecidophyopsis species collected in Europe (Serbia, Austria, Italy and Montenegro) are given: Cecidophyopsis vermiformis from Corylus avellana L. (Betulaceae), C. hendersoni from Yucca gigantea Lem. (Asparagaceae), C. verilicis from Ilex aquifolium L. (Aquifoliaceae), C. psilaspis from Taxus baccata L. (Taxaceae) and C. malpighianus from Laurus nobilis L. (Lauraceae). Males of C. vermiformis, C. verilicis, C. hendersoni and C. malpighianus, and immatures of C. hendersoni and C. verilicis, are described for the first time. C. verilicis is recorded for the first time in the fauna of Serbia and the European region. Female cuticle-lined internal genitalia of five Cecidophyopsis species are studied under confocal laser scanning microscopy. A several steps of oviposition in cecidophyines is proposed based on CLSM observations on their internal genitalia. Sequences of the barcoding region of the mitochondrial cytochrome oxidase subunit I (mtCOI) gene are given for the following species: C. hendersoni, C. verilicis, C. psilaspis and C. malpighianus.

AB  - Brojnost virusa infektivnih za vinovu lozu stalno se povećava, tako da je do sada opisano više od 60 vrsta koji mogu da zaraze vinovu lozu. Paralelno sa globalnom ekspanzijom vinogradarske i vinarske industrije zapaženo je povećanje učestalosti i ekonomskog značaja oboljenja uvijenosti lišća vinove loze (Grapevine Leafroll Disease, GLD). GLD, osim što smanjuje vitalnost i dugovečnost vinove loze, značajno smanjuje prinos i kvalitet grožđa. Epidemiologija oboljenja je nedovoljno istražena zbog kompleksnosti ove bolesti, genetički i serološki različitih vrsta unutar kompleksa i različitosti simptoma kod crvenih i belih sorti vinove loze. Кompleks virusa prouzrokovača uvijenosti lišća vinove loze (Grapevine leafrollassociated viruses, GLRaVs) čini pet različitih virusa koji pripadaju familiji Closteroviridae: jedna vrsta koja pripada rodu Closterovirus (GLRaV-2), tri rodu Ampelovirus (GLRaV-1, GLRaV -3, GLRaV -4, i njegovi sojevi: GLRaV-4 soj 5, GLRaV-4 soj 6, GLRaV-4 soj 9, GLRaV-4 soj Pr, GLRaV-4 soj De i GLRaV-4 soj Car) i jedna vrsta iz roda Velarivirus (GLRaV-7). GLRaVs se prenose kalemljenjem i propagativnim materijalom (podloga i vijoka), a neke vrste i štitastim vašima. Virusi koji izazivaju uvijenost lišća se obično javljaju u mešanoj infekciji, a često su prisutni i u mešanoj infekciji sa drugim virusima vinove loze. Ranija istraživanja sprovedena u vinogorjima u Srbiji utvrdila su prisustvo GLRV-1, -2 i -3, pri čemu je GLRaV-3 bio najzastupljeniji. Ukoliko je širenje virusa unutar zasada putem vektora izraženo, krčenje ili uklanjanje zaraženih čokota, kontrola štitastih vaši i adekvatne sanitarne mere mogu redukovati nove infekcije.

AB  - Hyalesthes obsoletus, vektor biljnog patogena 'Candidatus (Ca.) Phytoplasmasolani', smatra se polifagnom štetočinom od ekonomskog značaja u centralnojEvropi i Mediteranu. Istraživanjima sprovedenim u jugoistočnoj Evropi, kojagravitira centru arela vrste i gde je zabeležen značajan broj 'Ca. P. solani'-uslovljenih biljnih bolesti, utvrđeno je postojanje četiri asocijacije vektora sabiljkama dualnim domaćinima: Convolvulus arvensis (Ca), Urtica dioica (Ud), Vitexagnus-castus (Vac) i Crepis foetida (Cf). Populaciono-genetičkim analizamamitohondrijskih i nuklearnih markera (mikrosatelita) utvrđena je kriptičnadiferencijacija simpatričkih (uključujući sintopske) i geografski udaljenih H.obsoletus populacija na tri filogenetičke linije uslovljene biljkom domaćinom: (1)Ca-Ud, (2) Vac i (3) Cf. Pored jasne diferencijacije na mitohondrijskim genima(1.1% - 1.5%) i mikrosatelita koji potvrđuju izdvajanje populacija u jedan od triklastera prema biljci domaćinu (pripadnost >90%), podršku za izdizanje genetičkedivergencije H. obsoletus sensu lato na nivo kriptične specijacije daje registrovanadistanca u odnosu na morfološki najsrodniju vrstu - Hyalesthes thracicus (2.1% -3.3%). Posledice specijalizacije i diferencijacije H. obsoletus sensu lato premabiljkama domaćinima na epidemiologiju 'Ca. P. solani' procenjene su multigenskomkarakterizacijom izolata fitoplazme detektovanih (1) in situ u prirodnimepidemiološkim sistemima mediteranskih vinograda i (2) eksperimentimaunakrsne inokulacije biljaka putem prirodno inficiranih populacija vektora.Potvrđena je osnovna epidemiološka divergencija 'Ca. AB  - The cixiid planthopper Hyalesthes obsoletus is generally considered as apolyphagous vector of 'Candidatus (Ca.) P. solani' associated with numerous wildand cultivated plants in central Europe and Mediterranean. Research conducted insoutheastern Europe, the distribution centre of H. obsoletus and the area of most'Ca. P. solani'-inflicted crop diseases, points toward specific dual host-plantassociations with: Convolvulus arvensis (Ca), Urtica dioica (Ud), Vitex agnus-castus(Vac) and Crepis foetida (Cf). Population-genetic analysis of mitochondrial andnuclear genetic markers (microsatellites) have revealed cryptic differentiationbetween sympatric (including syntopic) and geographically distant H. obsoletuspopulations into three phylogenetic lineages delineated by host-plant preference:1) Ca-Ud, 2) Vac i 3) Cf. Pored jasne diferencijacije na mitohondrijskim genima(1.1% - 1.5%) i mikrosatelita koji potvrđuju izdvajanje populacija u jedan od triklastera prema biljci domaćinu (pripadnost >90%), podršku za izdizanje genetičkedivergencije H. obsoletus sensu lato na nivo kriptične specijacije daje registrovanadistanca u odnosu na morfološki najsrodniju vrstu - Hyalesthes thracicus (2.1% -3.3%). Posledice specijalizacije i diferencijacije H. obsoletus sensu lato premabiljkama domaćinima na epidemiologiju 'Ca. P. solani' procenjene su multigenskomkarakterizacijom izolata fitoplazme detektovanih (1) in situ u prirodnimepidemiološkim sistemima mediteranskih vinograda i (2) eksperimentimaunakrsne inokulacije biljaka putem prirodno inficiranih populacija vektora.Potvrđena je osnovna epidemiološka divergencija 'Ca. AB  - The stolbur phytoplasma vector Hyalesthes obsoletus is generally considered as a polyphagous species associated with numerous wild and cultivated plants. However, recent research in southeastern Europe, the distribution centre of H. obsoletus and the area of most stolbur-inflicted crop diseases, points toward specific host-plant associations of the vector, indicating specific vector-based transmission routes. Here, we study the specificity of populations associated with four host-plants using mitochondrial and nuclear genetic markers, and we evaluate the evolution of host-shifts in H. obsoletus. Host-plant use was confirmed for Convolvulus arvensis, Urtica dioica, Vitex agnus-castus and Crepis foetida. Mitochondrial genetic analysis showed sympatric occurrence of three phylogenetic lineages that were ecologically delineated by host-plant preference, but were morphologically inseparable. Nuclear data supported the existence of three genetic groups (Evanno's Delta K(3) = 803.72) with average genetic membership probabilities  gt  90%. While populations associated with C. arvensis and U. dioica form a homogenous group, populations affiliated with V. agnus-castus and C. foetida constitute two independent plant-associated lineages. The geographical signal permeating the surveyed populations indicated complex diversification processes associated with host-plant selection and likely derived from post-glacial refugia in the eastern Mediterranean. This study provides evidence for cryptic species diversification within H. obsoletus sensu lato: i) consistent mitochondrial differentiation (1.1-1.5%) among host-associated populations in syntopy and in geographically distant areas, ii) nuclear genetic variance supporting mitochondrial data, and iii) average mitochondrial genetic distances among host-associated meta-populations are comparable to the most closely related, morphologically distinguishable species, i.e., Hyalesthes thracicus (2.1-3.3%).

AB  - A new species of plant mite (Acari: Eriophyidae) was discovered on medusahead (Taeniatherum caputmedusae), an annual grass that is native to central Asia and the Mediterranean Basin. It is invasive in western North America. Aculodes altamurgiensis sp. nov., is described here and differentiated from other Aculodes spp., on the basis of morphology. Its DNA fingerprinting was reported and compared with Aculodes mckenziei collected from Elymus repens and Bromus inermis. Pairwise comparison of MT-001 sequences between A. altamurgiensis sp. nov., and A. mckenziei revealed 20.2-21.5% genetic divergence between these congeneric species. First collected in Parco Nazionale dell'Alta Murgia in Apulia, Italy in 2014,A. altamurgiensis sp. nov., has been subsequently collected from medusahead in Serbia, Bulgaria, Iran and Turkey. Based on these data and on preliminary observations on the effects of the mite on plant growth, A. altamurgiensis sp. nov., is currently being investigated as a candidate biological control agent of medusahead.

AB  - Archival specimens are a great resource for molecular research in population biology, taxonomy and conservation. A primary goal for researchers is to preserve specimens from collections by improving noninvasive methods for DNA extraction and to achieve successful amplification of the short fragments of a target gene in the event of DNA fragmentation. We tested the suitability of a noninvasive method of DNA extraction and amplification of the barcoding region of the mitochondrial gene cytochrome c oxidise subunit I from archival specimens of aphid parasitoids belonging to the genera Aphidius, Lysiphlebus and Praon (Aphidiinae, Braconidae, Hymenoptera). Using a commercial kit as a noninvasive method, we successfully extracted DNA from dry 7 to 41 year old samples of 26 different parasitoid species. However, amplification of the barcoding region failed using the standard primer pair LCO1490/HCO2198. In order to reconstruct DNA barcodes we designed internal genus-specific degenerative primers and a new amplification protocol to target the short fragments within the mitochondrial region. Novel primers were designed using as a template the reference sequences from congeners retrieved from the public database. The combination of standard primers with internal primers, in direct and nested amplification reactions, produced short overlapping subsequences, concatenated to recover long barcoding sequences. Additional analyses also confirmed that primers initially designed for Aphidius, Lysiphk.hus and Praon can be combined in a mixture, and successfully used to obtain short fragments of disintegrated DNA from archival specimens of several other braconid species from the genera Ephedrus and Monoctonus.

AB  - The relationship between the genera Cleopomiarus and Miarus of Mecinini (Curculionidae, Curculioninac) was tested on the basis of morphological characters from the immature stages. The mature larvae of five Cleopomiarus species (C. distinctus (Boheman, 1845), C. graminis (Gyllenhal, 1813), C. longirostris (Gyllenhal, 1838), C. medius (Desbrochers des Loges, 1893), and C. meridionalis (H. Brisout de Barneville, 1863)), three Miarus species (M. abnormis Solari, 1947, M. ajugae (Herbst, 1795), and M. campanulae (Linnaeus, 1767)), and the pupae of four Cleopomiarus species (C. distinct us, C. graminis, C. longirostris, and C. medius) and two Miarus species (M. abnormis and M. ajugae) are described in detail for the first time. To confirm the taxonomic identification of some larvae, DNA COI barcode was obtained and compared with those of adults. The immature stages of the species herein studied were compared with those known from other genera in tribe Mecinini. It is suggested that Miarus and Cleopomiarus may be monophyletic based on several shared distinctive characters. Larvae of Miarus have a characteristic maxil-lary mala with six finger-like dms of two sizes (one or two elms very long and the rest of medium length), this feature being apparently unique among weevils. Other genus-specific character states are observed in the pupae, such as the length of setae on the head, rostrum and pronotum, including the number of rs on the rostrum, ds on pronotum, and finally the shape of the urogomphi. A key to the described larvae and pupae were respectively presented. New biological and distributional data on some species are reported.

AB  - The European lantern fly, Dictyophara europaea, is an alternative vector of the Flavescence doree phytoplasma (FDp) disease of grapevine in European vineyards, enabling infection initiation from wild reservoir compartment (Clematis vitalba). Heretofore recorded rate of D. europaea FDp-infection has been very low (3%), making it less epidemiologically significant than would be expected based on reservoir plant infection rate (30%). In this study we present findings on a heavily FDp-infected D. europaea population ( gt 60%), on the natural Wolbachia infection of populations with low FDp-infection rates (DeWo+) and on Wolbachia absence in highly FDp-infected population (DeWo-). We examine several possible causes underlying the differences in vector infection rates: (a) population genetic characteristics of D. europaea and correlation with Wolbachia strain wEur natural infections, (b) Wolbachia effects on fitness components of DeWo+ laboratory colony and (c) rate of reservoir plant FDp-infection and differences in FDp genotypes harboured by low and highly infected vector populations. The vector genetic diversity level was found to be lower in DeWo+ than in uninfected individuals and to exhibit a different evolution of fixed haplotypes. All DeWo+ populations were infected with the same strain of wEur. The FDp was found to be genetically diversified (five genotypes) but had no relation to infection rates. We did not find evidence of fitness upgrades with regard to Wolbachia infection status. Although more experimentation is needed, it seems that Wolbachia confers protection against FDp or is in competition with FDp according to the observed correlations: low FDp-infected vector populations are infected with Wolbachia and vice versa.

These Linaria species have been the targets of classical biological control programmes in Canada and the USA since the 1960s. The first effective toadflax biological control agent, the stem-mining weevil Mecinus janthinus (Coleoptera: Curculionidae) was introduced from Europe in the 1990s. This weevil has become established on L. dalmatica and L. vulgaris in both countries, although it has shown greater success in controlling the former toadflax species. Genetic and ecological studies of native range M. janthinus populations revealed that weevils previously identified as a single species in fact include two cryptic species, now recognised as M. janthinus, associated with yellow toadflax, and the recently confirmed species Mecinus janthiniformis, associated with Dalmatian toadflax. The results of a comprehensive study characterising haplotype identities, distributions and frequencies within M. janthinus s.l. native range source populations were compared to those populations currently established in the USA and Canada. The presence of both Mecinus species in North America was confirmed, and revealed with a few exceptions a high and consistent level of host fidelity throughout the adopted and native ranges. Genetic analysis based on mitochondrial cytochrome oxidase subunit II gene (mtCOII) defined the origin and records the subsequent North American establishment, by haplotype, of the European founder populations of M. janthinus (northern Switzerland and southern Germany) and M. janthiniformis (southern Macedonia), and provided population genetic indices for the studied populations. This analysis together with existing North American shipment receipt, release and rearing records elucidates probable redistribution routes and sources of both weevil species from initially released and established adopted range populations.
